Abstract
一种神经(鞘)磷脂的提取工艺及其在化妆品和
食品中的应用,其特征在于将动物脏器粉碎,丙酮抽
提胆固醇,热乙醇抽提神经(鞘)磷脂(含卵磷脂及可
溶蛋白),乙醚抽提除去卵磷脂,丙酮洗涤除去蛋白,
得可用于化妆品和食品的神经(鞘)磷脂。所得神经
(鞘)磷脂经硅胶双向薄层色谱及磷定量检验,神经
(鞘)磷脂含量≥%,胆固醇≤3%,水分<1%。
Description
本发明涉及神经(鞘)磷脂类(Sphingomyelins)的提取以及用神经(鞘)磷脂作脂质体(Liposomes)膜材用于日用化妆品和食品添加剂。
神经(鞘)磷脂类是非甘油性磷脂,在生物体内分布广泛,功能重要,但含量较低。曾有从红细胞提取神经(鞘)磷脂的报道(1961),但技术流程复杂,产量低,成本高,仅用作生化试剂。(Hanahan D.J.,Biochem.Prep 1961;8:121)
Hanahan报告的方法主要内容为:抗凝牛血10L,4℃下离心取红血球,生理盐水清洗后再离心,得红血球3.9L。所得红细胞加0.25体积蒸馏水混合室温静置3~5′,加3体积95%乙醇,搅拌后室温放置2h。加入1体积二乙乙醚搅拌后,置4h,过滤取滤液保存(4℃),重复上述步骤,合并滤液,放置减压除去有机溶剂后,层析柱分离,洗脱液低温冷藏后收集沉淀物,真空干燥,得2.2g神经(鞘)磷脂,其中卵磷脂含量不大于5%。产量/原料为0.022%,生产中使用乙醇13L,二乙乙醚5L以及其它药品。
本发明的目的在于避免上述现有技术中的不足之处而提供一种用动物脏器作为神经(鞘)磷脂的提取原料,且工艺简单、成本低、产量高,可广泛应用于化妆品和食品之中。
本发明的目的可以通过以下措施来达到:
1、理论根据:
a、动物脏器含有较高比例的神经(鞘)磷脂,例如:主动脉38.1%(占脂类的百分比,下同),脑5.6%,周围神经30.2%,肺11.1%。
b、不同脂类在不同有机溶剂中溶解度不同。

2、工艺流程、
脏器粉碎,丙酮抽提胆固醇,热乙醇抽提神经(鞘)磷脂(含卵磷脂及可溶蛋白),乙醚抽提除去卵磷脂,丙酮洗涤除去蛋白,得可用于化妆品和食品的神经(鞘)磷脂(同时可得胆固醇,卵磷脂)。
如使用引产胎儿脏器为原料,可得人源性神经(鞘)磷脂。
本发明相比现有技术具有如下优点:
现用作脂质体膜料的磷脂主要为卵磷脂和脑磷脂。它们最大的缺点是不稳定性,在空气中常温下极易氧化变质,失去效果。和卵磷脂、脑磷脂相比,神经(鞘)磷脂不仅在空气中、光照下稳定,而且含有皮肤间质重要成分神经酰氨。所以用它作成的脂质体添加到化妆品中,得到特有效果。另外,由于它的稳定和多方面功能,作为食品脂质体添加剂也将受到重视。
本品经检测无毒性,无刺激性,无过敏性,卫生诸指标符合国家有关规定。
用神经(鞘)磷脂制作的脂质体,颗粒细少、分布均匀,形态多为球形,少数为推进器形(电镜照片图象分析,直径平均124.25±44.33nm,(50~300nm))。
制作的单纯神经(鞘)磷脂质体及神经(鞘)磷脂-蛇油脂质体添加到化妆品后人群实验结果好。添加量10-4时,皮肤润泽、舒适。每日一次,一周后皮肤变细嫩、有弹性、皱纹减少。一月后皮肤黄褐斑变淡。同时不粘、不腻、通气性好。
实施例1、
猪脑匀浆200g+丙酮300ml,搅拌4h,过滤。
滤饼加二倍量无水乙醇,60℃水溶。搅拌4h,过滤,重复操作3次。
合并滤液,-15℃下析出沉淀。冷过滤,减压干燥,得粗品。
粗品加二倍量乙醚,于三角烧瓶或烧杯内盖封,振荡4h,取沉淀物,重复乙醚洗涤一次。
乙醚沉淀物丙酮洗涤2次,冷冻干燥,得神经(鞘)磷脂0.8g。
实施例2、
牛坐骨神经匀浆100g+丙酮200ml,搅拌4h,过滤。
以下步骤同实施例1、
得神经(鞘)磷脂2.5g。
所得神经(鞘)磷脂经硅胶双向薄层色谱及磷定量检验,神经(鞘)磷脂含量≥96%,胆固醇≤3%,水份<1%。
实施例3、脂质体-蛇胆霜
硬脂酸12%,硬脂酸醇8%,羊毛脂4%,甘油10%,香料1%,防腐剂适量,精水加至100%,磷脂酸1.5%,胆固醇0.5%,神经(鞘)磷脂0.01%,蛇胆0.1%。
将磷脂酸、胆固醇、神经(鞘)磷脂、蛇胆制成脂质体,其余部分制成膏体,然后充分混合即成。
脂质体-蛇胆霜特别适用于油性皮肤。
实施例4、脂质体口服液
海藻浓缩液3%,大豆提取液3%,白糖1%,蜂蜜1%,香料适量,防腐剂适量,NaHCO30.2%,脑活素0.1%,Vitc0.1%,神经(鞘)磷脂0.01%,磷脂酸1%,精水加至100%。
将脑活素,VitC,神经(鞘)磷脂,磷脂酸制备成脂质体,然后和其他成分共同配成脂质体口服液。
此种口服液活性成分受到磷脂保护,具有明显健脑功能。
Claims (3)
1、一种神经(鞘)磷脂的提取工艺及其在化妆品和食品中的应用,其特征在于将动物脏器粉碎,丙酮抽提胆固醇,热乙醇抽提神经(鞘)磷脂(含卵磷脂及可溶蛋白),乙醚抽提除去卵磷脂,丙酮洗涤除去蛋白,得可用于化妆品和食品的神经(鞘)磷脂。
2、根据权利要求1所述的神经(鞘)磷脂在化妆品中的应用,其特征在于将磷脂酸、胆固醇、神经(鞘)磷脂、蛇胆制成脂质体,硬脂酸、硬脂酸醇、羊毛脂、甘油、香料、防腐剂制成膏体,然后充分混合即成脂质体-蛇胆霜。
3、根据权利要求1所述的神经(鞘)磷脂在食品中的应用,其特征在于将脑活素、Vitc、神经(鞘)磷脂、磷脂酸制备成脂质体,然后和海藻浓缩液、大豆提取液、白糖、蜂蜜、香料、防腐剂、NaHCO3共同配成脂质体口服液。
