Abstract
La presente invención se relaciona con la rama de la Inmunología y en particular con la generación y selección de anticuerpos monoclonales (AcM) contra gangliósidos (Ab1s), que poseen la característica de ser específicos, de portar idiotipos recurrentes y que tienen valor como inmunoreguladores de la respuesta inmune contra estos antígenos; y con la obtención de AcM anti-idiotipos (Ab2s) generados a partir de la inmunización con dichos AcM anti-gangliósidos. De acuerdo con lo expuesto, un objeto de esta invención es proporcionar AcMs contra gangliósidos (Ab1s), portadores de idiotipos capaces de inducir una respuesta antígeno específica (Ab1´) en ausencia del antígeno, y los métodos para su generación. Otro objeto de esta invención es proveer de AcMs anti-idiotipo (Ab2s), generados mediante la inmunización con AcM antigangliósidos poseedores de las propiedades antes expuestas, que tienen la capacidad también de producir respuesta Ab1´. Otro importante objetivo de la invención consiste en generar AcM específicos contra gangliósidos (Ab1), inmunizar ratones y otras especies con dichos anticuerpos y seleccionar aquellos capaces de generar una respuesta antígeno específica (Ab1´). Generar AcM anti-idiotipo a partir de la inmunización de ratones u otras especies animales con los AcM anti-gangliósidos antes mencionados.The present invention is related to the branch of Immunology and in particular to the generation and selection of monoclonal antibodies (mAbs) against gangliosides (Ab1s), which have the characteristic of being specific, of carrying recurrent idiotypes and that have value as immunoregulators of the immune response against these antigens; and with obtaining anti-idiotype mAbs (Ab2s) generated from immunization with said anti-ganglioside mAbs. In accordance with the foregoing, an object of this invention is to provide mAbs against gangliosides (Ab1s), carriers of idiotypes capable of inducing a specific antigen response (Ab1´) in the absence of the antigen, and the methods for its generation. Another object of this invention is to provide anti-idiotype mAbs (Ab2s), generated by immunization with antiganglioside mAbs possessing the above properties, which also have the ability to produce an Ab1 'response. Another important objective of the invention is to generate specific mAbs against gangliosides (Ab1), immunize mice and other species with said antibodies and select those capable of generating a specific antigen response (Ab1´). Generate anti-idiotype mAbs from the immunization of mice or other animal species with the aforementioned anti-ganglioside mAbs.
