本发明公开了一种烟草丝裂原活化蛋白激酶基因NtMAPK8及其应用,所述烟草丝裂原活化蛋白激酶NtMAPK8基因核苷酸序列如SEQ ID NO:1或SEQ ID NO:3所示,编码的氨基酸序列如SEQ ID NO:2或SEQ ID NO:4所示;本发明首次克隆烟草丝裂原活化蛋白激酶基因NtMAPK8,并构建得到了克隆载体及通过基因编辑载体构建NtMAPK8基因缺失的转基因突变株,从而使烟草叶片颜色由正常的绿色变成花白;因此可以利用基因编辑载体使NtMAPK8基因不表达,进行植物品种育种,获得植物花叶的观赏植物品种。
Description
技术领域
本发明涉及分子生物学技术领域,尤其涉及一种烟草丝裂原活化蛋白激酶基因NtMAPK8及其应用。
背景技术
在植物中,烟草是重要的一种经济作物。烟草叶片的颜色,直接影响烟草烘烤后的烟丝色泽,其为衡量烟叶品质的一个重要指标。
MAPKKK-MAPKK-MAPK级联途径在植物中非常保守,其不仅在植物的防御系统中扮演着重要的角色,还在植物的生长发育中也起着十分重要的调节作用。但是目前尚未有MAPK基因在叶片颜色方面的相关报道。
发明内容
针对现有技术的不足,本发明提供了一种烟草丝裂原活化蛋白激酶基因NtMAPK8,其如SEQ ID NO:1或SEQ ID NO:2所示,该基因编码的氨基酸序列如SEQ ID NO:3或SEQ IDNO:4所示。
本发明另一目的是提供含有上述的烟草丝裂原活化蛋白激酶基因NtMAPK8的重组载体。
本发明所述的重组载体包括克隆载体和表达载体;所述的克隆载体的制备方法包括以烟草的cDNA为模板,用如SEQ ID NO:5和SEQ ID NO:6所示的引物,进行PCR扩增;将所得PCR产物连接到T载体上,转化大肠杆菌获得克隆载体。
本发明另一目是提供一种基因敲除载体,其是利用CRISPR/Cas9基因编辑技术对烟草NtMAPK8基因设计靶位点后基因编辑获得,所述靶位点敲除序列的核苷酸序列如SEQID NO:7所示;在所述靶位点两侧加上人工合成的接头序列后得到的引物对的核苷酸序列如SEQ ID NO:8和SEQ ID NO:9所示;将所述引物对经退火后,连接到经BsaI酶切的载体pORE-Cas9上,获得基因敲除载体pORE-Cas9/NtMAPK8。
本发明另一目是提供一种转基因植物,所述转基因植物的制备方法包括:将基因敲除载体pORE-Cas9/NtMAPK8转化农杆菌感受态细胞,再转化侵染植物即得。
所述转基因植物为转基因烟草;转化侵染转基因烟草时采用pORE-Cas9/NtMAPK8载体转化农杆菌感受态,利用农杆菌介导的烟草叶盘转化法对烟草进行遗传转化,获得不同颜色的烟草植株。具体地,其叶片颜色部分由绿色变成浅绿色。
本发明另一目是将上述的烟草丝裂原活化蛋白激酶基因NtMAPK8应用在制备不同叶片颜色植物品种中。
与现有技术相比,本发明具有如下优点和效果:
本发明首次克隆获得烟草丝裂原活化蛋白激酶基因NtMAPK8,并构建得到了克隆载体及敲除载体;NtMAPK8蛋白参与MAPK信号传递,可通过基因编辑载体构建NtMAPK8基因缺失的转基因突变株,从而使烟草叶片颜色由正常的绿色变成花白,具体原因仍然不清楚。
本发明所得的NtMAPK8基因敲除的突变株系,其叶片颜色部分变成白色,因此可以利用基因编辑载体使NtMAPK8基因基因不表达,进行植物品种育种,获得植物花叶的观赏植物品种。

具体实施方式
下面结合附图和实施例对本发明作进一步详细说明,但本发明保护范围不局限于所述内容,实施例中使用的试剂和方法,如无特殊说明,均采用常规试剂和使用常规方法。
本实施例中涉及的生物材料、实验试剂和耗材等情况如下:
烟草品种:红花大金元,其种子由西南大学家蚕基因组生物学国家重点实验室提供;
载体:CRISPR/Cas9基本敲除载体由西南大学家蚕基因组生物学国家重点实验室提供;pEASY®-Blunt Zero Cloning Kit载体,购自北京全式金生物技术有限公司;
菌株:Trans1-T1化学感受态细胞、LBA4404农杆菌化学感受态细胞,均购自北京全式金生物技术有限公司;
引物和测序:引物合成以及DNA测序均由北京华大基因完成;
实验试剂:植物DNA提取试剂盒EasyPure Plant Genomic DNA Kit (含RNase A)和DNA纯化试剂盒EasyPure® PCR Purification Kit PCR均购自北京全式金公司;RNA提取用Trizol® reagent (Invitrogen, USA);反转录试剂盒,TransScript® One-StepgDNA Removal and cDNA Synthesis SuperMix,购自北京全式金公司;T4连接酶、限制性内切酶BsaI、DNA扩增酶,购自NEB公司;PrimeSTAR Max DNA polymerase购自Takara公司。
实验设备:PCR扩增仪器Veriti(基因有限公司);凝胶成像系统紫外凝胶成像仪(BIO-RAD);定量PCR仪器,ABI 7500 fast real-time PCR system(赛默飞)。
实施例1:烟草丝裂原活化蛋白激酶基因NtMAPK8的克隆与序列分析
本发明基于烟草全基因组测序、进行大量生物信息学分析筛选获得了烟草丝裂原活化蛋白激酶基因NtMAPK8;接着以烟草全基因组cDNA为模板克隆烟草丝裂原活化蛋白激酶基因NtMAPK8,具体地:
1、cDNA模板制备
按照制造商的说明,使用试剂(美国Invitrogen公司)从烟草植物的新鲜组织中提取总RNA。使用TransScript® One-Step gDNA Removal and cDNA Synthesis SuperMix(中国全式金公司)将总RNA反转录为cDNA。
2、PCR产物扩增及回收
用Primer5设计引物,引物序列如下:
CDS-F:5’- ATGGGAAGTTCAGGAGATGCATGGAAATT -3’(SEQ ID NO:5所示)
CDS-R: 5’- CTAGTTGTCAACAACCTCAATCAAGCTTG-3’(SEQ ID NO:6所示)
以步骤1中制备的cDNA为模板,并利用设计的引物进行PCR扩增,条件是:98℃2min,28个循环(98℃ 5s,55℃ 15s,72℃ 15s),72℃ 5min。
3、连接
将PCR产物用EasyPure® PCR Purification Kit PCR试剂盒纯化回收,连接到pEASY®-Blunt Zero载体上,体系(5μL):T载体1μL、片段4μL;反应条件:轻轻混合,25℃反应10min。
4、转化
转化到Trans1-T1感受态细胞(冰浴30min→42℃热激30s→冰浴2min),加250μL无抗LB液体培养基,220rpm,37℃培养1h,1500转离心1min,弃部分上清,取100-150μL涂板。
5、挑斑
取1.5mL的离心管,加入10μL ddH2O,挑斑将白枪头打到离心管中,然后左右前后晃动板子,弃白枪头;从10μL ddH2O中吸4μL ddH2O做菌落PCR。
6、测序验证
菌落PCR阳性克隆菌液摇混后,送华大基因测序验证。
经过公司测序后,分析得到基因NtMAPK8的两个转录本:NtMAPK8a基因序列全长2976 bp,其核苷酸序列为SEQ ID NO:1所示,所编码的烟草NtMAPK8a蛋白的氨基酸序列如SEQ ID NO:2所示;NtMAPK8b基因序列全长3947 bp,其核苷酸序列为SEQ ID NO:3所示。所编码的烟草NtMAPK8b蛋白的氨基酸序列如SEQ ID NO:4所示。
实施例2 烟草NtMAPK8基因在不同组织中的表达模式特征分析
利用荧光定量qRT-PCR方法检测NtMAPK8基因在烟草不同组织(根、茎、叶、花等)表达模式。采用FastStart UniversalSYBR Green Master(ROX)Realtime试剂盒(Roche)。选用相对定量的方法,以烟草EF1α基因作为内参基因,模板为上述不同组织下得到的cDNA。每个样品设置3个重复,同时设置阴性对照和阳性对照,反应体系如表1所示。

反应体系充分混匀后放入ABI7500荧光定量PCR系统进行PCR反应,采用三步法的PCR反应程序,利用2-ΔΔCt方法确定目标基因的相对表达量。
结果如图1所示,结果表明,NtMAPK8基因的两个转录本均在花药中表达量最高。
实施例3:基因编辑载体的构建
利用CRISPR/Cas9基因编辑技术对烟草NtMAPK8基因靶位点进行设计,构建NtMAPK8基因敲除载体;对烟草品种进行遗传转化,获得转基因烟草植株;
1、设计靶位点:
Target:GAGGCTAATGATGACTTGACA(如SEQ ID NO:7所示);
人工合成加接头序列
Target-F:GATTGAGGCTAATGATGACTTGACA(如SEQ ID NO:8所示);
Target-R:AAACTGTCAAGTCATCATTAGCCTC(如SEQ ID NO:9所示);
2、敲除载体构建
①单链oligo DNA退火形成双链DNA
将合成的2条单链的引物(Target-F以及Target-R)稀释成50μmol\L,然后退火,退火反应体系如表2所示:

将反应体系PCR仪中95℃孵育3min,孵育后自然冷却至室温。
②表达载体的酶切
表达载体:pORE-Cas9/gRNA,用BsaI酶切表达载体,酶切体系如表3所示,37℃酶切1h,回收酶切产物;

③靶位点连接到表达载体上,连接体系如表4所示,25℃连接10min;

④连接产物转化感受态细胞,经PCR验证获得阳性克隆;经测序并比对后表明克隆成功。
实验例4 烟草NtMAPK8基因敲除植株的获得
1、将构建好的pORE-Cas9/NtMAPK8载体转化农杆菌感受态LBA4404,利用农杆菌介导的烟草叶盘转化法对栽培烟草品种K326进行了遗传转化,获得了具有抗卡那霉素的T0代阳性转化苗,具体转化步骤为:
(1)农杆菌转化
①将构建好的敲除载体加到100μL农杆菌感受态细胞LBA-4404中;
②冰浴30min 后液氮速冻1min,再37℃水浴5min;
③加入1ml YEB液体无抗培养基,28℃、200rpm处理3h 取出;
④ 3000rpm离心3min后,去部分上清,取150μL 涂到含有利福平,链霉素和卡那霉素的平板上;
⑤ 28℃,黑暗倒置培养2-3天;
(2)烟草遗传转化
用MS液体培养基悬浮农杆菌至OD值为0.6-0.8,侵染烟草叶盘10min,后黑暗共培养3天,选择培养至生长出愈伤组织并切芽。
2、分子检测
用特异性引物扩增再生烟草基因组中sgRNA附近的基因组序列是否发生编辑,具体地:提取阳性烟苗叶片基因组DNA,用如SEQ ID NO:8和SEQ ID NO:7所示的引物,以基因组DNA为模板,对目的片段进行PCR扩增,对PCR产物进行纯化回收,连接克隆载体进行测序,用以检测靶位点片段突变形式。
收取靶位点被编辑的T0代转基因阳性植株种子,本实验中T0代转基因株系敲除靶位点的基因编辑情况图如表5所示;

由上可知,烟草植株突变系中NtMAPK8基因被成功编辑。
3、转基因株系的表型观察
图3为本发明实验中T0代转基因株系的表型图,由图3可知,NtMAPK8基因敲除的突变株系,其烟草叶片颜色由正常的绿色变成花白。
综上可知,本实施例中基于基因编辑载体CRISPR/Cas9,利用叶盘遗传转化方法将该基因敲除载体转化到烟草叶盘中,获得了NtMAPK8基因的敲除株系,其叶片颜色部分变成白色,因此可以利用基因编辑载体使NtMAPK8基因基因不表达,进行植物品种育种,获得植物花叶的观赏植物品种。

Claims (2)
1.烟草丝裂原活化蛋白激酶基因NtMAPK8或NtMAPK8基因敲除载体在制备不同颜色叶片植物品种中的应用,其特征在于,基因NtMAPK8核苷酸序列如SEQ ID NO:1或SEQ ID NO:2所示;
所述的基因敲除载体利用CRISPR/Cas9基因编辑技术对烟草NtMAPK8基因设计靶位点敲除序列后进行基因编辑获得。
2.根据权利要求1所述的烟草丝裂原活化蛋白激酶基因NtMAPK8或NtMAPK8基因敲除载体在制备不同颜色叶片烟草品种中的应用,其特征在于,所述的基因敲除载体的获取方式为:靶位点敲除序列的核苷酸序列如SEQ ID NO:7所示,在靶位点两侧加上人工合成的接头序列后,得到核苷酸序列如SEQ ID NO:8和SEQ ID NO:9所示的引物对,引物对经退火后,将其连接到经BsaⅠ酶切的载体pORE-Cas9上,即得基因敲除载体pORE-Cas9/NtMAPK8。
