背景技术
炎症,是机体对感染或组织损伤所产生的一种防御措施,可发生在人体的各种 组织和器官中,例如常见的肝炎,胃炎,肺炎等。其中细菌的感染或病毒的感染、 烧伤和割伤、强酸和强碱腐蚀以及人体的免疫系统失调,都可以引起机体的炎症, 将这些因子称为炎症因子。
冷蒿(ArtemisiafrigidaWilld)为菊科(Compositae)蒿属(Artemisia)的一种多年 生草本植物。冷蒿的总黄酮提取物具有明显的抗炎效果。但冷蒿抗炎作用的有效活 性成分及其作用机制尚不清楚。
发明内容
本发明的目的是提供具有抗炎作用的冷蒿醇提取物。
本发明的再一目的是提供上述冷蒿醇提取物的制备方法。
本发明的再一目的是提供上述冷蒿醇提取物的应用。
根据本发明的具有抗炎作用的冷蒿醇提取物,其通过以下方法制备而得,
将冷蒿粉碎并过筛;
加入95%的酒精,放入超声提取机中搅拌均匀进行超声提取,超声功率500W, 温度50℃,时间30min,酒精与冷蒿粉的体积质量比为40:1(mL/g);
超声提取后,将上层提取液倒入离心管中,收集离心后的上清,蒸发浓缩获得 蒿醇提取物。
本发明提供了冷蒿醇提取物的制备方法,所述方法包括以下步骤:
将冷蒿粉碎并过筛;
加入95%的酒精,放入超声提取机中搅拌均匀进行超声提取,超声功率500W, 温度50℃,时间30min,酒精与冷蒿粉的体积质量比为40:1mL/g;
超声提取后,将上层提取液倒入离心管中,收集离心后的上清,蒸发浓缩获得 蒿醇提取物。

具体实施方式
本发明以19种中草药为原料,采用超声提取法,以乙醇作为提取溶剂,分别获 得19种中草药的醇提物,发现不同中草药的抗炎症特性不同。将其作用于LPS诱导 的RAW264.7细胞模型,通过检测提取物对细胞存活率和炎症介质NO、IL-6、1L-1β、 TNF-α的影响,确定抗炎作用研究的对象;
(2)确定抗炎作用研究对象的有效活性成分。利用LPS诱导的小鼠巨噬细胞RAW264.7炎症模型,检测中草药提取物对炎症介质NO和炎症因子IL-6、IL-1β、 TNF-α表达量的影响,确定有效抗炎中草药提取物;利用不同的有机溶剂萃取确定 的抗炎中草药提取物,考察提取物对细胞存活率及炎症介介质NO和炎症因子IL-6、 IL-1β、TNF-α表达量的影响,确定有效组分;利用高效液相制备色谱技术分馏,考 察各馏分对细胞存活率及炎症介质NO和炎症因子IL-6、IL-1β、TNF-α表达量的影 响,确定有效馏分;
实施例1中草药醇提物的制备
1、中草药粉碎
使用微型植物粉碎机将黄花蒿、冷蒿、白头翁、龙蒿、白鲜果皮、多叶棘豆、 广布野豌豆、防风、斜茎黄耆种皮、斜茎黄耆叶、灵芝菌棒、东北珍珠梅、苍术茎 叶、苍术根须、苍耳、波叶大黄、山韭、茜草、地梢瓜瓜皮粉碎,过60目的分析筛, 于密封袋中保存,备用。
2、醇提物的制备
(1)在超声提取机中加入800mL95%酒精,称取黄花蒿粗粉20g放入超声 提取机中搅拌均匀进行超声提取,超声条件:超声功率500W,温度50℃,时间30 min。
(2)超声提取后,取出容器,将上层提取液倒入离心管中,5000rpm离心20min, 将离心后的上清收集或转移至旋转蒸发瓶中,用旋转蒸发仪旋转浓缩至5mL左右, 用移液枪将液体转移至合适的容器中,用保鲜膜包上,并在上端扎几个孔,放入-80℃ 冰箱,待冻成固体后,将其放入冻干机中,冻干成粉末,将保鲜膜取下来,盖上离 心管的盖子,作为待测样品,做好标记,放入-80℃冰箱备用;
以同样的方法制备冷蒿、白头翁、龙蒿、白鲜果皮、多叶棘豆、广布野豌豆、 防风、斜茎黄耆种皮、斜茎黄耆叶、灵芝菌棒、东北珍珠梅、苍术茎叶、苍术根须、 苍耳、波叶大黄、山韭、茜草、地梢瓜瓜皮的乙醇提取物。
称取醇提物10mg置于1.5mLEP管中,在无菌的环境中配置成浓度为10mg/mL 的母液,用移液枪吹打混匀或者上下颠倒,使其溶解,用无菌的0.45μm滤膜过滤至 新的无菌离心管中,放入﹣80℃冰箱,保存备用。
实施例2中草药提取物的抗炎作用
1、醇提物对细胞存活率的影响
确定实施例1的中草药提取物对细胞的存活率有无影响。将各提取物以不同的 浓度(80μg/mL、100μg/mL、150μg/mL、200μg/mL、250μg/mL)作用于RAW264.7 细胞,用CCK-8试剂检测其对细胞存活率的影响。
由图1~图4可知,上述19种中草药的醇提取物质作用于RAW264.7细胞后, 其中广布野豌豆醇提物、防风醇提物、斜茎黄耆叶醇提物、东北珍珠梅醇提物、苍 术茎叶醇提物、苍耳醇提物、波叶大黄醇提物、茜草醇提物和地梢瓜瓜皮的醇提物 都会对细胞产生不同程度的损伤。
2、醇提物的抗炎作用
研究无细胞毒性的黄花蒿醇提物、冷蒿醇提物、白头翁醇提物、龙蒿醇提物、 白鲜果皮醇提物、多叶棘豆醇提物、斜茎黄耆种皮醇提物、灵芝菌棒醇提物、苍术 根须醇提物、山韭醇提物的抗炎作用。
以LPS诱导的小鼠单核巨噬细胞RAW264.7为模型,设置对照组、LPS组(LPS+ 培养基)、LPS+加药组。将各提取物以100μg/mL的浓度处理RAW264.7细胞,2小 时后,除对照组外,均加入终浓度为1000μg/mL的LPS共培养24小时,收集细 胞上清,检测抗炎因子的含量。
利用ELISA试剂盒检测各醇提物对细胞上清中NO、IL-6、IL-1β和TNF-α表达 量的影响。
(2-1)中草药醇提物对NO含量的影响
由图5可知,黄花蒿醇提物、冷蒿醇提物、白头翁醇提物和龙蒿醇提物处理组 的NO含量的含量高于对照组,低于LPS模型组,说明黄花蒿醇提物、冷蒿醇提物、 白头翁醇提物和龙蒿醇提物均可抑制炎症介质NO的释放,抑制NO释放的能力为 黄花蒿醇提物>冷蒿醇提物=白头翁醇提物>龙蒿醇提物。
(2-2)中草药醇提物对IL-6含量的影响
由图6可以看出,与LPS模型组相比,黄花蒿醇提物、冷蒿醇提物、苍术根须 醇提物处理RAW264.7细胞后,IL-6的含量没有显著的变化。白头翁醇提物和灵芝 菌棒的醇提物处理RAW264.7细胞后,不仅没有降低细胞上清中IL-6的含量,反而 增加。龙蒿醇提物、白鲜果皮醇提物、多叶棘豆醇提物、斜茎黄耆种皮醇提物和山 韭的醇提物使细胞上清中IL-6的含量明显下降,这说明它们可以抑制细胞上清中 IL-6的释放。抑制IL-6的能力为龙蒿醇提物=白鲜果皮醇提物=多叶棘豆醇提物 =山韭醇提物>斜茎黄耆种皮醇提物。
(2-3)中草药醇提物对IL-1β含量的影响
由图7可知,黄花蒿醇提物和山韭的醇提物作用于细胞后,炎症因子IL-1β的含 量没有下降反而上升,其余8种醇提物与LPS模型组相比,没有显著的变化,说明 这10种中草药的醇提物对细胞上清中IL-1β没有显著的抑制作用。
(2-4)中草药醇提物对TNF-α含量的影响
由图8可知,白鲜果皮醇提物和山韭醇提物TNF-α的表达量比对照组高,比LPS 组高,说明白鲜果皮醇提物和山韭醇提物都可以抑制细胞上清中TNF-α的释放与表 达。抑制TNF-α的能力为山韭醇提物>白鲜果皮醇提物。其余8种醇提物对TNF-α 的释放与表达没有明显的抑制作用。
综上所述,由图5~图8可知,醇提物的抗炎作用各不相同,其中冷蒿的醇提物 可较好的抑制炎症介质NO的释放。
Claims (5)
1.具有抗炎作用的冷蒿醇提取物,其特征在于,所述冷蒿醇提取物通过以下方法制备而得:
将冷蒿粉碎并过筛;
加入95%的酒精,放入超声提取机中搅拌均匀进行超声提取,超声功率500W,温度50℃,时间30min,酒精与冷蒿粉的体积质量比为40:1mL/g;
超声提取后,将上层提取液倒入离心管中,收集离心后的上清,蒸发浓缩获得蒿醇提取物。
2.冷蒿醇提取物的制备方法,其特征在于,所述方法包括以下步骤:
将冷蒿粉碎并过筛;
加入95%的酒精,放入超声提取机中搅拌均匀进行超声提取,超声功率500W,温度50℃,时间30min,酒精与冷蒿粉的体积质量比为40:1mL/g;
超声提取后,将上层提取液倒入离心管中,收集离心后的上清,蒸发浓缩获得蒿醇提取物。
3.权利要求1所述的具有抗炎作用的冷蒿醇提取物用于抑制炎症因子的应用。
4.根据权利要求3所述的应用,其特征在于,根据本发明的应用,所述炎症因子为一氧化氮。
5.权利要求1所述的具有抗炎作用的冷蒿醇提取物用于制备抑制炎症因子一氧化氮的制剂的应用。
