Abstract

Изобретение относится к медицине, а также биохимии и биотехнологии и представляет собой способ стимуляции регенерации соединительной ткани (сухожилия и связки) при травмах и заболеваниях опорно-двигательного аппарата у лошади. Способ стимуляции репаративного ангиогенеза и регенерации соединительной ткани, при её повреждении и/или заболевании, в медицине, заключающийся в том, что его выполняют введением в организм животного генно-инженерной ДНК-конструкции, в которую клонированы гены, кодирующие экспрессию видоспецифичных (для конкретного вида животного) белковых факторов, а именно сосудистого эндотелиального фактора роста изоформы 164 (VEGF164) и фактора роста фибробластов 2 (FGF2), способ по п.1 характеризуется тем, что генно-инженерная ДНК-конструкция выполнена в виде плазмидной конструкций, способ по п.1 характеризуется тем, что генно-инженерная ДНК-конструкция выполнена в виде вирусного вектора либо вирусных частиц, способ по п.1 характеризуется тем, что генно-инженерная ДНК-конструкция выполнена в любом известном виде, способ по пп.1-4, отличающийся тем, что для реализации способа используют фармацевтическую композицию, содержащую генетическую ДНК-конструкцию в эффективном количестве и фармацевтически допустимые вспомогательные вещества, а также возможно вирусы, микроорганизмы или клетки, генетическая ДНК-конструкция SEQ ID NO:1, характеризующаяся тем, что для реализации способа по п.1 используют двухкассетную экспрессионную плазмидную ДНК-конструкцию, созданную на базе вектора pBudCE4.1, в котором клонированы видоспецифично кодон-оптимизированные гены, кодирующие экспрессию видоспецифичных для лошади домашней (Equus ferus caballus) белковых факторов, а именно сосудистого эндотелиального фактора роста изоформы 164 (VEGF164) и фактора роста фибробластов 2 (FGF2).The invention relates to medicine, as well as biochemistry and biotechnology, and is a method of stimulating the regeneration of connective tissue (tendons and ligaments) in injuries and diseases of the musculoskeletal system in a horse.
