Abstract
本发明涉及一种具有抗氧化、美白和保护细胞活性的酒制滇黄精提取物及其制备方法与应用,该方法首先取酒制滇黄精干燥至水分含量≤4%后,粉碎到40目,加入粉末质量15‑20倍的浓度为55%‑65%食用乙醇,超声提取,过滤,滤液经闪式蒸发浓缩至原体积的1/8‑1/10后,冷冻干燥成粉末,即得。本发明提取物中多糖、多酚、皂苷和黄酮含量分别为39.68%、4.11%、8.54%和0.470%,该提取物具有抗氧化活性与酪氨酸酶抑制活性,在1000pm浓度以内无毒性,并具有保护纤维细胞和角质形成细胞活性,将其添加应用于在日化妆品中,能制备保湿、抗氧化、美白等功能的面膜、精华、护手霜等化妆品,应用前景广泛。
Description
技术领域
本发明属于植物深加工技术领域,具体涉及一种具有抗氧化、美白和保护细胞活性的酒制滇黄精提取物及其制备方法与应用。
背景技术
黄精是中国传统中药材,据2015版《药典》记载,黄精为黄精(Polygonatum sibiricum)、多花黄精(Polygonatum cyrtonema Hua)和滇黄精(Polygonatum kingianum Coll. et Hemsl)的干燥块根。其中滇黄精又名大黄精,主产地为云南,是云南道地药材。滇黄精含有多糖、甾体皂苷、蒽醌、生物碱等化学成分,具有降血糖、降血脂、抗炎抗菌、抗肿瘤、调节免疫等作用。
目前已经有一些专利文章报道了酒制黄精的加工利用方法,但主要集中在相关食品与药品的开发。如ZL2019106839111发明了一种黄精炮制方法;ZL2018105464706发明了利用黄酒和黄精浸泡的酒制黄精炮制方法;ZL2003101032158通过酒制黄精与其他中药材配比,发明了一种治疗贫血的再造生血中药制剂及制备方法。
黄精提取物在国家食品药品监督管理总局发布的《已使用化妆品原料名称目录》中,可以添加进入日化产品中,但是未见相关的发明报道。
发明内容
本发明的目的是为了解决现有技术的不足,提供一种酒制滇黄精提取物及其制备方法与应用,该提取物具备清除自由基、抑制酪氨酸酶、增强细胞活性等功效,在化妆品领域具有广泛的应用前景。本发明促进了该黄精资源的高效利用。
为实现上述目的,本发明采用的技术方案如下:
本发明除非另有说明,否则百分号为质量百分数,比例为质量比。
一种具有抗氧化、美白和保护细胞活性的酒制滇黄精提取物的制备方法,包括如下步骤:
称取酒制滇黄精,干燥至水分含量≤4%后,粉碎到40目,加入粉末质量15-20倍的体积浓度为55%-65%的食用乙醇,超声提取,过滤,滤液经闪式蒸发浓缩至原体积的1/8-1/10后,冷冻干燥成粉末,即得具有抗氧化、美白和保护细胞活性的酒制滇黄精提取物。
进一步,优选的是,所述的超声提取时间为3h-5h。
进一步,优选的是,所述的滤液以55℃~65℃在闪式蒸发器中浓缩
进一步,优选的是,所述的冷冻干燥温度为-55℃,气压为0.12 MPa。
本发明提供上述具有抗氧化、美白和保护细胞活性的酒制滇黄精提取物的制备方法制得的酒制滇黄精提取物。
进一步,优选的是,所述的酒制滇黄精提取物中多糖、多酚、皂苷和黄酮含量分别为39.68%、4.11%、8.54%和0.470%。
本发明还提供上述酒制滇黄精提取物作为日化用品添加剂的应用。
本发明同时提供酒制滇黄精提取物在制备日化用品中的应用。
进一步,优选的是,所述的日化用品为面膜、精华或护手霜。
进一步,优选的是,所述的日化用品为抗衰、美白、修复类日化用品。
本发明所得到的酒制滇黄精提取物为黑褐色粉末,通过福林酚比色法、苯酚-硫酸法、香草醛-高氯酸比色法和硝酸铝比色法测定本发明提取物中多酚、多糖、皂苷和黄酮物质含量分别为4.11%、39.68%、8.54%和0.470%。
本发明酒制滇黄精为普通市售品。
本发明中“粉碎到40目”是指粉末过40目筛,取筛下物。
本发明与现有技术相比,其有益效果为:
本发明制备方法简单可靠,适合大工业生产,制得的提取物对DPPH自由基、超氧阴离子和羟自由基清除率最高分别为94.4%、47.5%、109.7%。该自由基能在体内肆意掠夺其它分子的电子,攻击生物大分子,引发细胞结构和功能被破坏,从而导致皮肤形成色板、皱纹,造成皮肤衰老,本发明提取物能够通过清除自由基达到延缓皮肤衰老等问题
本发明提取物具有酪氨酸酶抑制活性,实验表明,在待测液中加入250 μL浓度为100mg/mL的本发明提取物水溶液情况下抑制率为105.8%,说明本发明酒制滇黄精提取物能利用抑制酪氨酸酶和体外抗氧化能力来抑制黑色素形成,进而具有美白活性,可以开发为美白的化妆品。
本发明提取物在1000ppm浓度内对角质形成细胞及成纤维细胞的活性无影响,对角质细胞核纤维细胞活性有增强作用,表明该提取物对皮肤细胞无毒性,且能增强皮肤组织损伤、重建修复的功能。
本发明提取物应用于制备面膜、护手霜、精华等化妆品,具有保湿、减缓皮肤衰老、增强细胞活性等功能。

下面结合实施例对本发明作进一步的详细描述。
本领域技术人员将会理解,下列实施例仅用于说明本发明,而不应视为限定本发明的范围。实施例中未注明具体技术或条件者,按照本领域内的文献所描述的技术或条件或者按照产品说明书进行。所用材料或设备未注明生产厂商者,均为可以通过购买获得的常规产品。
实施例1
一种具有抗氧化、美白和保护细胞活性的酒制滇黄精提取物的制备方法,包括如下步骤:
称取酒制滇黄精片2.5kg,干燥至水分含量≤4%后,粉碎到40目后加入粉末质量15倍的体积浓度为55%的食用乙醇,采用超声波提取3h;过滤后使用闪式蒸发器,使滤液在55℃-65℃之间通过闪式蒸发器蒸发浓缩至原体积的1/8(呈黑色浓稠状液体),接着在-55℃、0.12MPa下冷冻干燥,得酒制滇黄精提取物600g。
实施例2
一种具有抗氧化、美白和保护细胞活性的酒制滇黄精提取物的制备方法,包括如下步骤:
称取酒制滇黄精片20kg,干燥至水分含量≤4%后,粉碎到40目后加入粉末质量20倍的体积浓度为65%的食用乙醇,采用中试超声波连续逆流提取线提取5h,过滤后使滤液在55℃-65℃之间通过闪式蒸发器蒸发浓缩至原体积的1/10(呈黑色浓稠状液体),接着在-55℃、0.12MPa下冷冻干燥,得酒制滇黄精提取物4.8kg。
实施例3
一种具有抗氧化、美白和保护细胞活性的酒制滇黄精提取物的制备方法,包括如下步骤:
称取酒制滇黄精片20kg,干燥至水分含量≤4%后,粉碎到40目后加入质量18倍的体积浓度为60%的食用乙醇,采用中试超声波连续逆流提取线提取4h,过滤后使滤液在55℃-65℃之间通过闪式蒸发器蒸发浓缩至原体积的1/9(呈黑色浓稠状液体),接着在-55℃、0.12MPa下冷冻干燥,得酒制滇黄精提取物4.9kg。
实施例4 酒制滇黄精提取物的成分检测:
应用福林酚比色法、苯酚-硫酸法、香草醛-高氯酸比色法和硝酸铝比色法测定本发明实施例2提取物中多酚、多糖、皂苷和黄酮物质含量分别为4.11%、39.68%、8.54%和0.470%。
实施例5 酒制滇黄精提取物的OH-自由基清除活性检测:
设置实验组、空白组和对照组。其中实验组和空白组加入25 μL、50 μL、125 μL、250 μL浓度为100mg/mL的本发明提取物水溶液,用蒸馏水补足到250 μL,对照组加入250 μL蒸馏水。之后各组中均加入500 μL蒸馏水,250 μL 7.5 mM FeSO4溶液,充分混匀后在实验组和对照组中加入250 μL 0.01%(v/v)双氧水,空白组中加入250 μL蒸馏水。37℃水浴放置10min,保温结束加入250μL 2.5 mM水杨酸的乙醇溶液,继续置于37℃水浴保温30 min,在520 nm处测定其吸光值。计算公式为:
其中,A1为实验组的吸光度均值;A2为空白组的吸光度均值;A3为对照组的吸光度均值。
测定发现,提取物水溶液添加量为250 μL的时候,清除率最高为109.7%,结果表明提取物羟自由基清除能力明显。
实施例6 酒制滇黄精提取物酪氨酸酶抑制活性检测:
设置实验组、空白组和对照组。其中实验组和空白组加入25 μL、50 μL、125 μL、250 μL浓度为100mg/mL的本发明提取物水溶液,用PBS缓冲液(0.1 M ,pH 6.8)补足到250μL。对照组取250μL PBS缓冲液。加入750μL L-酪氨酸溶液(0.09mg/mL,PBS缓冲液定容)后,于37℃下保温1 h;加入酪氨酸酶溶液(1mg/mL, PBS缓冲液定容)25 μL,于37℃下反应30 min,于450 nm下测定其吸光度值。计算公式为:
其中,A0为对照组吸光度值;A1为实验组吸光度值;A2为空白组吸光度值;
测得结果表明,当提取物水溶液添加量在250 μL的时候,酪氨酸酶抑制活性最高为105.8%。结果表明抑制酪氨酸酶活性显著。
实施例7 酒制滇黄精提取物对角质形成细胞及成纤维细胞的活性检测:
取对数生长期的对角质形成细胞及成纤维细胞作为受试对象,设置溶剂对照孔(对照组,不添加提取物)、调零孔(实验组,添加提取物)和给药孔(SC,添加4%DMSO)。分别设置终浓度为7.8、15.6、31.3、62.5、125、250、500、1000ppm浓度的8组滇黄精提取物(即添加90μL滇黄精提取物溶液至细胞培养板,滇黄精提取物终浓度为上述8个浓度),添加90μL到至少含有40%细胞铺板率的培养板中,于37℃、5%的CO2培养箱中培养24h。检测时,先弃掉上清,加入事先配好的10μL 0.5%MTT 溶液(Solarbio),37℃避光培养。4h后弃掉上清,每孔加100μL 二甲基亚砜,酶标仪490nm 读取 OD 值。计算公式:
根据细胞活性检测结果和形态学检测结果,最终认为样品滇黄精提取物在1000ppm添加范围内无细胞毒性,并且对角质形成细胞和成纤维细胞活性起保护作用。当添加量在62.5ppm时,对角质形成细胞迁移率为108.47%。当添加量在31.3ppm时,对成纤维细胞迁移率为117.37%。
实施例8 酒制滇黄精提取物的应用
① 酒制滇黄精提取物添加在面膜的成分配置
制作面膜母液规格为25mL/片,其中添加各组分量分别为(液体v/v,固体m/v):1%的酒制滇黄精提取物、0.2%龙胆提取物、0.2%积雪草提取物、0.2%黄芪提取物、0.2%母菊花提取物、0.2%迷迭香提取物,3%甘油、2%戊二醇、3%丙二醇中,再搭配2%海藻糖、0.1%辛酰羟肟酸、2%卡波姆、1%PEG-60氢化蓖麻油和0.1%黄原胶。
②酒制滇黄精提取物添加在护手霜的成分配置
制作规格为50mL/瓶,其中添加各组分量分别为(液体v/v,固体m/v):2%的液晶型乳化剂 TUE-20、1%的鲸蜡醇、1.5%的新戊二醇二庚酸酯、3%山茶籽油、5%的角鲨烷、2%的二甲基硅油 DC200、1.5%的季戊四醇四椰油酸酯、0.5%的维E醋酸酯、0.1%的EDTA-2Na、5%的甘油、3%的1,3-丙二醇、0.1%的黄原胶、2%的海藻糖、1.5%的塞比克 305、8%的铁皮石斛提取物、5%的青稞β葡聚糖、2%的IG0798英肤安(云南英格生物技术有限公司)、0.5%的酒制滇黄精提取物、0.5%的PE9010混合与水配置成护手霜。
③酒制滇黄精提取物添加在精华液的成分配置
制作规格为10mL/瓶,其中添加各组分量分别为(液体v/v,固体m/v):0.1%的聚丙烯酸、0.1%的黄原胶(透明)、0.05%的小核菌胶 (粉状)、3%的甘油、5%的1,3-丙二醇、2%的北美金缕梅提取物、2%的仟敏舒、3%的1%透明质酸溶液、2%的海藻糖、0.1%的三乙醇胺、0.5%的酒制滇黄精提取物、0.5%的PE9010,加水配置成精华液。
实施例9 酒制滇黄精面膜使用感官评价:
针对酒制滇黄精面膜各项参数指标及添加的酒制滇黄精提取物的功效,开展感官评价调查。以下为面膜使用情况调查评分表,根据所述情况符合与否进行0-2分的评分,0分为不符合,2分为特别符合,精确到小数点后两位,综合所有评分取平均数即为最终结果。调查人员情况占比:共计50人,其中女性占比40人,男性10人,0-10岁0人,10岁-25岁15人,25岁-60岁35人,60岁以上0人。表1为使用滇黄精面膜感官评价,表2为不添加滇黄精提取物面膜(其余组分含量相同)的感官评价。

上述结果表明,酒制滇黄精提取物能增加面膜保湿能力,在长时间使用情况下,能够体现其美白和抗氧化作用,使得肤色更加红润白皙,且部分人会有斑痕减轻的情况。

1.一种具有抗氧化、美白和保护细胞活性的酒制滇黄精提取物的制备方法,其特征在于,包括如下步骤:
称取酒制滇黄精,干燥至水分含量≤4%后,粉碎到40目,加入粉末质量15-20倍的体积浓度为55%-65%的食用乙醇,超声提取,过滤,滤液经闪式蒸发浓缩至原体积的1/8-1/10后,冷冻干燥成粉末,即得具有抗氧化、美白和保护细胞活性的酒制滇黄精提取物。
2.根据权利要求1所述的具有抗氧化、美白和保护细胞活性的酒制滇黄精提取物的制备方法,其特征在于,所述的超声提取时间为3h-5h。
3.根据权利要求1所述的具有抗氧化、美白和保护细胞活性的酒制滇黄精提取物的制备方法,其特征在于,所述的滤液以55℃~65℃在闪式蒸发器中浓缩。
4.根据权利要求1所述的具有抗氧化、美白和保护细胞活性的酒制滇黄精提取物的制备方法,其特征在于,所述的冷冻干燥温度为-55℃,气压为0.12 MPa。
5.权利要求1-4任意一项所述的具有抗氧化、美白和保护细胞活性的酒制滇黄精提取物的制备方法制得的酒制滇黄精提取物。
6.根据权利要求5所述的酒制滇黄精提取物,其特征在于,所述的酒制滇黄精提取物中多糖、多酚、皂苷和黄酮含量分别为39.68%、4.11%、8.54%和0.470%。
7.权利要求5所述的酒制滇黄精提取物作为日化用品添加剂的应用。
8.权利要求5所述的酒制滇黄精提取物在制备日化用品中的应用。
