Abstract
一种具有抑菌修护作用的水凝胶敷料,包括以下重量份的各组分:金银花纯露18‑22份、桑黄多糖提取物6‑8份、明胶1‑3份、保湿剂2‑4份以及营养剂1‑3份。本发明采用超声协同纤维素酶法提取桑黄粉中的多糖,利用纤维素酶能够有效降解植物中的纤维素,进而有效破除植物细胞壁,使植物细胞多糖有效溶出,从而提高桑黄多糖的提取量,此过程简单、价格低廉、条件温和、对环境无污染,利于绿色工业化生产。本发明所制备的桑黄多糖不仅抗菌能力强,且抗氧化能力好,更易深层滋养肌肤。采取消炎抗菌能力强、来源天然的弱酸性金银花纯露代替去离子水制备化妆品,不仅因其呈弱酸性而更贴合肌肤,其所自行散发的特殊的植物香味还能愉悦身心。

背景技术
桑黄作为我国传统名贵中草药,在民间素有“森林黄金”之尊称,多部中医药典籍均有记载其药用价值,如《本草纲目》中曾提及桑黄“利五脏,宣肠胃气,排毒气”。它属于寒性药物,味道苦涩。研究表明桑黄的主要活性成分包括多糖、黄酮、麦角甾醇、三萜类等(赵鹏,李彩霞,左凡娇,等. 桑黄化学成分、药理活性研究进展及质量标志物(Q-Marker)预测[J]. 药物评价研究),其中桑黄多糖作为核心活性成分,具有抗肿瘤、抗氧化、抗糖、提高免疫力、抗突变、清除自由基等功效。

植物纯露是指芳香植物经过蒸馏萃取精油的过程中得到的副产物,有特殊植物香气,含有微量的酯类、酮类、烯类以及醇类,具有亲水性,大多数纯露呈弱酸性,作为化妆品对肌肤具有舒缓、消炎作用。研究表明,金银花纯露富含绿原酸等,较其精油而言,其具有良好的消炎抗菌能力(王嘉璐,张彩凤,徐润杨,等. 金银花活性成分的功能及应用研究进展[J].食品安全导刊)。

本发明为解决现有技术中存在的问题,提供了一种将具有优良抗菌抗氧化作用的桑黄多糖等作为有效成分,应用于制备具备抑菌修护功能的水凝胶敷料的方法。
本发明属于植物提取和化妆品领域,本发明采用桑黄多糖和金银花纯露为主要原料,制备了一种具有抗氧化、温和保湿的护肤品,其工艺简单、绿色环保,填补了桑黄多糖用于化妆品领域的空白。
本发明的一种具有抑菌修护作用的水凝胶敷料,包括以下重量份的各组分:
金银花纯露 18-22份
桑黄多糖提取物 6-8份
明胶 1-3份
保湿剂 2-4份
营养剂 1-3份。
优选的是,桑黄多糖提取物通过超声协同纤维素酶法从桑黄粉中提取,提取率为2%~5%。
优选的是,桑黄多糖提取物的制备方法包括以下步骤:
脱脂处理:桑黄粉与90%乙醇按料液比1:20~1:30混合,80~85℃回流2~3 h;

一种具有抑菌修护作用的水凝胶敷料的制备方法,包括以下步骤:
S1、称取明胶粉末置于容器中,并向其中加入金银花纯露,持续搅拌,使明胶完全溶解得到凝胶水溶液;
S2、将桑黄多糖溶液、保湿剂、营养剂加入凝胶水溶液中继续搅拌均匀,冷却至室温,即可形成所需水凝胶敷料。
优选的是,凝胶水溶液中明胶质量浓度为10%~20%,搅拌温度为40~50℃。
本发明的优点:

<实施例1>
(1)桑黄多糖的制备:
取5.0 g桑黄粉于250 mL圆底烧瓶中,加入90%的乙醇溶液125 mL,置于80℃的油浴锅内回流两小时,在该过程中桑黄粉中的脂质会溶出且溶于乙醇,两小时后,将烧瓶内的溶液进行抽滤,之后再加入90%的乙醇溶液125 mL,重复上述操作,进而使桑黄粉中的脂质全部溶出。抽滤得脱脂后桑黄粉,置于60℃真空干燥箱烘干至恒重,并将脱脂后的桑黄粉封存于自封袋保存备用。
取3.0 g脱脂后桑黄粉加入80 mL的去离子水中,加入0.5 g纤维素酶,反应温度为45℃,提取90 min。然后升温至80℃,灭酶5 min。在温度为80℃、功率为360 w的条件下超声1 h后,待温度降至50℃后,调节体系的pH至4。之后将上述溶液进行离心,由于所要提取的多糖溶于水,所以保留上清液,同时,为了使残渣去除干净,将离心后的上清液再进行抽滤,保留滤液。再将滤液进行旋转浓缩至20 mL左右,得到高浓度提取液。由于多糖不溶于乙醇,故而可向提取液中加入79%的乙醇使提取液中的多糖沉淀,桑黄浓缩液与乙醇体积比为1:5。静置12 h,使沉淀完全,再将醇沉后溶液进行离心,取沉淀,最后将沉淀冷冻干燥,得到桑黄多糖提取物,红外谱图如图1所示,其他提取方法如下表1所示。

(2)金银花纯露的制备:
将新鲜的3 kg金银花枝叶剪段放至蒸馏瓶中,按照料液比为1:2添加6 kg去离子水,设置蒸馏温度为120℃,待水沸腾开始产生水蒸气则开启冷凝装置,设置加热温度为100℃,3 h后,将蒸馏出的芳香物在收集装置进行冷却、静置,收集下层的液体则为金银花纯露。
(3)成品的制备:
按照质量比重加入下列物质。将浓度为10%的明胶2份加入步骤(2)所制备的金银花纯露20份,50℃下制备明胶水溶液作为A相备用。步骤(1)制备的7份桑黄多糖加入A相中,随后加入保湿剂乙酰化透明质酸钠3份,营养剂维生素C 2份,搅拌均匀。直至产品冷却至室温,凝固成为成品试样。
<测试实验>
(1)抗菌性能实验
将160 μL水凝胶预聚液平铺到一个48孔板中,进行蓝光光照30 min,形成水凝胶支架。再配制105 CFU/L的金黄色葡萄球菌菌悬液。将70 μL菌悬液涂布到M-H平板上,再将水凝胶支架平铺到M-H平板上,在恒温37℃环境中培养1 d,对平板上的抑菌圈进行观察并测量。如表所示,试样对于金黄色葡萄球菌的抑制作用较好,如表2所示。

(2)水凝胶性能测试实验(溶胀比实验)
将试样浸没到磷酸盐缓冲溶液中,在恒温37℃环境下放置2 d。再取出,吸干材料表面多余的磷酸盐缓冲溶液,称重记录,记为V1。之后,再将试样干燥,称重记录,记为V2。然后计算水凝胶溶胀比,公式为:
实验结果参见图2,结果表明试样水凝胶溶胀比较好。
(3)创面愈合情况
创面愈合情况:根据患者面部皮肤缺损修补治疗手术中的创面愈合治疗和时间等,对创面愈合情况进行评价,具体分为优、良、差3个等级。并以此计算创面愈合优良率。创面瘢痕增生情况:根据温哥华瘢痕量表,对各组患者术后一定时间的创面瘢痕增生情况进行VSS评价。该评价采用Linker六级评分,即0~5分。VSS评分越高,则代表患者术后一定时间的创面瘢痕增生情况越严重。患者满意度情况:采用调查问卷的方式,对面部皮肤缺损修补治疗手术术后6个月的患者进行满意度访问。该评价总分设定为10分,其中,低于6分为不满意;6~7分表示满意;8~10分表示非常满意。以此计算患者满意度。本研究中各数据均使用SPSS 25.0软件进行处理,其中,“P<0.05”表示各组数据差异存在统计学意义。选取进行面部皮肤缺损修补治疗的患者92例。并进行随机分组,对照组和试验组的面部皮肤缺损修补治疗患者例数均为46例。患者纳入标准为:(1)知晓且自愿同意参与本研究;(2)患者年龄在18~60岁;(3)在面部受伤6h内接受面部手术治疗;(4)患者均为面部皮肤缺损。患者排除标准为:(1)曾接受过同类手术治疗;(2)患者有精神类疾病或感染类疾病;(3)患者的心、肝、肾功能严重异常。在面部皮肤缺损修补治疗中,对照组的各患者均通过普通皮瓣修复术治疗,试验组的各患者则均通过水凝胶敷料进行治疗。各组患者换药日期、包扎固定方式等均无差异。
实验结果参见下表3,试样水凝胶的创面愈合情况较好。

(4)皮肤刺激性情况
使用0.5 mL水凝胶溶液进行试验,根据《消毒技术规范》(2002年版)中多次完整皮肤刺激试验方法进行。在试验前24 h,将豚鼠背部脊柱两侧的毛去掉,不得损伤皮肤。去毛范围,左、右各约3 cm×3 cm。试验时,将水凝胶溶液0.5 mL直接滴于面积为2.5 cm×2.5cm的2层~4层纱布上并敷贴在左侧去毛皮肤表面,然后用一层无刺激塑料膜覆盖,再用无刺激胶布固定。右侧涂0.5 mL 0.9%氯化钠注射液作对照,在涂抹后4 h,用温水清洗,除去残留物。每天涂抹一次,连续涂抹14 d,每次涂抹受试物4 h,用水或无刺激的适宜溶剂清洗残留受试物,在涂抹受试物24 h观察动物皮肤的红斑和水肿情况,根据皮肤刺激反应的评分标准进行评分,发现该水凝胶对豚鼠的多次完整皮肤刺激强度属无刺激性,符合《消毒技术规范》(2002年版)的要求。
实验结果如下表4所示,样品几乎无刺激性。

上述实施案例只为说明本发明的技术方案及特点,其目的在于更好的让熟悉该技术的人士予以实施,并不能以此限制本发明的保护范围,凡根据本发明精神实质所作的等效变化或修饰,均在本发明保护范围之内,其中未详细说明的为现有技术。
Claims (7)
1.一种具有抑菌修护作用的水凝胶敷料,其特征在于,包括以下重量份的各组分:
金银花纯露 18-22份
桑黄多糖提取物 6-8份
明胶 1-3份
保湿剂 2-4份
营养剂 1-3份;
所述水凝胶敷料的制备方法,包括以下步骤:
S1、称取明胶粉末置于容器中,并向其中加入金银花纯露,持续搅拌,使明胶完全溶解得到凝胶水溶液;
S2、将桑黄多糖溶液、保湿剂、营养剂加入凝胶水溶液中继续搅拌均匀,冷却至室温,即可形成所需水凝胶敷料;
凝胶水溶液中明胶质量浓度为10%~20%,搅拌温度为40~50℃。
2.根据权利要求1所述的一种具有抑菌修护作用的水凝胶敷料,其特征在于,桑黄多糖提取物通过超声协同纤维素酶法从桑黄粉中提取,提取率为2%~5%。
3.根据权利要求2所述的一种具有抑菌修护作用的水凝胶敷料,其特征在于,桑黄多糖提取物的制备方法包括以下步骤:
脱脂处理:桑黄粉与90%乙醇按料液比1:20~1:30混合,80~85℃回流2~3 h;
酶解超声:脱脂桑黄粉按固液比1:20~1:30加水,加入0.5~1.0 g生物酶,40~55℃酶解90 min,80~100℃灭酶5~10 min,再经300~360 w超声0.5~2 h;
纯化:酶解液与79%乙醇按体积比1:5混合沉淀10~12 h,离心干燥得桑黄多糖提取物。
4.根据权利要求3所述的一种具有抑菌修护作用的水凝胶敷料,其特征在于,酶解超声步骤中,脱脂桑黄粉加水配置成固液比为1:20~1:30的水溶液,酶解采用的生物酶为纤维素酶、木瓜蛋白酶、果胶酶、淀粉酶中的至少一种。
5.根据权利要求1所述的一种具有抑菌修护作用的水凝胶敷料,其特征在于,保湿剂选自乙酰化透明质酸钠、甘油、聚谷氨酸钠中的至少一种。
6.根据权利要求1所述的一种具有抑菌修护作用的水凝胶敷料,其特征在于,营养剂选自维生素C、泛醇、烟酰胺中的至少一种。
7.根据权利要求1所述的一种具有抑菌修护作用的水凝胶敷料,其特征在于,金银花纯露制备方法为:金银花枝叶按料液比1:2加水,120℃蒸馏3 h,收集下层冷凝液即可得到金银花纯露。
