Abstract
本发明公开了一种治疗和/或改善功能性消化不良和/或小儿厌食的组合物及其制备方法,它是以新鲜中草药沙棘为原料采用压榨、原汁浓缩、分离、萃取提油、制粒、干燥、分装等工序制备获得,通过将汁与油溶为一体制成,既保持了沙棘中的活性成份,又提高脂溶性药物疗效及疾病治愈率。本发明的组合物具有消食化滞,活血散瘀,理气止痛之功效,用于治疗和/或改善功能性消化不良,小儿厌食所致胃腹胀痛,食欲不振,纳差食少,恶心呕吐等症,具有服用和携带方便、易于保存和运输、质量稳定、疗效确切等优点。
Description
本发明属于医药品的制备技术领域,涉及一种治疗和/或改善功能性消化不良和/或小儿厌食疾病的组合物,具体涉及一种以中草药沙棘为原料制备的中成药或保健食品,本发明还涉及该组合物的制备方法。
功能性消化不良、小儿厌食疾病是临床上的常见病、多发病,目前,本领域用于治疗功能性消化不良、小儿厌食疾病的药物已有不少,特别是一些中医药制剂在临床实践中也形成了颇为独到的疗效优势。但由于此类疾病的发病率高且病理机制较为复杂,现有的一些药物制剂尚不能完全满足临床和市场的需要,依然有待于科研机构及人员做进一步的探索以研制出更为理想的中药新药。沙棘为胡颓子科植物沙棘(HippophaeRhamnoidos L.)的新鲜成熟果实,秋、冬果实成熟时采收,除去杂质即得。主产于我国西北、华北及西南广大地区。沙棘性温、味酸,入肝胃、小肠经,在《兰毗璃璃》第49页中记载了沙棘具有止咳化痰、消食化滞、活血化淤的作用,于1977年被卫生部正式列入《中国药典》。由于本领域以往对沙棘的化学成分、药理作用及新剂型的研究较少,兼之受使用途径及其它一些因素的限制,目前尚未有关于用沙棘果汁和沙棘籽油组成的用于治疗和/或改善功能性消化不良及小儿厌食等疾病的组合物的研制报道。
本发明的目的在于针对本领域现有技术存在的问题,提供一种服用方便、易于保存和运输、对治疗和/或改善功能性消化不良和/或小儿厌食疾病有确切疗效的组合物及其制备方法。
本发明所述组合物的组方根据是在总结藏医民间用药的基础上提出的。它是以新鲜中草药沙棘为原料采用压榨、原汁浓缩、分离、萃取提油、制粒、干燥、分装等工序制备获得,通过将汁与油溶为一体制成,它既保持了沙棘中的活性成份,又提高脂溶性药物疗效及疾病治愈率。
本发明提供的组合物的制备方法,包括以下步骤:
(1)取洗净沥干的沙棘果,榨取果汁,果渣另器收集,果汁离心除去上层油状物后,薄膜蒸发浓缩至可溶性固形物为60%~70%,备用;
(2)分取(1)所述果渣中的种子干燥,粉碎成粗粉,用正己烷在70~80℃条件下回流提取2次,第一次加正己烷4倍量,浸泡1小时后,回流3小时:第二次加正己烷2倍量,回流2小时,冷却抽滤,合并滤液,在70℃、-0.05Mpa条件下回收至无溶媒滴出时充氮气25分钟得沙棘籽油;
(3)取上述浓缩汁,沙棘籽油混合均匀,和/或
(4)加入辅料,即制得本发明所述的组合物。
本发明所述的辅料是药学领域制备颗粒剂及乳化剂时的常用辅料,包括赋形剂如庶糖,乳化剂如阿拉伯胶。
本发明所述的组合物具有消食化滞,活血散瘀,理气止痛之功效,用于治疗功能性消化不良和/或小儿厌食所致胃腹胀痛,食欲不振,纳差食少,恶心呕吐等症,具有服用和携带方便、易于保存和运输、质量稳定、疗效确切等优点。
下面通过实施例及毒理药理实验结果对本发明作进一步说明。
实施例
取新鲜沙棘果,榨取果汁,果汁蒸发浓缩至可溶性固形物为60~70%。取果渣中的种子干燥,粉碎成粗粉,用正己烷在70~80℃条件下回流提取2次,在70℃、-0.05Mpa条件下回收至无溶媒滴出时充氮气25分钟得沙棘油。取阿拉伯胶30g与上述沙棘籽油研磨混匀后加上述浓缩果汁中,混匀,加入过80目筛的原糖粉400g搅拌均匀,制粒,60℃以下干燥,整粒,包装即得沙棘干乳剂。
本发明提供的是一种治疗和/或改善功能性消化不良和/或小儿厌食疾病的组合物,为证明其疗效及安全性,发明者使用按上述实施例提供的方法制备得到的沙棘干乳剂进行了毒理和药效学试验,研究结果如下:
一、急性毒性试验
选80只小白鼠(18~22g),雌雄各半,分4组。第1、2、3组鼠灌胃给沙棘干乳剂<上、下午分两次灌胃间隔6h>,剂量分别为20ml/kg次、30ml/kg/次、40mlkg/次。第4组灌胃给水40ml/kg/次。室温下观察7天,各组动7物皆无死亡,结果表明,小白鼠灌胃的LD50大于20g/kg。按化合物急毒分级标准,属无毒性级化合物。
二、长期毒性试验
沙棘干乳剂(溶液)按10ml/kg、15ml/kg及20ml/kg三个剂量给大鼠灌胃,每天一次,连续三个月。每周测量体重,增长正常,每天观察动物活动行为均正常,毛色光亮、润泽,粪便正常,3个月后,取血测定血液指标、肝肾功能,有关的脏器系数及病理检验均与正常组大鼠无明显差异。显示临床试用剂量安全,无明显毒性反应。
三、与功能主治有关的主要药效学试验
<一>抗炎作用
1、巴豆油致小白鼠耳部肿胀试验:用沙棘干乳剂<溶液>5ml/kg和10ml/kg给小鼠灌胃,每天一次,连续3天。对巴豆油所致耳肿胀的抑制率分别为17.3与38.1。
2、醋酸治小白鼠腹膜炎试验: 用沙棘干乳剂<溶液>5ml/kg和10ml/kg给小鼠灌胃,每天一次,连续3天。对小白鼠醋酸致腹膜炎血管通透性有明显抑制作用。
<二>沙棘干乳剂对大白鼠胃蛋白酶的影响
选取体重为250~350克健康大鼠45只,雌雄各半,随机分为5组,沙棘干乳剂按小、中、大剂量<5ml/kg、10ml/kg、20ml/kg>,体重灌胃:对照组喂等体积水和大山楂颗粒(10ml/kg),每天一次,连续20天,于末次给药后禁食12小时,在戊巴比妥麻醉下结扎幽门,然后在十二指肠给药2ml/100g体重,5h取胃液离心后用蛋白毛细管法测胃蛋白酶活力,结果表明沙棘干乳剂可显著增强大白鼠胃液消化蛋白的能力。
(三)对消化道运动功能的影响
1、对家兔离体肠管的作用:取家兔10只,击毙,迅速剖取回盲部段回肠,立即于冷冻的台氏液中保养。实验时剪取长约1.5cm肠段,用DC-001型离体器管测定仪记录,低速20mm/min,待静15min后,描记一段正常收缩曲线。依次给药:沙棘干乳剂按小、中、大剂量(03ml、0.6ml、0.9m1)给药,对照组给0.5ml蒸馏水,大山楂颗粒按小、中剂量(0.3ml、0.6ml)给药,记录曲线,结果表明,沙棘干乳剂对离体家兔回肠平滑肌收缩力有明显的增强作用,同时收缩频率下降。
2、对胃肠推进运动的影响:取体重为18~22g的健康小鼠50只雌雄各半,随机分成5组。实验前禁食12h,不禁水。对照组给蒸馏水、沙棘干乳剂组(小、小、大)按5ml/kg、10ml/kg、20mlkg体重给药,大山楂颗粒组按10ml/kg给药,第二次灌胃给药后2小时,分别灌胃炭末阿拉伯胶混悬液0.1ml/10g,20分钟后,将动物处死测定炭末推进百分率,结果表明:沙棘干乳剂能明显促进小鼠胃肠的推进运动。
Claims (8)
1、一种治疗和/或改善功能性消化不良和/或小儿厌食的组合物,它是由沙棘果汁和沙棘籽油和/或辅料组成。
2、权利要求1所述的组合物,其中的沙棘果汁由新鲜的沙棘果,压榨、浓缩得到。
3、根据权利要求2所述的组合物,其中的沙棘果汁经薄膜蒸发浓缩至可溶性固形物为60-70%。
4、权利要求1-3中任何一个所述的组合物,其中的沙棘籽油是从沙棘种子中萃取提油得到。
5、权利要求4所述组合物,其中的沙棘籽油按下述方法得到:沙棘种子干燥,粉碎成组粉,用正己烷在70-80℃条件下回流提取2次,第一次加正乙烷4倍量,浸泡1小时后,回流3小时;第二次加正乙烷2倍量,回流2小时;冷却抽滤,合并滤液,在70℃、-0.05Mpa条件下回收至无溶媒滴出时充N225分钟得沙棘籽油。
6、权利要求1、2、4或5所述组合物的制备方法:它包括下列步骤:
A、取新鲜沙棘果压榨、浓缩得到沙棘果汁、备用;
B、将沙棘种子萃取提油得沙棘籽油,备用;
C、将上述沙棘果汁和沙棘籽油混合均匀;和/或
D、加入辅料制成。
7、根据权利要求6所述的制备方法,其中步骤A为沙棘果汁经薄膜蒸发浓缩至可溶性固形物为60-70%。
8、根据权利要求6或7所述的制备方法,其中的沙棘籽油按下述方法得到:沙棘种子干燥,粉碎成粗粉,用正乙烷在70-80℃条件下回流提取2次,第一次加正乙烷4倍量,浸泡1小时后,回流3小时,第二次加正乙烷2倍量,回流2小时,冷却抽滤,合并滤液,在70°、-0.05Mpa条件下回收至无溶媒滴出时充N225分钟得沙棘籽油。
