背景技术:
随着人们生活水平的显著提高,人们对膳食、保健品和药品的要求也越来越严格。以天然动植物为原料提取的天然色素具有安全无毒、富有营养和保健医疗的特点,受到消费者的重视,并有逐步取代合成色素的趋势。在天然色素中,类胡萝卜素是最重要的一种。类胡萝卜素包括胡萝卜素和叶黄素。其中,叶黄素是一种天然存在的不表现维生素A生物活性的类胡萝卜素。其广泛存在于水果和蔬菜中。在绿色蔬菜和水果中,如甘蓝、豌豆、菠菜、莴苣、猕猴桃和蜜汁等,并以天然未酯化状态和其他胡萝卜素共同存在。叶黄素也存在于许多黄色的蔬菜和水果中。如桃子、木瓜、芒果、南瓜和橘子等,在其中与高级脂肪酸形成脂肪酸酯的形式存在。

在国内外,长期以来就有多种制备黄体素类天然产物的研究和实例,如美国专利US005648564A等,这些制备方法尽管可以制备黄体素,但其产品品质低,而且工业化大批量生产的可行性不强,难以取得精制的医用品和食品类及化妆级的产品,所以一直使黄体素的应用局限于饲料行业的工业化生产,而对保健品和医药品的黄体素的工业化生产一直处于探索阶段和实验室规模。尤其是对叶黄素的提纯和保护,工业化的难度比较大。本发明是以万寿菊花为原料,通过物理和化学的手段,设计出黄体素的萃取、浓缩、皂化、洗涤、结晶、包埋、干燥等加工工艺路线和方法,以制备医药品和保健品级的工业化原料,为人类的健康作出努力。
发明内容:
本发明的主要技术工艺包括制粒、萃取、浓缩、脱残、皂化、洗涤、结晶、制粉、包埋及干燥等步骤。先将鲜万寿菊花经发酵、压榨、干燥过程制成干品颗粒后,按重量比例加入溶剂反复进行萃取,合并萃取液减压浓缩,再将浓缩液进行脱除残留溶剂制得膏状叶黄素树脂,将叶黄素树脂与丙二醇混合溶解,控制温度和时间进行皂化,使叶黄素脂肪酸酯转化为具有生物活性的黄体素单体,黄体素转化为类胡萝卜素;再将皂化后的叶黄素树脂用丙酮洗涤三次,在油水分离器中静置分层,收集上层油相并浓缩后在结晶器中进行结晶,然后离心过滤收集黄体素和玉米黄素晶体,再将结晶体溶于二硫化碳和石油醚的二元溶液中进行重结晶,再离心过滤收集晶体,在氮气保护下将黄体素和玉米黄素低温干燥成粉末状;然后取叶黄素粉末与抗氧化剂混合,分散于明胶溶液中,进行沸腾造粒干燥,制成微囊状的黄体素产品。采用本技术工艺过程制备黄体素,其工艺过程安全可靠,采用常规设备和溶剂可以降低生产成本,其制成的产品抗氧化性能好,纯度高,安全使用性强,可以作为医用和食用天然着色剂及保健品的主要原料。

1、万寿菊花在黄体素含量高峰期采摘后,经过避光发酵、压榨、干燥等工艺过程,制成直径为2-3mm,长度为10mm的颗粒。其含量大约15g/kg,贮存在低温干燥避光的库房,成为萃取黄体素的主要原料。
2、以6号溶剂油或烷烃类有机溶剂为萃取剂在萃取釜中控温35-45℃范围内进行三次逆流萃取。其中溶剂与万寿菊花颗粒的重量比为4∶1。萃取时间分别为1小时、2/3小时、1/3小时。
3、合并三次萃取液,进入浓缩罐,在温度40℃、真空度为0.085MPa的操作条件下进行减压浓缩。
4、将浓缩液进行脱除残留溶剂,向反应釜通入氮气,连续搅拌,抽真空,温度控制在40℃,时间12小时,使叶黄素树脂中总类胡萝卜素的含量达到100-150g/kg.叶黄素树脂中有机溶剂残留量小于100ppm,在常温条件下,叶黄素树脂以深黄色的浸膏状态存在。将该浸膏贮存在低温、干燥、避光的环境中。
5、将叶黄素浸膏和食品级或医药级丙二醇以1∶1的重量比例在小于60℃的反应釜中混合溶解,配制氢氧化钾或氢氧化钠或碳酸钠的饱和水溶液,以一定流速缓慢加入反应釜中,连续搅拌,控温65℃,时间3小时,用薄层色谱法测定皂化率为95%以上,皂化过程即可结束。黄体素由原来稳定态转变成具有生物活性的类胡萝卜素。采用美国官方分析协会(AOAC)法测定,其中具有生物活性的总类胡萝卜素约为60-80g/kg.采用高压液相色谱的方法测定黄体素占总类胡萝卜素的94-97%,玉米黄素占总类胡萝卜素的3-6%。
6、皂化的叶黄素在常温下溶于丙酮溶剂,过滤,除去丙酮不溶物。不溶物用丙酮洗涤三次,直至洗液成浅黄色为止,丙酮溶液和正己烷以1∶1的重量比例再混合,加入油水分离器中,静置分层。可适当加入少量的水,加速分层速度,收集上层油相,用10%的无水硫酸钠水溶液洗涤,除去水溶物。
7、将洗涤后的上层油相浓缩,在0-5℃条件下在结晶器中进行结晶,时间6-24小时,离心过滤,收集黄体素和玉米黄素晶体。采用高压液相色谱(HPLC)法测定总类胡萝卜素含量30-50%。
8、将上述晶体溶于二硫化碳和石油醚(按重量比1∶1)的二元溶液中进行重结晶。
9、离心过滤收集晶体,在氮气保护下进行黄体素和玉米黄素低温干燥成粉末状,纯度可达70-98%,得率在60%以上。
10、将高纯度的叶黄素粉末和二叔丁基羟基甲苯或叔丁基羟基茴香醚或叔丁基对苯二酚或维生素E等较强的抗氧化剂以1000∶5的重量比混合,分散于明胶溶液中,进行沸腾造粒干燥,制成稳定的微囊产品,成为医药级黄体素的主原料。
Claims (7)
1、一种黄体素的制备方法,以万寿菊花为原料,用物理和化学手段,经制粒、萃取、浓缩、脱残、皂化、洗涤、结晶、制粉、包埋及干燥等步骤,其特征在于先将鲜万寿菊花经发酵、压榨、干燥过程制成干品颗粒后,按重量比例加入溶剂反复进行萃取,合并萃取液减压浓缩,再将浓缩液进行脱除残留溶剂制得膏状叶黄素树脂,将其与丙二醇混合溶解,控制温度和时间进行皂化,使黄体素转化为类胡萝卜素;再将皂化后的叶黄素树脂用丙酮洗涤,在油水分离器中静置分层,收集上层油相并浓缩后在结晶器中进行结晶,然后离心过滤收集黄体素和玉米黄素晶体,再将结晶体溶于二硫化碳和石油醚的二元溶液中进行重结晶,再离心过滤收集晶体,在氮气保护下将黄体素和玉米黄素低温干燥成粉末状;然后取叶黄素粉末与抗氧化剂混合,分散于明胶溶液中,进行沸腾造粒干燥,制成微囊状的黄体素产品。
2、根据权利要求1所述的黄体素的制备方法,其特征在于萃取溶剂为6号溶剂油或烷烃类有机溶剂,其与万寿菊花颗粒的重量比为4∶1。
3、根据权利要求书1所述的黄体素的制备方法,其特征在于将叶黄素浸膏和丙二醇以1∶1的重量比例在小于60℃的反应釜中混合溶解,配制氢氧化钾或氢氧化钠或碳酸钠的饱和水溶液,缓慢加入反应釜中,连续搅拌,控温65℃完成皂化过程。
4、根据权利要求书1所述的黄体素的制备方法,其特征在于将洗涤后的上层油相浓缩,在0-5℃下结晶器中进行结晶,离心过滤,收集黄体素和玉米黄素晶体。
5、根据权利要求书1所述的黄体素的制备方法,其特征在于将黄体素和玉米黄素溶于二硫化碳和石油醚(按重量比1∶1)的二元溶液中进行重结晶。
6、根据权利要求书1所述的黄体素的制备方法,其特征在于离心过滤收集晶体,在氮气保护下干燥成粉末。
7、根据权利要求书1所述的黄体素的制备方法,其特征在于粉末和抗氧化剂以1000∶5的重量比例混合,分散于明胶溶液中,进行沸腾造粒干燥,制成微囊产品。
