本发明涉及医药保健品技术领域,具体是一种抗病毒生物肽护理液的制备方法。本发明将脂肪酶,蛋白酶,超氧化物歧化酶和水混合,得混合酶,将磷脂与豆甾醇混合,接着加入混合酶和无水乙醇,得1号混合液,将过硼酸钠与磷酸盐缓冲溶液混合,接着加入牛奶磷脂,胆固醇,维生素C和乙醇搅拌混合,即得2号混合液,将壳聚糖与水混合,静置溶胀后,加热搅拌溶解,降温,随后加入壳聚糖酶,恒温搅拌反应,灭酶,得酶化壳聚糖液,将抗病毒肽,氯化钠,糖蛋白,磷脂,羽毛水解浓缩液,1号混合液,2号混合液,酶化壳聚糖液搅拌混合,即得抗病毒生物肽护理。本发明提供的抗病毒生物肽护理液具有优异的杀菌和抗菌的作用。

本发明涉及医药保健品技术领域,具体是一种抗病毒生物肽护理液的制备方法。
背景技术
隐形眼镜(contactlens),或角膜接触镜,是一种戴在眼球角膜上,用以矫正视力或保护眼睛的镜片。它包括硬性和软性两种,隐形眼镜不仅从外观上和方便性方面给近视、远视、散光等屈光不正患者带来了很大的改善,而且视野宽阔、视物逼真,此外在控制青少年近视、散光发展,治疗特殊的眼病等方面也发挥了特殊的功效。
越来越多的人使用隐形眼镜,使得隐形眼镜行业发展迅速,这也使得隐形眼镜护理液多种多样。已有隐形眼镜消毒护理液产品品种很多,常见的有酸、碱、氯制剂、高锰酸钾、双氧水等。抗菌消毒性能不一,有高、中、低效之分。往往是高效消毒剂的有效成分及含量与价格成正比;且有毒性、腐蚀性、刺激性气味,对眼睛有损害:清洗消毒护理不能同步,因蛋白质的沉淀影响佩带者的舒适度和眼镜的性能增加了并发感染的可能性,可能造成角膜损伤。因此,隐形眼镜行业一直希望能有一种抗菌消毒效率高、安全稳定、对眼睛无损坏、使佩带者感到安全的隐形眼镜消毒护理液。
隐形眼镜护理液的主要成分是氯化钠和防腐剂等,主要用于对生物材料的清洗、消毒等,对于配戴隐形眼镜的人来说,无疑是必不可少的。但部分消费者反映配带隐形眼镜后出现眼部发红,感觉不适等不良反应。其主要原因是配带者在隐形眼镜护理液的使用中存在一些误区造成的。目前隐形眼镜护理液的杀菌效果不够理想,无法达到预期的效果,从而导致对人体造成了一定的损害具有一定的安全隐患。
发明内容
本发明的目的在于提供一种抗病毒生物肽护理液的制备方法,以解决现有技术中的问题。
为实现上述目的,本发明提供如下技术方案:
一种抗病毒生物肽护理液,包括以下重量份数的原料:5~8份抗病毒肽,2~3份氯化钠,2~3份糖蛋白,2~3份磷脂,8~10份羽毛水解浓缩液,8~10份1号混合液,8~10份2号混合液,20~30份酶化壳聚糖液。
所述糖蛋白为猪血清蛋白,羊血清蛋白或马血清蛋白中的任意一种。
所述磷脂为大豆磷脂。
所述羽毛水解浓缩液包括以下重量份数的原料:2~3份鸽毛,0.01~0.02份地衣芽胞杆菌,2~3份葡萄糖溶液,10~20份水。
所述混合酶包括以下重量份数的原料:2~3份脂肪酶,2~3份蛋白酶,1~2份超氧化物歧化酶,10~20份水。
所述1号混合液包括以下重量份数的原料:20~30份蛋黄磷脂,10~20份豆甾醇,2~3份混合酶,3~5份无水乙醇。
所述2号混合液包括以下重量份数的原料:5~8份过硼酸钠,60~80份磷酸盐缓冲溶液,20~30份牛奶磷脂,8~10份胆固醇,3~5份维生素C,30~80份乙醇。
所述酶化壳聚糖液包括以下重量份数的原料:2~3份壳聚糖,100~120份水,0.1~0.2份壳聚糖酶。
所述抗病毒生物肽护理液包括以下重量份数的原料:8份抗病毒肽,3份氯化钠,3份糖蛋白,3份磷脂,10份羽毛水解浓缩液,10份1号混合液,10份2号混合液,30份酶化壳聚糖液
一种抗病毒生物肽护理液的制备方法,具体制备方法为:
(1)水解鸽毛;
(2)制备混合酶;
(3)制备1号混合液;
(4)制备2号混合液;
(5)制备酶化壳聚糖液;
(6)混料;
(7)检测。
所述抗病毒生物肽护理液具体制备步骤如下:
(1)将鸽毛,葡萄糖溶液,水恒温混合,接着加入地衣芽胞杆菌,恒温搅拌发酵,升温灭菌,过滤,浓缩,得羽毛水解浓缩液,在使用过程中,由于原料中的磷脂带有负电荷,可与1号混合液中的糖蛋白中带正电荷的疏水端产生静电吸附,并在隐形眼镜表面形成以磷脂,糖蛋白和酶化壳聚糖和羽毛水解蛋白形成双亲型隔膜,由于双亲形成的隔膜的存在,增加了隐形眼镜的舒适度,同时双亲型的隔膜能够良好的负载体系中的抗病毒肽,使得体系中的抗病毒肽能够良好的分散并附着在隐形眼镜表面,起到了良好的抗病毒作用;
(2)将脂肪酶,蛋白酶,超氧化物歧化酶和水混合,得混合酶;
(3)将磷脂与豆甾醇混合,接着加入混合酶和无水乙醇,得1号混合液;
(4)将过硼酸钠与磷酸盐缓冲溶液混合,接着加入牛奶磷脂,胆固醇,维生素C和乙醇搅拌混合,即得2号混合液,在使用过程中,首先,1号混合液和2号混合液中都含有脂质体,脂质体对细菌生物膜具有良好的靶向性,因此使得产品能够更加有效快速的作用与体系中细菌细胞膜,从而使得产品的杀菌性能得到提升,其次,混合酶脂质体在与隐形眼镜表面的细菌生物膜接触后,脂质体中的混合酶可快速破坏细菌生物膜,从而在生物膜表面形成孔隙或裂缝,随着脂质体添加剂与隐形眼镜表面的细菌生物膜接触,脂质体添加剂中的过氧化物可从混合酶在细菌生物膜表面形成的孔隙和裂缝进入细菌生物膜,对生物膜中的氨基进行氧化,使氨基氧化为硝基,使原本带正电荷的氨基转变成带负电的硝基,从而使细菌生物膜表面的防护层因同种电荷相互排斥而脱落,细菌生物膜可快速解离从隐形眼镜表面除去,从而使得产品的杀菌性能得到进一步的提升;
(5)将壳聚糖与水混合,静置溶胀后,加热搅拌溶解,降温,随后加入壳聚糖酶,恒温搅拌反应,灭酶,得酶化壳聚糖液;
(6)将抗病毒肽,氯化钠,糖蛋白,磷脂,羽毛水解浓缩液,1号混合液,2号混合液,酶化壳聚糖液搅拌混合,即得抗病毒生物肽护理液;
(7)产品性能检测。
所述抗病毒生物肽护理液具体制备步骤如下:
(1)将鸽毛,质量分数为0.3~0.5倍的葡萄糖溶液,水置于1号烧杯中,接着将1号烧杯置于数显测速恒温磁力搅拌器中,于温度为30~35℃,转速为400~600r/min条件下,恒温搅拌混合40~60min,得混合浆体,接着将混合浆体与地衣芽胞杆菌置于发酵釜中,于温度为30~35℃,转速为300~500r/min条件下,恒温搅拌发酵3~5天,接着升温至90~95℃灭菌5~10min后,过滤,得滤液,接着将滤液置于旋转蒸发仪中,于温度为60~70℃,压力为500~600Pa条件下,浓缩40~60min,得羽毛水解浓缩液;
(2)将脂肪酶,蛋白酶,超氧化物歧化酶和水置于单口烧瓶中,于转速为300~500r/min条件下,搅拌混合40~60min,得混合酶;
(3)将磷脂与豆甾醇置于2号烧杯中,于转速为600~800r/min条件下,搅拌混合40~60min,接着向2号烧杯中加入混合酶和无水乙醇,于转速为400~600r/min条件下,加班混合40~60min后,得1号混合液;
(4)将过硼酸钠与磷酸盐缓冲溶液置于3号烧杯中,接着将3号烧杯置于数显测速恒温磁力搅拌器中,于温度为40~45℃,转速为400~600r/min条件下,接着向3号烧杯中加入牛奶磷脂,胆固醇,维生素C和乙醇,于温度为40~45℃,转速为500~600r/min条件下,搅拌混合40~60min,即得2号混合液;
(5)将壳聚糖与水置于4号烧杯中,并用玻璃棒搅拌10~20min后,静置溶胀3~5h后,将4号烧杯置于数显测速恒温磁力搅拌器中,于温度为90~95℃,转速为400~600r/min条件下,加热搅拌溶解40~60min后,降温至30~35℃,随后向4号烧杯中加入壳聚糖酶,于温度为30~35℃,转速为400~600r/min条件下,恒温搅拌反应40~60min,随后升温至90~95℃灭酶5~10min后,得酶化壳聚糖液;
(6)将抗病毒肽,氯化钠,糖蛋白,磷脂,羽毛水解浓缩液,1号混合液,2号混合液,酶化壳聚糖液置于混料机中,于转速为1000~1200r/min条件下,高速搅拌混合40~60min,即得抗病毒生物肽护理液;
(7)产品性能检测。
所述抗病毒生物肽护理液具体应用:用水把手掌润湿,把隐形眼镜镜片放在手掌,正面朝下,在镜片上滴抗病毒生物肽护理液,用另一只手中指的指腹轻轻的顺时针方向柔,两只隐形眼镜镜片都一样洗完后镜片先放在盒盖上,这个时候把盒子里原来的抗病毒生物肽护理液倒掉,清洗一下,换上新的抗病毒生物肽护理液,然后把隐形眼镜镜片放入盒子,既可。
与现有技术相比,本发明的有益效果是:
(1)本发明在使用过程中,由于原料中的磷脂带有负电荷,可与1号混合液中的糖蛋白中带正电荷的疏水端产生静电吸附,并在隐形眼镜表面形成以磷脂,糖蛋白和酶化壳聚糖和羽毛水解蛋白形成双亲型隔膜,由于双亲形成的隔膜的存在,增加了隐形眼镜表面的润滑度,有利于隐形眼镜贴合眼球,增加了隐形眼镜的舒适度,同时双亲型的隔膜能够良好的负载体系中的抗病毒肽,使得体系中的抗病毒肽能够良好的分散并附着在隐形眼镜表面,起到了良好的抗病毒作用;
(2)本发明在使用过程中,首先,1号混合液和2号混合液中都含有脂质体,脂质体对细菌生物膜具有良好的靶向性,因此使得产品能够更加有效快速的作用与体系中细菌细胞膜,从而使得产品的杀菌性能得到提升;
其次,混合酶脂质体在与隐形眼镜表面的细菌生物膜接触后,脂质体中的混合酶可快速破坏细菌生物膜,从而在生物膜表面形成孔隙或裂缝,随着脂质体添加剂与隐形眼镜表面的细菌生物膜接触,脂质体添加剂中的过氧化物可从混合酶在细菌生物膜表面形成的孔隙和裂缝进入细菌生物膜,对生物膜中的氨基进行氧化,使氨基氧化为硝基,使原本带正电荷的氨基转变成带负电的硝基,从而使细菌生物膜表面的防护层因同种电荷相互排斥而脱落,细菌生物膜可快速解离从隐形眼镜表面除去,从而使得产品的杀菌性能得到进一步的提升。

为了更清楚的说明本发明提供的方法通过以下实施例进行详细说明,在以下实施例中制作的抗病毒生物肽护理液的各指标的测试方法如下:
分别取试件样片(2cm×3cm),置于无菌平皿内,在每个样片上均匀滴加试验菌悬液0.1mL,作用5min,将样片投入含5.0mL中和剂的试管中,充分混匀并中和作用10min;然后将其充分震荡洗脱,取洗脱液0.5mL倾注接种平皿,倒入融化的营养琼脂培养基,混匀,凝固后置于37℃培养48h,作活菌计数,计算抑菌率;试验重复3次,取平均值。
实例1
一种抗病毒生物肽护理液,包括以下重量份数的原料:8份抗病毒肽,3份氯化钠,3份糖蛋白,3份磷脂,10份羽毛水解浓缩液,10份1号混合液,10份2号混合液,30份酶化壳聚糖液。
所述糖蛋白为猪血清蛋白。
所述磷脂为大豆磷脂。
(1)按重量份数计,依次取3份鸽毛,0.02份地衣芽胞杆菌,3份葡萄糖溶液,20份水,将鸽毛,质量分数为0.5倍的葡萄糖溶液,水置于1号烧杯中,接着将1号烧杯置于数显测速恒温磁力搅拌器中,于温度为35℃,转速为600r/min条件下,恒温搅拌混合60min,得混合浆体,接着将混合浆体与地衣芽胞杆菌置于发酵釜中,于温度为35℃,转速为500r/min条件下,恒温搅拌发酵5天,接着升温至95℃灭菌10min后,过滤,得滤液,接着将滤液置于旋转蒸发仪中,于温度为70℃,压力为600Pa条件下,浓缩60min,得羽毛水解浓缩液;

(5)按重量份数计,依次取3份壳聚糖,120份水,0.2份壳聚糖酶,将壳聚糖与水置于4号烧杯中,并用玻璃棒搅拌20min后,静置溶胀5h后,将4号烧杯置于数显测速恒温磁力搅拌器中,于温度为95℃,转速为600r/min条件下,加热搅拌溶解60min后,降温至35℃,随后向4号烧杯中加入壳聚糖酶,于温度为35℃,转速为600r/min条件下,恒温搅拌反应60min,随后升温至95℃灭酶10min后,得酶化壳聚糖液;
(6)按重量份数计,依次取8份抗病毒肽,3份氯化钠,3份磷脂,10份羽毛水解浓缩液,10份1号混合液,10份2号混合液,30份酶化壳聚糖液,将抗病毒肽,氯化钠,磷脂,羽毛水解浓缩液,1号混合液,2号混合液,酶化壳聚糖液置于混料机中,于转速为1200r/min条件下,高速搅拌混合60min,即得抗病毒生物肽护理液;
(7)产品性能检测。
