技术领域
本发明涉及卫生消毒领域,尤其涉及一种胍类消毒剂的增效技术。
背景技术
胍类消毒剂因其化学结构式中具有生物活性的烷基胍而得名,主要分为双胍类消毒剂和单胍类消毒剂两大类,其中双胍类消毒剂有氯己定、聚六亚甲基双胍盐、聚亚己基双胍、聚胺丙基双胍等;单胍类消毒剂有聚六亚甲基胍盐酸盐聚六亚甲基胍硬脂酸盐、聚六亚甲基胍丙酸盐、聚六亚甲基胍磷酸盐等。
胍类消毒剂在杀菌、消毒与防腐领域均有应用,对细菌繁殖体有很好的杀灭作用,但对抗力较强的真菌和分枝杆菌以及某些亲水性病毒等杀灭效果存在变化,对细菌芽孢基本没有杀灭作用,只能抑制其发芽繁殖,比如醋酸氯己定,对黑曲霉菌杀灭能力较弱,浓度为80mg/L醋酸氯己定对黑曲霉孢子作用20min,平均杀灭对数值只有0.60,故而胍类消毒剂普遍被认为属于低效消毒剂,其单方水溶液只适用于冲洗消毒或抗菌应用,对于消毒要求更高的场合,往往需要和其他消毒成分复配后,才能使用,如醇类消毒剂。因此,发明一种胍类消毒剂的增效技术,对于深化胍类消毒剂的应用具有非常现实的价值和意义。
发明内容
针对现有技术中的上述不足,本发明提供了一种胍类消毒剂的增效技术。
为了达到上述发明目的,本发明采用的具体方案为:
一种胍类消毒剂的增效技术,包括制配胍类消毒剂的增效剂,所述胍类消毒剂中胍活性成分与有机酸盐和表面活性剂的配伍按重量的比例范围为1:0.8~1.5:1.5~2.5,制备成含3%胍的浓缩型增效剂,所述有机酸盐包括乙二胺二琥珀酸二钠、亚氨基琥珀酸二钠、谷氨酸二乙酸四钠、氨基三乙酸三钠、乙二胺四乙酸四钠,所述表面活性剂包括异构脂肪醇(C13/15)聚氧乙烯醚-10、吐温-20、吐温-40、三醋酸乙二胺椰烷基乙酰胺、牛脂醇聚氧乙烯醚-8、异构脂肪醇(C9/11)聚氧乙烯醚-6、椰油酰胺丙基甜菜碱、异构脂肪醇(C9/11)聚氧乙烯醚-6、异构脂肪醇(C12/13)聚氧乙烯醚-7、异构脂肪醇(C12/15)聚氧乙烯醚-7。
作为改进,将配置好的增效剂使用悬液定量法测试杀菌率,用200ppm的PHMB(增效后),对铜绿假单胞菌,在污染条件下,作用5min,对比200ppm的PHMB(增效前)同等条件下,杀灭对数值。
作为改进,所述悬液定量法的操作过程包括:
(1)按产品说明书要求配制消毒液,无特殊说明者,一律使用无菌硬水配制,配制的浓度为待测浓度的1.25倍(例如要评价的消毒液浓度为200mg/L,则应配制250mg/L),置20℃±1℃水浴备用;
(2)配制实验用菌悬液,浓度为1×108cfu/ml~5×108cfu/ml;
(3)取消毒试验用无菌大试管,先加入0.5ml试验用菌悬液,再加入0.5ml有机干扰物质,混匀,置20℃±1℃水浴中5min后,用无菌吸管吸取上述浓度消毒液4.0ml注入其中,迅速混匀并立即记时;
(4)待试验菌与消毒剂相互作用至各预定时间,分别吸取0.5ml试验菌与消毒剂混合液加于4.5ml经灭菌的中和剂中,混匀;
(5)各管试验菌与消毒剂混合液经加中和剂作用10min后,分别吸取1.0ml样液,按活菌培养计数方法测定存活菌数,每管样液接种2个平皿即可。如平板上生长的菌落数较多时,可进行系列10倍稀释后,再进行活菌培养计数;
(6)同时用稀释液代替消毒液,进行平行试验,作为阳性对照;
(7)所有试验样本均在37℃温箱中培养,对细菌繁殖体培养48h观察最终结果;对细菌芽孢需培养72h观察最终结果;
(8)试验重复3次,计算各组的活菌浓度(cfu/ml),并换算为对数值(N),然后计算杀灭对数值。
作为改进,所述悬液定量法使用的公式为:杀灭对数值(KL)=对照组平均活菌浓度的对数值(No)-试验组活菌浓度对数值(Nx),数值取小数点后两位。
