Abstract
本发明公开一种壳聚糖—明胶网络/磷酸钙多孔复合支架材料的制备方法。该方法采用壳聚糖与明胶比例为1∶1,磷酸钙为10-70wt%,通过葡萄糖二醛交联制备而成,其中磷酸钙是羟基磷灰石,或者是α型和β型磷酸三钙。本发明制造的支架材料不但具有良好的力学强度和良好的细胞亲和性,以及大的孔隙率,而且孔隙率和在体内降解速度均具有可调控性,并且没有细胞毒性。
Description
本发明属于生物医学工程组织移植用支架材料制备技术。
本申请人在公开号为“CN 1272384A”申请专利中,公开了一种壳聚糖/明胶网络支架材料的制备方法。依该方法所制备的支架材料生物相容性好,具有大量相互贯通的微孔,孔隙率可达80%以上,亲/疏水平衡性可调,并且有良好的力学强度和细胞亲和力。但是该材料仍然存在着不足之处,如在制备过程中采用了有细胞毒性的戊二醛作为交联剂。虽然微量戊二醛的存在不会对支架材料的细胞亲和性产生影响,但探索生物相容性良好的替代交联剂仍具有积极意义。
本发明的目的在于提出一种壳聚糖-明胶/磷酸钙多孔复合支架材料的制备方法。该方法过程简单,所制造的支架材料不但有良好的力学强度和良好的细胞亲和性,以及大的孔隙率,而且该支架材料的孔隙率和在体内降解速度均具有可调节性,并且完全没有细胞毒性。
为达到上述目的,本发明是通过下述技术方案加以实现的。以壳聚糖、明胶和磷酸钙为原料,将磷酸钙粉末、水和微量乙酸组成弱酸性乳液状混合物,在4-40℃条件下将壳聚糖和明胶溶解,用交联剂进行交联;交联后的产物经冻干处理后,以适量氢氧化钠的乙醇溶液中和材料中的乙酸,用硼氢化钠溶液将材料中残留的醛基转变,然后再经二次冻干、洗涤和去水过程制得支架材料,其特征在于:其中磷酸钙采用的是羟基磷灰石(Hydroxyapatite,简称HA)或者是α型和β型磷酸三钙(Tricalcium phosphate简称TCP),磷酸钙的含量为10-70wt%;其中壳聚糖和明胶的含量比例为1∶1;采用的交联剂是无细胞毒性的葡萄糖二醛。
下面对本发明进行详细说明。
本发明采用了生物相容性良好的天然可降解生物材料,从仿生的角度出发,制备骨组织工程用多孔复合网络支架材料,能够适应成骨细胞粘连和增殖及同组织修复和重建的需要。该支架材料采用壳聚糖、明胶网络和磷酸钙复合而成。
壳聚糖是一种生物可降解的天然聚阳离子多糖(pKa=6.3),结构上类似GAGs,具有良好的生物相容性、生物降解性。它能与细胞膜负电成分呈现非特异相互作用[Razania A andHealy KE.Biomimetic peptide surfaces that regulate adhesion,spreading,cytoskeleted organization,and mieralization of the matrix deposited by osteoblast-like cells.Biotechnol Prog,1999;15:19-32],并可调控生物活性分子释放[Park YG,Lee YM,Park SN,et al.Plateset derived growth factorrelease chitosan spone for perildontal bone regeneration.Biomaterial,2000;21:153-159.],呈碱性,在生物医学方面应用广泛。
明胶是胶原蛋白的部分水解产物,生物相容性良好,且无抗原反应。明胶的结构单元为甘氨酸-脯氨酸和羟脯氨酸与赖氨酸(Gly-Pro-Hyp-Hyl),系柔性两性电解质,等电点pHiso=4.7,有利于细胞粘连。
磷酸钙是骨的主要无机成分。一方面可以增强多孔复合支架材料的力学性能,另一方面通过调控其降解速率,使之与新生组织的长入相匹配。
采用上述原料进行支架材料制备过程中,通过以明胶仿生胶原,以壳聚糖仿生糖胺聚糖,并利用没有细胞毒性的葡萄糖二醛作为交联剂,制备壳聚糖-明胶网络。由于壳聚糖是刚性聚合物,可以有效改善明胶的力学性能较差的弱点,不仅提高了聚合物网络的力学性能,同时保持了壳聚糖和明胶优异的生物相容性。
下面再以实施例地本发明加以进一步说明。
实施例一:
将5gHA加入40g去离子水,搅拌0.5h,然后将其用超声波处理。在处理后的乳液状混合物中加入2.5g壳聚糖,再加入0.8ml冰乙酸,待壳聚糖完全溶解后加入2.5g明胶,当明胶全部溶解后加入一定量的葡萄糖二醛溶液。将该混合物置于40℃的水浴中搅拌1h。将组成一定的混合物倾入塑料培养皿,在4℃下冷藏2小时,然后在-20℃下预冷冻8小时后冻干30小时。以适量NaOH的乙醇溶液中和材料中的乙酸,用硼氢化钠溶液将材料中残留醛基转变,然后将其二次冻干,置于干燥器中。将得到的CS-Gel/HA复合支架材料裁剪成直径7mm,厚1.5mm的圆盘片,置于48孔培养板的孔底,用75%的乙醇溶液浸泡1h,用蒸馏水反复冲洗,除去水分。在密闭环境中有高效紫外灯照射灭菌。灭菌后的复合支架材料用聚乙烯薄膜密封后待用。
实施例二:
HA含量调整为1.5g,壳聚糖和明胶的含量均调整为3.5g,其他制备过程同实施例1。
实施例三:
将3gβ-TCP加入40g去离子水,搅拌0.5h,然后将其用超声波处理。在处理后的乳液状混合物中加入3.5g壳聚糖,再加入0.8ml冰乙酸,待壳聚糖完全溶解后加入2.5g明胶,当明胶全部溶解后加入一定量的葡萄糖二醛溶液。将该混合物置于40℃的水浴中搅拌1h。将组成一定的混合物倾入塑料培养皿,在4℃下冷藏2小时,然后在-20℃下预冷冻8小时后冻干30小时。以适量NaOH的乙醇溶液中和材料中的乙酸,用硼氢化钠溶液将材料中残留醛基转变,然后将其二次冻干,置于干燥器中。将得到的CS-Gel/HA复合支架材料裁剪成直径7mm,厚1.5mm的圆盘片,置于48孔培养板的孔底,用75%的乙醇溶液浸泡1h,用蒸馏水反复冲洗,除去水分。在密闭环境中有高效紫外灯照射灭菌。灭菌后的复合支架材料用聚乙烯薄膜密封后待用。
本发明过程简单,采用的原料来源广泛,价格低廉,所制备出的支架材料有大量的相互贯通的微孔,其孔隙率在80-95%。实验证明,该支架材料有良好的细胞亲和性,并有适宜的力学强度。
Claims (1)
1.一种壳聚糖-明胶网络/磷钙多孔复合支架材料的制备方法,以壳聚糖、明胶和磷酸钙为原料,将磷酸钙粉末、水和微量乙酸组成弱酸性乳液状混合物,在4-40℃条件下将壳聚糖和明胶溶解,用交联剂进行交联;交联后的产物经冻干处理后,以适量氢氧化钠的乙醇溶液中和材料中的乙酸,用硼氢化钠溶液将材料中残留的醛基转变,然后再经二次冻干、洗涤和去水过程制得支架材料,其特征在于:其中磷酸钙采用的是羟基磷灰石,或者是α型和β型磷酸三钙,磷酸钙的含量为10-70wt%;其中壳聚糖和明胶的含量比例为1∶1;采用的交联剂是无细胞毒性的葡萄糖二醛。
