Abstract
本发明提供一种具有降血糖功效的茄皮组合物及其制备方法和应用,所述茄皮组合物按重量份计,包括以下组分:茄皮醇提物40‑50份、桑叶醇提物10‑20份、葛根醇提物10‑20份和青钱柳醇提物10‑20份。本发明的茄皮组合物充分发挥天然活性物质的生物活性,具有协同作用,作用靶点多,可以通过刺激胰岛素释放、增加胰岛素敏感性、抑制α‑糖苷酶或增加外周糖的利用等机制有效地起到降血糖的作用。本发明的茄皮组合物具有降低血糖、增强免疫、防治并发症以及毒副作用小等优点,而且所用原料廉价易得,生产成本低,保健价值大,是高血糖患者的福音。
Description
技术领域
本发明涉及降血糖药物领域,更具体地,涉及一种具有降血糖功效的茄皮组合物及其制备方法和应用。
背景技术
正常情况下,人体能够通过激素调节和神经调节这两大调节系统确保血糖的来源与去路保持平衡,使血糖维持在一定水平。但是在遗传因素(如糖尿病家族史)与环境因素(如不合理的膳食、肥胖等)的共同作用下,两大调节功能发生紊乱,就会出现血糖水平的升高。糖尿病是血糖升高容易导致的一种疾病,其已经成为人类的主要死因之一。
目前临床上治疗糖尿病采取静脉注射人胰岛素类似物和口服降糖药,但是长期应用机体易产生低血糖、胰岛素抵抗和耐受性不佳等不良反应,导致血糖控制不佳,容易引起急性心血管疾病等事件的发生,且用药受限,不能适应现在快节奏的生活需求和不断提升的生活品味。
天然产物具有多途径、多靶点、副作用小等特点,国内外医药工作者已把治疗糖尿病的目标从化学药物转向从天然产物中筛选降血糖的活性成分,充分发挥天然活性成分在治疗糖尿病及其并发症上的独特优势。
发明内容
针对现有技术存在的问题,本发明提供一种具有降血糖功效的茄皮组合物及其制备方法和应用。
本发明第一目的为提供一种具有降血糖功效的茄皮组合物,按重量份计,包括以下组分:茄皮醇提物40-50份、桑叶醇提物10-20份、葛根醇提物10-20份和青钱柳醇提物10-20份。
茄子属茄科茄属植物,果的形状有长或圆,颜色有白、红、紫等,国内外广泛种植。茄子的根、茎、叶、花、蒂、果实均可药用,具有清热血、祛风利湿、散瘀消肿、止痛等功效;具体地,茄根和茎用于治疗风湿热痹、久痢不止、女阴挺出以及冻疮等症;茄叶主治血淋、血痢、肠风下血、痈肿、冻伤;茄花主治创伤、牙痛;茄蒂可治疗肠风下血;果实晒干研末可治乳腺溃烂。
本发明研究发现,之前被废弃的茄皮中含有一定比例的酚酸类和黄酮类物质,具有很好的降血糖功效,将茄皮醇提物与桑叶醇提物、葛根醇提物和青钱柳醇提物配伍,充分发挥这些天然活性物质的生物活性,具有协同作用,可以通过刺激胰岛素释放、增加胰岛素敏感性、抑制α-糖苷酶或增加外周糖的利用等机制有效地起到降血糖的作用。本发明解决了茄皮浪费的问题,并且为今后茄皮的深加工提供了一种可行路径。
进一步地,所述茄皮醇提物为紫色茄皮的醇提物。
进一步地,所述茄皮醇提物的活性成分包括酚酸类和黄酮类物质。
进一步地,所述酚酸类和黄酮类物质的含量为30%~65%,大于茄皮醇提物中其他活性成分的含量。
进一步地,所述茄皮组合物还包括黄芪醇提物和枸杞醇提物,按重量份计,所述茄皮组合物包括以下组分:茄皮醇提物40-50份、桑叶醇提物10-20份、葛根醇提物10-20份、青钱柳醇提物10-20份、黄芪醇提物10-20份和枸杞醇提物10-20份。
本发明发现黄芪提取物中主要含有多糖、皂苷类、黄酮、氨基酸等活性成分;枸杞提取物中含有枸杞多糖、黄酮多酚、类胡萝卜素、生物碱和其它化合物等活性成分;黄芪和枸杞提物中的多糖成分能够刺激胰岛素分泌,在所述茄皮组合物中加入这两种组分,降血糖效果进一步增强,且增长幅度较大。
本发明中的各醇提物可以由市购获得,也可以购买新鲜优质材料然后自行加工处理,具体制备方法为:(1)将新鲜紫茄皮冻干粉碎过60目筛,用30倍量的60%的食品级乙醇超声辅助提取1h,收集上清液冻干粉碎并过80目筛备用。(2)购买优质的葛根、桑叶、青钱柳、黄芪、枸杞粉碎过60目筛,加入40-50倍量的70%食品级乙醇40℃超声波辅助提取1.5h,收集提取液冻干粉碎过80目筛备用。
本发明第二目的为提供一种具有降血糖功效的制剂,包括上述茄皮组合物和药学上接受的辅料。
进一步地,所述制剂为片剂、胶囊剂、丸剂或冲剂等常见剂型。其中优选制成口含片,方便携带和服用。
本发明第三目的为提供上述茄皮组合物的制备方法,包括:将各组分分别过筛,然后按配比混合均匀。优选地,将各组分过80目筛。
本发明第四目的为提供上述茄皮组合物在制备治疗糖尿病的药物中的应用。
本发明的有益效果:
本发明的茄皮组合物将茄皮醇提物与桑叶醇提物、葛根醇提物、青钱柳醇提物配伍,充分发挥这些天然活性物质的生物活性,具有协同作用,作用靶点多,可以通过刺激胰岛素释放、增加胰岛素敏感性、抑制α-糖苷酶或增加外周糖的利用等机制有效地起到降血糖的作用。本发明的茄皮组合物具有降低血糖、增强免疫、防治并发症以及毒副作用小等优点,而且所用原料廉价易得,生产成本低,保健价值大,是高血糖患者的福音。
具体实施方式
以下实施例用于说明本发明,但不用来限制本发明的范围。实施例中未注明具体技术或条件者,按照本领域内的文献所描述的技术或条件,或者按照产品说明书进行。所用试剂或仪器未注明生产厂商者,均为可通过正规渠道商购买得到的常规产品。
以下实施例中,紫茄皮、葛根、青钱柳、桑叶、黄芪和枸杞均为市购的优质材料,然后进行醇提得到相应醇提物;淀粉、木糖醇、甘露醇等辅料为直接市购获得。
原料制备:(1)将新鲜紫茄皮冻干粉碎过60目筛,用30倍量的60%的食品级乙醇超声辅助提取1h,收集上清液冻干粉碎并过80目筛备用。(2)购买优质的葛根、桑叶、青钱柳、黄芪、枸杞粉碎过60目筛,加入40-50倍量的70%食品级乙醇40℃超声波辅助提取1.5h,收集提取液冻干粉碎过80目筛备用。
实施例1
本实施例提供一种具有降血糖功效的茄皮组合物,由以下组分组成:茄皮醇提物40份、桑叶醇提物20份、葛根醇提物20份和青钱柳醇提物20份。
实施例2
本实施例提供一种具有降血糖功效的茄皮组合物,由以下组分组成:茄皮醇提物50份、桑叶醇提物10份、葛根醇提物10份、青钱柳醇提物10份、黄芪醇提物10份和枸杞醇提物10份。
实施例3
本实施例提供一种茄皮口含片,其制备方法如下:
将紫茄皮烘干粉碎过60目筛,用超声辅助乙醇浸提提取其活性成分,冷冻干燥后粉碎过80目筛备用;将葛根、青钱柳、桑叶、黄芪、枸杞,分别用高速中药粉碎机粉碎过60目筛,再利用乙醇超声辅助提取其活性成分,将提取物制成干粉并过80目筛备用;准备淀粉、甘露醇、木糖醇,过80目筛备用。分别称取紫茄皮醇提物40份、葛根醇提物10份、青钱柳醇提物10份、桑叶醇提物10份、黄芪醇提物10份、枸杞醇提物10份、淀粉10份和甘露醇10份,充分混匀过80目筛,置于压片机压制成片剂并灭菌和包装。
实施例4
本实施例提供一种茄皮口含片,其制备方法如下:
将紫茄皮烘干粉碎过60目筛,用超声辅助乙醇浸提提取其活性成分,冷冻干燥后粉碎过80目筛备用;将葛根、青钱柳、桑叶、黄芪、枸杞用高速中药粉碎机粉碎过60目筛,再利用乙醇超声辅助提取其活性成分,将提取物制成干粉并过80目筛备用;准备淀粉、甘露醇、木糖醇、过80目筛备用。分别称取紫茄皮醇提物45份、葛根醇提物15份、青钱柳醇提物15份、桑叶醇提物15份、黄芪醇提物15份、枸杞醇提物15份、淀粉15份和甘露醇15份,充分混匀过80目筛,置于压片机压制成片剂并灭菌和包装。
实施例5
本实施例提供一种茄皮口含片,其制备方法如下:
将紫茄皮烘干粉碎过60目筛,用超声辅助乙醇浸提提取其活性成分,冷冻干燥后粉碎过80目筛备用;将葛根、青钱柳、桑叶、黄芪、枸杞用高速中药粉碎机粉碎过60目筛,再利用乙醇超声辅助提取其活性成分,将提取物制成干粉并过80目筛备用;准备淀粉、甘露醇、木糖醇、过80目筛备用。分别称取紫茄皮醇提物50份、葛根醇提物20份、青钱柳醇提物20份、桑叶醇提物20份、黄芪醇提物20份、枸杞醇提物20份、淀粉20份和甘露醇20份,充分混匀过80目筛,置于压片机压制成片剂并灭菌和包装。
实施例6
本实施例提供一种茄皮口含片,组分为紫茄皮醇提物40份、葛根醇提物20份、青钱柳醇提物20份、桑叶醇提物20份、淀粉10份和甘露醇10份,其制备方法同实施例3。
对比例1
本对比例提供一种茄皮口含片,组分为紫茄皮醇提物90份、淀粉10份和甘露醇10份,其制备方法同实施例3。
对比例2
本对比例提供一种茄皮口含片,组分为葛根醇提物30份、青钱柳醇提物30份、桑叶醇提物30份、淀粉10份和甘露醇10份,其制备方法同实施例3。
实验例
对实施例提供的茄皮口含片的降血糖活性进行评价测试:
一、试验方法
1、造模:选取健康成年的SD雄性大鼠约250只,体重150±20g,随机选取20只作为空白对照组,其余进行造模:24h后,腹腔注射链唑霉素(剂量为45mg/kg),注射72h后,不禁水禁食6h,进行尾部取血检测血糖;取血糖值≥16.8mM的大鼠继续进行造模,饲喂高脂饲料;空白对照组在同等条件腹腔注射相同计量的生理盐水,饲喂普通饲料。四周后,尾部静脉取血检测血糖以及测24h尿蛋白含量,以空腹血糖值≥16.8mM,同时尿蛋白质≥20mg/24h,判定糖尿病大鼠造模成功。
2、给药:将造模成功的大鼠随机分成10组,每组20只,分别为模型组、阳性对照组、实施例3低剂量组(即以低剂量给予实施例3中的茄皮口含片)、实施例3中剂量组(即以中剂量给予实施例3中的茄皮口含片)、实施例3高剂量组(即以高剂量给予实施例3中的茄皮口含片,其余以此类推)、实施例4高剂量组、实施例5高剂量组、实施例6高剂量组、对比例1高剂量组和对比例2高剂量组。将对应的茄皮口含片研磨后加纯净水配置成溶液进行灌胃,每日现配,给药量:低剂量组150mg/kg、中剂量组200mg/kg、高剂量组400mg/kg;给阳性对照组的大鼠灌胃盐酸二甲双胍,给药量为200mg/kg;模型组和空白对照组在等同条件下灌胃生理盐水。每组每日灌胃一次,连续灌胃30天。
3、检测:末次灌胃后2h后进行尾部静脉取血,测大鼠糖含量以及糖化血红蛋白含量,结果见表1所示。

由表1可知,相比于空白对照组,模型组的空腹血糖和糖化血红蛋白的含量均大幅度升高,因此说明糖尿病大鼠造模成功。与模型组相比,给药组和阳性对照组的空腹血糖值和糖化血红蛋白的含量均有所下降,但对比例1-2的下降不明显,只有实施例3-6的给药组下降明显,呈显著性差异,其中,以药物高剂量组的下降更为明显。这表明本发明实施例的茄皮口含片对糖尿病有一定的治疗效果,紫茄皮醇提物、葛根醇提物、青钱柳醇提物和桑叶醇提物相辅相成,缺少其中一种或几种组分都不能取得较好的降血糖功效。
实施例3-6中,实施例4、实施例5的结果与实施例3类似,实施例6相较实施例3-5的下降效果差一点,说明黄芪醇提物和枸杞醇提物这两种组分能使降血糖效果进一步增强,且增长幅度较大。
虽然,上文中已经用一般性说明及具体实施方案对本发明作了详尽的描述,但在本发明基础上,可以对之作一些修改或改进,这对本领域技术人员而言是显而易见的。因此,在不偏离本发明精神的基础上所做的这些修改或改进,均属于本发明要求保护的范围。
Claims (7)
1.一种具有降血糖功效的茄皮组合物,其特征在于,按重量份计,由以下组分组成:紫茄皮醇提物40-50份、桑叶醇提物10-20份、葛根醇提物10-20份、青钱柳醇提物10-20份、黄芪醇提物10-20份和枸杞醇提物10-20份。
2.根据权利要求1所述的茄皮组合物,其特征在于,所述紫茄皮醇提物的活性成分包括酚酸类和黄酮类物质。
3.根据权利要求2所述的茄皮组合物,其特征在于,所述酚酸类和黄酮类物质的含量为30%~65%。
4.一种具有降血糖功效的制剂,其特征在于,包括权利要求1~3任一项所述的茄皮组合物和药学上接受的辅料。
5.根据权利要求4所述的制剂,其特征在于,所述制剂为片剂、胶囊剂、丸剂或冲剂。
6.权利要求1~3任一项所述的茄皮组合物的制备方法,其特征在于,包括:将各组分分别过筛,然后按配比混合均匀。
7.权利要求1~3任一项所述的茄皮组合物在制备治疗糖尿病的药物中的应用。
