Abstract
Description
- 1. Vorspülen (Füllen) des gesamten Systemes
- - P1 saugt ca. 600 ml "Arbeitspuffer" über V9 durch lau fende Zentrifuge und pumpt weiter durch Magnetfilter über V2 in "kranke Zellen".
- 2. waschen der zellulären Fraktion sowie Magnetfilter-
Reinigung
- - P1 saugt (25-50 ml) "zelluläre Fraktion" über V5 in laufende Zentrifuge. [Wascheinheit]
- - P1 saugt ca. 250 ml "Arbeitspuffer" über V9 durch laufende Zentrifuge und pumpt weiter durch Magnetfil ter über V2 in "kranke Zellen".
- 3. Magnetteilchenzugabe, Inkubation und Stopplösungszugabe
- - P1 saugt ca. 50 ml "Magnetteilchen-Lösung" über V8 in laufende Zentrifuge. [Mischeinheit]
- - P1 saugt ca. 50 ml "Stopplösung" über V7 in laufender Zentrifuge.
- 4. Magnetfiltration
- - Zentrifuge stoppt, somit verteilt sich die zelluläre Fraktion in der Zentrifuge.
- - {Magnetfeld einschalten}
- - P1 pumpt Zentrifugeninhalt über V4 durch [Magnetfil ter] und Zentrifuge im Kreis.
- 5. Konzentration und Aufreinigung gesunder Zellen sowie
Magnetfilter-Reinigung
- - Zentrifuge startet, somit sammelt sich die zelluläre Fraktion in der Zentrifuge. [Konzentrator]
- - {Magnetfeld ausschalten}
- - P1 saugt ca. 125 ml "Arbeitspuffer" über V9 durch laufende Zentrifuge und pumpt weiter durch Magnetfil ter über V2 in "kranke Zellen".
- - Zentrifuge bremst langsam ab (geringe Vermischung).
- - P1 drückt (5-30 ml) zelluläre Fraktion aus Zentri fuge über V6 in "gesunde Zellen".
Claims (11)
gekennzeichnet durch
eine Fraktioniereinheit (10),
eine Wascheinheit (20),
eine Mischeinheit (30),
eine zweite Wascheinheit (40),
einer Magnetfiltereinheit (50),
einen Konzentrator (60),
eine Meßeinrichtung (70) und
eine Steuereinheit (80).
gekennzeichnet durch die folgenden Verfahrensschritte:
- a) Vorbereitung der flüssigen biologischen Dispersion
- b) Inkubation von Magnetteilchen in die Dispersion in nerhalb eines festgelegten Zeitregimes
- c) Abstoppen des Inkubationszeitregimes durch Zugabe einer Stopplösung
- d) Abtrennung aller Magnetteilchen von der Dispersion durch Einwirkung eines Magnetfeldes
- e) Rückführung der biologischen Dispersion in ein bio logisches System.
