本发明公开了一种制备积雪草苷巴布剂的方法。该方法过程包括,以质量20%~30%的溶胀明胶赋形剂,在45℃~65℃水浴加热与质量5%~35%的高岭土填充剂先进行搅拌混合均匀,然后保持温度加入质量1%~10%的聚乙烯醇和1%~10%的聚乙烯吡咯烷酮或1%~10%的羧甲基纤维素钠、及质量10%~20%的聚乙烯酸钠黏性剂,和质量8%~15%的甘油与1%~10%的丙二醇保湿剂搅拌均匀制成巴布剂的基质,最后加质量1%~25%的含积雪草苷20%~90%的提取物干粉,均匀搅拌炼和15分钟至45分钟,炼和温度为40℃至60℃,趁热涂布,静置,冷却,干燥,即得巴布剂。本发明制备过程简单,所制备巴布剂毒副作用低,药效作用时间长。

技术领域
本发明涉及一种制备积雪草苷巴布剂的方法,属于积雪草药物制备技术。
背景技术
积雪草(Centella asiatica(L.)Urban),是伞形科积雪草属植物,为《中国药典》收载的常用中药。积雪草具有清热利湿、解毒消肿的功效,临床上多用于治疗跌打损伤、皮肤病等。在印℃它被称为Mandukaparni,通常作为伤口愈合剂和增强大脑的补益品。由于积雪草具有较强的药理活性,国外于1937年开始研究其化学成分,至今已从中分离得到了三萜类、多炔烯类、单萜、倍半萜、生物碱、黄酮类等多种活性化学成分。目前已证实主要活性成分是积雪草苷A、积雪草苷B、积雪草苷、马达积雪草酸等五环三萜类成分。以此类活性成分申请了许多专利,如US2004151789、US2004121027、NZ515900、CN1429553、JP8133952、JP2004059482等。在上述专利中,虽然涉及到利用积雪草提取物或主要活性成分积雪草苷等制备软膏或化妆品,但没有关于积雪草苷巴布剂的制备。
发明内容
本发明的目的在于提供一种制备积雪草苷巴布剂的方法。该方法制备过程简单,所制备巴布剂毒副作用低,药效作用时间长。
本发明是通过下述技术方案加以实现的,一种制备积雪草苷巴布剂的方法,该方法其特征在于包括以下过程:以质量含量20%~30%溶胀明胶赋形剂,在45℃~65℃水浴加热与质量含量5%~35%高岭土填充剂先进行搅拌混合均匀,然后保持温度加入质量含量1%~10%聚乙烯醇和1%~10%聚乙烯吡咯烷酮或羧甲基纤维素钠、及质量含量10%~20%聚乙烯酸钠黏性剂,和质量含量8%~15%甘油与1%~10%丙二醇保湿剂搅拌均匀制成巴布剂的基质,最后再加积雪草苷质量含量20%~90%的提取物干粉,均匀搅拌炼和15分钟至45分钟,炼和温度为40℃至60℃,趁热涂布,静置,冷却,干燥,即得巴布剂。
本发明的优点在于,制备过程简单,以含量大于20%积雪草苷主要活性成分为主要原料制备成巴布剂,其透皮给药方法简单,毒副作用发生度较低,同时可以根据要求随时中断给药。此外,还减少了过去由积雪草提取物或其主要活性成分制备的各种软膏、油等易被污染、易脱落等缺点。这也减少了用药次数延长了作用时间。
具体实施方式:
【实施例1】
本发明首先是提取积雪草有效成分,具体程序是称取适量药材加入8~12倍体积乙醇或甲醇0~99%,采用加热回流提取三次,每次2小时;或用超声提取每次0.5小时,共三次。然后将提取液浓缩制备成粉状,此为粗提物。粗提物用硅胶柱层析方法,用氯仿∶甲醇∶水(7∶3∶0.3)等度洗脱,收集所得馏分,获得20%~90%积雪草苷等主要活性成分,并制备成干粉。或者将粗提物用大孔吸附树脂,采用0~100%乙醇洗脱,收集10%~60%洗脱液,获得20%~90%积雪草苷等主要活性成分,并制备成干粉。
20%积雪草苷巴布剂的制备:积雪草提取物∶明胶∶聚乙烯醇∶聚乙烯吡咯烷酮∶聚乙烯酸钠∶甘油∶丙二醇∶高岭土,它们配比为20g∶20g∶4g∶3g∶12g∶10g∶5g∶10g,各组分添加顺序为高岭土与明胶在水浴55℃下搅拌均匀混合,然后在同温度下加入聚乙烯醇、聚乙烯吡咯烷酮、聚乙烯酸钠、甘油、丙二醇再搅拌混合均匀后,加入20%积雪草苷提取物,搅拌炼和30min,炼合温度为50℃趁热涂布,静置,冷却,干燥,即得20%积雪草苷巴布剂。
20%积雪草苷巴布剂有效成分的检测方法:利用高效液相法测巴布剂中积雪草苷的含量。色谱条件:zorbax SB c18分析柱(4.6×250mm),流动相为乙腈∶水(28∶72),柱温25℃,流速:1ml/min,检测波长205nm。检测物质为积雪草苷,保留时间为8分钟。此色谱条件下检测无干扰,主峰分离度好,结果准确、稳定、可靠。
【实施例2】
90%积雪草苷巴布剂的制备:积雪草提取物∶明胶∶聚乙烯醇∶聚乙烯吡咯烷酮∶聚乙烯酸钠∶甘油∶丙二醇∶高岭土,它们配比为5g∶25g∶5g∶5g∶15g∶12g∶5g∶28g,各组分添加顺序为高岭土与明胶在水浴55℃下搅拌均匀混合,然后在同温度下加入聚乙烯醇、聚乙烯吡咯烷酮、聚乙烯酸钠、甘油、丙二醇再搅拌混合均匀后,加入90%积雪草苷提取物,搅拌炼和30min,炼和温度为50℃,趁热涂布,静置,冷却,干燥,即得90%积雪草苷巴布剂。
【实施例3】
50%积雪草苷巴布剂的制备:积雪草提取物∶明胶∶聚乙烯醇∶羧甲基纤维素钠∶聚乙烯酸钠∶甘油∶丙二醇∶高岭土,它们配比为10g∶20g∶5g∶5g∶20g∶12g∶5g∶23g,各组分添加顺序为高岭土与明胶在水浴55℃下搅拌均匀混合,然后在同温度下加入聚乙烯醇、羧甲基纤维素钠、聚乙烯酸钠、甘油、丙二醇再搅拌混合均匀后,加入50%积雪草苷提取物,搅拌炼和30min,炼和温度为50℃,趁热涂布,静置,冷却,干燥,即得50%积雪草苷巴布剂。

1、一种制备积雪草苷巴布剂的方法,其特征在于包括以下过程:以质量20%~30%的溶胀明胶赋形剂,在45℃~65℃水浴加热与质量5%~35%的高岭土填充剂先进行搅拌混合均匀,然后保持温度加入质量1%~10%的聚乙烯醇和1%~10%的聚乙烯吡咯烷酮或1%~10%的羧甲基纤维素钠、及质量10%~20%的聚乙烯酸钠黏性剂,和质量8%~15%的甘油与1%~10%的丙二醇保湿剂搅拌均匀制成巴布剂的基质,最后再加质量1%~25%的含积雪草苷20%~90%的提取物干粉,均匀搅拌炼和15分钟至45分钟,炼和温度为40℃至60℃,趁热涂布,静置,冷却,干燥,即得巴布剂。
