技术领域
本发明涉及一种卡拉胶低聚糖精氨酸盐及其制备方法。它可用于生产放射防护剂、用作水产动物的药物或饲料添加剂,用于生产和研究其它生物活性物质或中间体。
背景技术
卡拉胶是一类红藻多糖,由于其独特的理化性质,长期以来广泛用在食品工业和医药、卫生和科学研究方面。卡拉胶具有多种生物活性,特别是作为一类硫酸半乳聚糖,其显著的生物活性,已引起医药界广泛关注。但卡拉胶分子量较大,粘度高,溶解性低等而使其作为药物应用受到极大限制。大量研究表明,具有一定分子量的硫酸低聚糖仍然保持其生物活性。而且,为提高低聚糖的生物活性,低聚糖的分子修饰和结构改造具有重要意义。目前,将卡拉胶转化为卡拉胶低聚糖精氨酸盐的研究未见报道。
发明内容
本发明的目的是提供一种卡拉胶低聚糖精氨酸盐及其制备方法,它能弥补现有技术上述的不足。

一种卡拉胶低聚糖精氨酸盐的制备方法,其特征在于首先向卡拉胶中加水,再加酸,在30-90℃下加热反应15-600min,反应结束后向反应液中加碱中和,再脱盐,将所得反应液离心分离后蒸发浓缩,向浓缩液中加入有机溶剂,离心分离,将上清液浓缩,通过色谱柱,收集洗脱液,加精氨酸,冷冻干燥。
本发明的卡拉胶低聚糖精氨酸盐可用于生产放射防护剂、用作水产动物的药物或饲料添加剂,用于生产和研究其它生物活性物质或中间体。
具体实施方式
实施例1.
取10g卡拉胶,加入1000ml蒸馏水并加热使其溶解,加入2N盐酸水溶液5ml,于80℃水浴搅拌水解240min,反应结束后,将水解液用冷水浴冷却至室温,用氢氧化钠中和至中性,脱盐,滤液于40℃下减压浓缩至100ml,加入3倍量乙醇,使其产生沉淀,离心分离,将上清液于40□减压浓缩,冷冻干燥得寡糖混合物。取2g该寡糖混合物,加1ml水溶解后注入Dowex 50色谱柱,室温下以蒸馏水进行洗脱,收集合并pH为3-6的洗脱液,加精氨酸调节pH为5.0,减压浓缩,冷冻干燥,得卡拉胶低聚糖精氨酸盐。该物质为白色,无定型粉末,易溶于水、稀酸和碱,通过核磁共振光谱、红外光谱、气质联用和质谱等技术确定了其结构。
实施例2.
取100g卡拉胶,加入5000ml蒸馏水并加热使其溶解,加入3N硫酸溶液15ml,于70℃水浴搅拌水解7h,反应结束后,将水解液用冷水浴冷却至室温,用氢氧化钠中和至中性,脱盐,滤液于40℃下减压浓缩至200ml,加入4倍量甲醇,使其产生沉淀,离心分离,上清液于40℃减压浓缩,冷冻干燥得寡糖混合物。取10g该寡糖混合物,加1ml水溶解后注入S Sepharose色谱柱,室温下以蒸馏水进行洗脱,收集合并pH为3-7的洗脱液,加精氨酸调节pH为6.0,减压浓缩,冷冻干燥,得卡拉胶低聚糖精氨酸盐。
本发明中所述的卡拉胶加水,使卡拉胶的重量百分浓度在0.1-10%。所用的酸为硫酸或盐酸或醋酸或柠檬酸,它们的浓度为1N-6N,卡拉胶与酸的重量比例为12∶1-8。反应温度和时间也影响产物分子量分布,并影响产物得率,可选择30-90□,反应时间为15-600min。所述的有机溶剂为乙醇或甲醇。所述的色谱柱为Dowex 50或SSepharose或SP Sepharose或SP Sephadex或Amberlite IR-120或Lewatit-100或DiaionSK-1或001X7阳离子交换树脂或732阳离子交换树脂等;所用洗脱剂为水,收集合并pH为1-7的洗脱液。所述的加精氨酸调节pH,加入精氨酸的量可调节收集液的pH为3-8。

2.一种卡拉胶低聚糖精氨酸盐的制备方法,其特征在于首先向卡拉胶中加水,再加酸,在30-90℃下加热反应15-600min,反应结束后向反应液中加碱中和,再脱盐,将所得反应液离心分离后蒸发浓缩,向浓缩液中加入有机溶剂,离心分离,将上清液浓缩,通过色谱柱,收集洗脱液,加精氨酸,冷冻干燥。
3.如权利要求2所述的方法,其特征在于向所述的卡拉胶加水,使卡拉胶的重量百分浓度在0.1-10%。
4.如权利要求2所述的方法,其特征在于所用的酸为硫酸或盐酸或醋酸或柠檬酸,它们的浓度为1N-6N,卡拉胶与酸的重量比例为12∶1-8。
5.如权利要求2所述的方法,其特征在于所述的有机溶剂为乙醇或甲醇。
6.如权利要求2所述的方法,其特征在于所述的色谱柱为Dowex 50或S Sepharose或SP Sepharose或SP Sephadex或Amberlite IR-120或Lewatit-100或Diaion SK-1或001X7阳离子交换树脂或732阳离子交换树脂等;所用洗脱剂为水,收集合并pH为1-6的洗脱液。
7.如权利要求2所述的方法,其特征在于所述的加精氨酸,加入精氨酸的量可调节收集液的pH为3-8。
