Abstract
一种氯化-N-三甲基壳聚糖季铵盐,其特征是它的分子式为(C8H13NO5)n(C8H17NO4)m-x(C9H20NO4Cl)x,式中n=0.07~0.19,x=0.39~0.75,n+m+x=1。制备时将壳聚糖溶胀于1-甲基-2-吡咯烷酮中,加入碘化钠,分批加入碘甲烷保持50~65℃搅拌反应,反应液冷却后倒入丙酮中析出沉淀,并用丙酮-水溶液洗涤沉淀物;将沉淀物溶于水中,滴加盐酸-乙醇溶液进行离子交换,然后向反应液加入丙酮,所得沉淀用丙酮-水溶液洗涤后再溶于水,再加入到丙酮中进行沉淀,该沉淀继续用丙酮-水溶液洗涤、丙酮洗涤,烘干。本发明的优点在于分子中季铵盐的取代度可通过控制碘甲烷的投料量而加以控制,产物的水溶性好,它能满足作为抗菌剂和大分子药物吸收增强剂的要求。制备时操作简单、成本低,适用于产业化制备。
Description
技术领域
本发明涉及一种水溶性壳聚糖季铵盐衍生物,特别是涉及一种氯化-N-三甲基壳聚糖季铵盐。
背景技术
目前在壳聚糖季铵盐衍生物开发方面的报道很少,其应用也仅限于絮凝剂及抑菌剂。而具有良好的促进水溶性大分子药物如多肽和蛋白类药物透过肠内上皮细胞单层吸收的作用,可用作大分子药物吸收增强剂的尚未见报道。
发明内容
本发明的目的是提供一种氯化-N-三甲基壳聚糖季铵盐,它能弥补现有技术的上述不足。
一种氯化-N-三甲基壳聚糖季铵盐,其特征是它的分子式为(C8H13NO5)n(C8H15NO4)m(C9H18NO4Cl)x,其结构式为:
式中n=0.07~0.19,x=0.39~0.75,n+m+x=1。
上述的氯化-N-三甲基壳聚糖季铵盐的制备方法,其特征是将壳聚糖溶胀于1-甲基-2-吡咯烷酮中,加入碘化钠,在碱性条件下分批加入碘甲烷保持50-65℃搅拌反应,所述的壳聚糖∶碘化钠∶碘甲烷的总摩尔比为:1∶2.5-3∶13.6-16.3;反应液冷却至室温后倒入丙酮中析出沉淀,并用70-80%的丙酮-水溶液洗涤所得沉淀物;所得沉淀物溶解于水中,搅拌下滴加3-3.5%盐酸-乙醇溶液进行离子交换,然后将反应液加入到丙酮中使充分沉淀,所得沉淀用70-80%丙酮-水溶液洗涤后再溶于水,再加入到丙酮中进行沉淀,所得沉淀用70-80%丙酮-水溶液洗涤、丙酮洗涤,70-80℃烘干。
本发明的优点在于分子中季铵盐的取代度可通过控制碘甲烷的投料量而加以控制,产物的水溶性好,它能满足作为抗菌剂和大分子药物吸收增强剂的要求。制备时操作简单、成本低,适用于产业化制备。
具体实施方式
下面通过实例说明本发明。
1、将壳聚糖粉碎、过120目筛网后的粉状颗粒40.0g(0.24mol)溶胀于120mL的1-甲基-2-吡咯烷酮中,加入96.0g(0.64mol)碘化钠固体,加热至60℃,搅拌下加入30%(重量百分浓度,下同)氢氧化钠水溶液220mL和碘甲烷液体120mL(1.93mol),60℃下恒温搅拌反应2h;再加入30%氢氧化钠水溶液80mL和碘甲烷80mL(1.29mol),60℃下恒温搅拌反应1h后继续加入30%氢氧化钠水溶液40mL和碘甲烷40mL(0.64mol),60℃下继续恒温搅拌反应1h,壳聚糖、碘化钠、碘甲烷的总摩尔比为1∶2.7∶16.1。反应完毕将反应液冷却至室温加入到工业丙酮中进行沉淀,抽滤收集沉淀物,并用70-80%丙酮-水溶液洗涤,将所得沉淀物溶于600mL蒸馏水中,搅拌下滴加1200mL3%盐酸-乙醇溶液进行离子交换反应,然后加入工业丙酮使充分沉淀,抽滤收集沉淀物,该沉淀物用70-80%丙酮-水溶液清洗;所得沉淀物再溶于200mL蒸馏水中,搅拌下加入到工业丙酮中进行沉淀,抽滤收集沉淀物,沉淀物再用80%丙酮-水溶液,丙酮依次洗涤,75℃下烘干即得到本发明的氯化-N-三甲基壳聚糖季铵盐33.5g,x=39.8%。
2、结构鉴定
测定所用仪器及条件:
红外光谱:NICOLET NEXUS-470智能光谱仪,KBr压片法测定。
核磁共振波谱:JEOL JUM-ECP-600核磁共振波谱仪。
以粘均分子量为5.0×104氯化-N-三甲基壳聚糖季铵盐为例:
1).FT-IR(KBr)cm1:3500-3300(-OH的伸缩振动峰),3000-2900(-CH3伸缩振动峰),2900-2800(-N+H(CH3)2伸缩振动峰),1639(酰胺I带、-CO-羰基伸缩振动),1475、1374、1310(C-H弯曲振动峰及酰胺III带的叠加峰),1237(C-N的伸缩振动峰),1152、1096、1059(C-O-C的伸缩振动峰)。与原料相比,1599处-NH2的峰消失,2955、1475、1374处-CH3明显增强及1237处C-N伸缩振动峰增强,C-O的伸缩振动峰无明显变化,表明产物中壳聚糖季铵盐的生成。
2).1H NMR(D2O,DSS为内标,80℃),δ:2.07(s,-COCH3),3.00-3.15(s,-N(CH3)2),3.30-3.40(s,-N+(CH3)3),3.40-3.50(s,6-O-CH3),3.50-3.60(s,3-O-CH3)。3.80-4.10(壳聚糖环C-2,C-3,C-4,C-5,C-6的H),4.80-5.50(C-1的H)。
3).取代度的确定:
取20mg样品溶解于1ml D2O中,放置2h,冷冻干燥;重复此交换操作2次。冻干后所得样品以D2O为溶剂,DSS为内标,80℃进行1H NMR波普分析。氯化-N-三甲基壳聚糖季铵盐的取代度由通过下列公式计算:
%DQ={[(CH3)3]/[H1]×1/9}×100%%DQ是氯化-N-三甲基壳聚糖季铵盐分子中季铵化部分的百分含量,[(CH3)3]是1H NMR谱图上-N+(CH3)3化学位移为3.3ppm的峰面积,[H1]是谱图上化学位移在4.7-5.7ppm的H1信号峰面积。
根据上述方法和相应的1H NMR波谱数据:δ=3.34ppm处-N+(CH3)3的H的峰面积为6.09,δ=4.88-5.43处H-1的峰面积之和为1.70,测得本发明产品的季铵盐取代度DQ%={[6.09/1.70]×1/9}×100%=39.8%。
Claims (1)
1.一种氯化-N-三甲基壳聚糖季铵盐的制备方法,其特征是将壳聚糖溶胀于1-甲基-2-吡咯烷酮中,加入碘化钠,在碱性条件下分批加入碘甲烷保持50-65℃搅拌反应,所述的壳聚糖∶碘化钠∶碘甲烷的总摩尔比为:1∶2.5-3∶13.6-16.3;反应液冷却至室温后倒入丙酮中析出沉淀,并用70-80%的丙酮-水溶液洗涤所得沉淀物;所得沉淀物溶解于水中,搅拌下滴加3-3.5%盐酸-乙醇溶液进行离子交换,然后将反应液加入到丙酮中使充分沉淀,所得沉淀用70-80%丙酮-水溶液洗涤后再溶于水,再加入到丙酮中进行沉淀,所得沉淀用70-80%丙酮-水溶液洗涤、丙酮洗涤,70-80℃烘干。
