Abstract
鸡用缓释复方免疫增强剂,由药物和基质两部分组成,药物2份、基质3份;药物中有:黄芪多糖1~5份、淫羊藿多糖1~5份、左旋咪唑2.5~5份、维生素E 2.5~5份、亚硒酸钠0.1~0.2份;基质中含有:环氧树脂3.70~3.75份、乙二胺0.25~0.30份、邻苯二甲酸二丁酯0.05~0.06份、滑石粉3.30~3.39份。将环氧树脂放入容器中,取邻苯二甲酸二丁酯和乙二胺溶液,反应完全后备用;将药物原料混合均匀,按药物与基质比例加入到环氧树脂凝胶中搅拌,再加入一定量的滑石粉,揉搓均匀后切块、团成丸剂。本发明工艺简单,投服方便,在机体内作用时间长,药物能被机体充分吸收利用。

技术领域
本发明涉及现代免疫学和中兽医学领域,具体说是一种鸡用缓释复方免疫增强剂。
背景技术
免疫增强剂(Immunopotentiator)又称免疫佐剂(Immunoadjurant),是一类通过非特异性途径提高动物机体对抗原或微生物特异性反应的物质。它有利于加强机体的免疫系统功能,改善机体的免疫应答,增强机体的免疫水平。自1925年法国免疫学家Caston Ramon发现非特异性免疫增强作用以来,免疫增强剂的研究逐渐引起人们的重视,其领域不断拓展,研究逐步深入,发现和合成了许多有免疫增强作用的物质,在免疫血清制造,增强疫苗保护效果,防治传染病的发生等方面得到了广泛应用,并发挥了积极的作用。蔡传贵等(1997)在《养禽与禽病防治》发表的“免疫增强剂对家禽疫苗免疫作用的试验”,应用中草药芦荟多糖和维生素E、亚硒酸钠对家禽接种ND疫苗和IBD疫苗的免疫作用和实际应用效果进行了一些探索性研究;梁荣等(1998)在《中国兽医科技》发表的“中药免疫增强剂提高鸡免疫功能的研究”,以鸡新城疫(ND)和传染性腔上囊(IBD)为模型研究自拟中草药免疫增强剂对其灭活苗的免疫增强效果,结果表明,该中药免疫增强剂与ND、IBD疫苗配合使用具有免疫增强作用,可以提高机体体液免疫水平;柳纪省等(1999)在《中国兽医科技》发表的“天然药物缓释免疫增强剂的制备及其安全效力试验”,进行了鸡用天然缓释免疫增强剂的研究,在完成天然免疫增强效药物及配方筛选的基础上进行了测定,以期为生产中减轻鸡群免疫压力提高鸡的生产性能及肉、蛋品质有所帮助;王福传等(2002)在《兽药与饲料添加剂》中发表的“复方中草药免疫增强剂对鸡免疫器官组织形态学影响研究”,说明复方中草药免疫增强剂对鸡免疫器官的发育具有明显的促进作用;陈春林等(2005)在《中兽医学杂志》发表的“复合免疫增强剂对肉仔鸡免疫指标及抗病力的影响”,证实了以女贞子为主的复合免疫增强剂,能提高鸡新城疫抗体效价,延长其在体内的存留时间,并提高对鸡群的保护率,还可显著提高肉鸡免疫器官发育,可提高鸡痘的抗病力。由此可见,由中草药黄芪、淫羊藿、左旋咪唑、维生素E等组成的复合免疫增强剂对鸡新城疫(ND)和传染性腔上囊病(IBD)疫苗的免疫增强作用的研究和天然药物缓释免疫增强剂的制备,在国内已有文献报导。
尽管近年来对免疫增强剂的研究取得了一些进展,涌现出了大量制剂,但尚处于初始阶段,缺乏深入系统的研究,开发研制技术尚未成熟。有些药物作用机理尚不清楚,有些药物研究报导结果不一,有些化学免疫增强剂存在毒副作用和污染残留等弊端。中草药免疫增强剂多以粉、散剂拌料应用,剂型单一,使用不方便,能在兽医临床上应用的产品不多,更缺乏以畜禽品种为单位,以多种疾病为目标的免疫增强剂研究。由于免疫失败和疫苗免疫力不强等因素,造成鸡新城疫、传染性支气管炎、传染性法氏囊病和减蛋综合症等传染病不断发生,给养鸡业造成较大经济损失。此外疫苗的多次注射也给生产带来不便,影响工作效率。
发明内容
本发明的目的是提供一种以中草药有效活性成份与西药免疫增强剂药物有机结合起来,发挥中、西药物各自优势,取长补短,相互协调,以粘合剂为骨架的鸡用缓释复方免疫增强剂,给药途经方便,药效维持时间长,既能增强鸡的体液免疫功能和细胞免疫功能,提高血清抗体水平,延长维持时间,还能补充鸡维生素E及微量元素硒的不足,达到一丸多用之功效。
本发明的目的是通过下述技术方案实现的:
一种鸡用缓释复方免疫增强剂,其特征在于原料由药物和基质两部分组成,药物与基质配比为:
药物2份 基质3份
所述的药物含有下述原料及配比:
黄芪多糖1~5份 淫羊藿多糖1~5份 左旋咪唑2.5~5份
维生素E 2.5~5份 亚硒酸钠0.1~0.2份
所述的基质含有的原料及配比为:
环氧树脂3.70~3.75份 乙二胺0.25~0.30份
邻苯二甲酸二丁酯0.05~0.06份 滑石粉3.30~3.39份
其制备方法为
a、基质环氧树脂凝胶的制备:将环氧树脂放入容器中,按比例量取邻苯二甲酸二丁酯和乙二胺溶液,边加边搅拌,待反应完全后备用;
b、缓释复方免疫增强剂的制备:
将所需药物原料按比例称重,并充分混合均匀,按药物与基质比例加入到备用的环氧树脂凝胶中,进行搅拌,再按比例加入滑石粉,揉搓均匀后切块,团成丸剂。
本发明以黄芪多糖、淫羊藿多糖与西药免疫增强剂维生素E、亚硒酸纳、左旋咪唑有机结合,可促进鸡的生长,提高鸡的抗病力和免疫功能,减少疫苗免疫次数和药物用量。投服方便,在机体内缓慢释放,作用时间长,对养鸡业的健康发展具有重要作用。生产工艺简单,耗资少,成本低。本缓释复方免疫增强剂在鸡体内存留时间长,药物又能被机体充分吸收利用,不会造成对环境的污染。既能增强鸡的体液免疫功能和细胞免疫功能,提高血清抗体水平,延长维持时间,还能补充鸡维生素E及微量元素硒的不足,达到一丸多用之功效。将制备的鸡用缓释复方免疫增强剂,与鸡ND、IB、IBD、EDS疫苗配合使用,效果显著,无明显的毒副作用。
具体实施方式
下面结合实施例对本发明进行进一步详细的说明:
本发明所用原料:
黄芪多糖、淫羊藿多糖由本研究室按常规方法制备。
左旋咪唑(有效含量99%):陕西天汉兽药厂生产提供。
维生素E(有效含量56%):国外进口,西安爱丽丝畜牧有限公司分装提供。
亚硒酸钠(CR):西安化学试剂厂生产。
环氧树脂:星晨化工无锡树脂厂生产。
邻苯二甲酸二丁酯(AR):天津市化学试剂二厂生产。
乙二胺(AR):天津市化学试剂一厂生产。
滑石粉(医用):平凉市医药批发超市购置。
实施例1
(1)取环氧树脂370g放入容器中,按比例量取邻苯二甲酸二丁酯5g和乙二胺溶液25g,边加边搅拌,待反应完全后备用。
(2)取黄芪多糖100g、淫羊藿多糖500g、左旋咪唑250g、维生素E 500g、亚硒酸钠20g;充分混合均匀,按药物与基质比例加入到备用的环氧树脂凝胶中,进行搅拌,再加入一定量的滑石粉,揉搓均匀后切块,团成丸剂(每丸重0.25g)。
(3)检验
将制备好的缓释复方免疫增强剂,放入磁盘中,待干燥后装袋,并贴上标鉴、注明批号、生产日期、数量,同时做好各项记录,然后由质检人员进行抽样检验,待合格后方可发出使用。
成品规格:
物理性状
制备好的缓释复方免疫增强剂呈褐色,坚硬光滑,在制成后4~12h完全硬化。
浸水性
将制备好的缓释复方免疫增强剂于37℃在水中浸泡90d,表面应无软化现象。
磨损量及药物有效释放量
该缓释复方免疫增强剂的日均磨损量前期为6.2~6.7mg,后期为6.0~6.3mg,日均药物有效释放量为2.40~2.52mg。
安全性
青年仔鸡经3倍量投服后,观察20d应无任何不良反应,饮、食欲正常,精神状态良好。
效力检验
在投服缓释复方免疫增强剂的同时,进行ND疫苗免疫,在免疫后10、20、30d采血,用β-微量血凝抑制试验、T淋巴细胞E玫瑰花环试验、外周血T淋巴细胞酸性α-醋酸奈酯酶阳性率测定,经方差分析应与同时设立的对照组相比差异显著或极显著。
保存
在干燥、通风的环境中,可常年保存。
实施例2
(1)取环氧树脂375g放入容器中,按比例量取邻苯二甲酸二丁酯6g和乙二胺溶液30g,边加边搅拌,待反应完全后备用。
(2)取黄芪多糖500g、淫羊藿多糖100g、左旋咪唑500g、维生素E 250g、亚硒酸钠18g;充分混合均匀,按药物与基质比例加入到备用的环氧树脂凝胶中,进行搅拌,再加入一定量的滑石粉,揉搓均匀后切块,团成丸剂(每丸重0.25g)。
(3)检验
同实施例1。
实施例3
(1)取环氧树脂375g放入容器中,按比例量取邻苯二甲酸二丁酯6g和乙二胺溶液25g,边加边搅拌,待反应完全后备用。
(2)取黄芪多糖300g、淫羊藿多糖200g、左旋咪唑250g、维生素E 250g、亚硒酸钠0.18g;充分混合均匀,按药物与基质比例加入到备用的环氧树脂凝胶中,进行搅拌,再加入一定量的滑石粉,揉搓均匀后切块,团成丸剂(每丸重0.25g)。
(3)检验
同实施例1。

1.一种鸡用缓释复方免疫增强剂,其特征在于原料由药物和基质两部分组成,药物与基质配比为:

黄芪多糖1~5份 淫羊藿多糖1~5份 左旋咪唑2.5~5份
维生素E 2.5~5份 亚硒酸钠0.1~0.2份
所述的基质由下述原料及配比制成:
环氧树脂3.70~3.75份 乙二胺0.25~0.30份
邻苯二甲酸二丁酯0.05~0.06份 滑石粉3.30~3.39份。
2.一种制备如权利要求1所述鸡用缓释复方免疫增强剂的方法,其特征在于由下述方法制备而成:
a、基质环氧树脂凝胶的制备:将环氧树脂放入容器中,按比例量取邻苯二甲酸二丁酯和乙二胺溶液,边加边搅拌,待反应完全后备用;
b、缓释复方免疫增强剂的制备:
将所需药物原料按比例称重,并充分混合均匀,按药物与基质比例加入到备用的环氧树脂凝胶中,进行搅拌,再按比例加入滑石粉,揉搓均匀后切块,团成丸剂。
