Abstract
本发明涉及生物技术领域,具体涉及基因MsGCHI和MsADCS在提高植物叶酸含量及促进植物生长中的应用。在温室条件下,转基因株系叶中叶酸及前体含量均显著高于野生型,且转基因株系叶中叶酸含量显著高于茎,茎中前体含量高于叶,说明MsGCHI和MsADCS的共表达能够显著提高百脉根中叶酸及前体含量。同时,转基因百脉根株系GA‑3的生长速度明显优于野生型,转基因百脉根株系GA‑11的分枝数和根长均显著优于野生型,说明叶酸生物强化对百脉根生长没有不利影响,且叶酸生物强化合成的内源叶酸还能促进转基因百脉根株系的生长。

叶酸(Folate)是一种水溶性B族维生素(维生素B9),是动植物生长发育所必需的微量营养素,其作为C1单位的供体或受体,参与嘌呤、泛酸盐和蛋白质等的生物合成、还原型辅酶Ⅱ(NADPH)的生成以及甲硫氨酸循环等生化途径,在动植物机体新陈代谢和生长发育过程中发挥着十分重要的作用,而人类和其他动物普遍存在叶酸摄入不足的问题。植物和微生物在体内可以合成叶酸,但人类与其他哺乳动物由于缺乏完整的生物合成系统,自身无法合成叶酸,必须依靠饮食获取,植物通常是动物主要的叶酸来源。不同食物中的叶酸含量差异较大,在动物肝脏、豆类和绿色组织中含量比较丰富,而四大粮食作物中叶酸的含量却很低,所以叶酸缺乏导致的营养不良是一个广泛而持续的世界性健康问题,叶酸摄入不足会引发各种发育缺陷和疾病,如巨幼细胞贫血症、神经管缺陷和心血管疾病等。
叶酸还被作为一种重要的饲料添加剂广泛应用于集约化和现代化的畜牧业生产中。研究发现饲料中的叶酸含量已不能满足舍饲饲养下优良畜禽品种的快速生长和高产性能的需求,因此必须通过提高牧草自身的叶酸含量或在日粮中添加叶酸辅酶和甲基供体,为动物的生长提供保证。
为解决叶酸匮乏这一问题,人们尝试了多种方法,比如调整饮食结构、添加人工合成的营养剂和食品强化。尽管人工合成的叶酸(Folic acid,FA)在一定程度上能够改善叶酸摄入不足的问题,但其弊端亦不容忽视。当前人工合成的叶酸几乎全部采用化学法生产,副反应多,且废水中含有大量的氨氮及无机盐对环境造成严重污染。此外,近年来研究表明,服用较高剂量人工合成的叶酸对人体具有一定副作用,包括基因突变导致人体叶酸代谢受损、掩盖维生素B12缺乏症状和致癌等不良影响。通过生物强化手段提高作物中的叶酸含量,是解决食物中叶酸普遍缺乏问题的有效策略。大量研究表明,GTP环化水解酶I(GCHI)与氨基脱氧分支酸合酶(ADCS)是植物叶酸合成过程中最主要的两种限速酶,二者编码基因GCHI和ADCS的共表达能够显著提高诸多转基因作物的叶酸含量。
紫花苜蓿(Medicago sativa)被誉为“牧草之王”,其营养价值高,提取物和浓缩物除含有蛋白质、维生素C、铜、锰、叶酸、核黄素、镁、铁等多种成分外,还含有生物碱、氨基酸、香豆素、消化酶和黄酮类等多种次生代谢产物。此外,研究发现目前种植的牧草中,紫花苜蓿是叶酸含量最为丰富的豆科牧草,这为研究牧草叶酸生物合成提供了良好的供体材料,发明人所在的课题组从紫花苜蓿中发现并克隆出了叶酸合成关键酶GCHI和ADCS的编码基因MsGCHI和MsADCS。
在前期研究的基础上,发明人所在的课题组构建了MsGCHI和MsADCS基因的双价植物表达载体,并将其导入优良豆科牧草百脉根(Lotus corniculatus)中,通过分子鉴定筛选出目的基因表达量相对一致、且表达水平较高的转基因百脉根株系;在温室条件下,对上述株系中的叶酸及其组分含量进行分析,并系统评价其生长性状。
发明内容
针对上述技术问题,本发明的首要目的是提供了基因MsGCHI和MsADCS在促进植物生长中的应用,所述的基因MsGCHI的核苷酸序列如SEQ ID No.1所示,所述的基因MsADCS的核苷酸序列如SEQ ID No.3所示。
优选的,所述的基因MsGCHI的氨基酸序列如SEQ ID No.2所示;所述的基因MsADCS的氨基酸序列如SEQ ID No.4所示。
本发明的第二目的是提供基因MsGCHI在提高植物叶酸含量的应用,所述的所述的基因MsGCHI的核苷酸序列如SEQ ID No.1所示。
本发明的第三目的是提供基因MsADCS在提高植物叶酸含量的应用,所述的基因MsADCS的核苷酸序列如SEQ ID No.3所示。
本发明的第四目的是提供基因MsGCHI和MsADCS在提高植物叶酸含量的应用,所述的基因MsGCHI的核苷酸序列如SEQ ID No.1所示,所述的基因MsADCS的核苷酸序列如SEQID No.3所示。
优选的,所述的促进植物根系生长包括促进根部分支数增多、促进根长变长和促进根体积变大。
本发明的有益效果是:本发明提供了基因MsGCHI和MsADCS在提高植物叶酸含量和促进植物生长中的应用,在温室条件下,转基因株系叶中叶酸及前体含量均显著高于野生型,且转基因株系叶中叶酸含量显著高于茎,茎中前体含量普遍高于叶。说明MsGCHI和MsADCS的共表达能够显著提高百脉根中叶酸及前体含量,并且百脉根中前体的转运也是影响叶酸合成的重要因素之一。同时,转基因百脉根株系的生长速度和分枝数均优于野生型。说明叶酸生物强化对百脉根生长没有不利影响,且叶酸生物强化合成的内源叶酸还能促进转基因百脉根株系的生长。

以下实施例所用pBIB-BASTA-GWR-YFP和pCAMBIA1302-Bar为本实验室保存;

实施例一、基因MsGCHI和MsADCS全序列的合成
1.载体构建的引物设计
表1载体构建引物设计
采用Primer 5.0分别设计MsGCHI、MsADCS和Bar基因以及Gmubi启动子的特异性引物,并在相应序列的5’和3’端分别引入Spe I(P1)与Bst EII(P2)、Xho I(P5)与Xho I(P6)、Hind III(P7)与Kpn I(P8)酶切位点及保护碱基(表1);另外在MsGCHI基因下游引物5′端加上FLAG标签序列(CATTACAAGGATGACGACGATAAG),在MsADCS基因的下游引物5′端中加入HA标签序列(TACCCATACGATGTTCCAGATTACGCT)。
以pMD19-T-GCHI,pMD19-T-ADCS质粒为模板,P1、P2,P3、P4为特异引物,进行MsGCHI-FLAG融合基因和MsADCS-HA融合基因的扩增;以pCAMBIA1300-pGmUbi质粒为模板,P7、P8为引物,进行启动子Gmubi的扩增,1.0%琼脂糖凝胶电泳检测PCR产物,并纯化回收目的条带。
1.1 pCAMBIA1302-Bar-MsGCHI-FLAG载体的构建
用T4连接酶将MsGCHI-FLAG融合基因的回收产物连接到Spe I/Bst EII双酶切后的pCAMBIA1302-Bar载体,构建流程如图1所示,转化大肠杆菌,进行菌液PCR并测序。
1.2 pBIB-BASTA-Gmubi-MsADCS-HA载体的构建
按照In-Fusion HD Cloning Kit说明将Gmubi启动子回收纯化产物连接到经HindIII与Kpn I双酶切的pBIB-BASTA-GWR-YFP载体,构建流程如图2所示。转化大肠杆菌,通过菌液PCR检测阳性克隆后测序。
对MsADCS-HA融合基因PCR扩增所得目的条带进行纯化回收,分别在上下游引物的5′端加入一段与pBIB-BASTA-Gmubi载体末端15bp大小的序列,引物设计如下:

用MsADCS-HA融合基因回收产物为模板,用Phusion酶扩增目的片段,纯化回收,用Kpn I与Sst I双酶切pBIB-BASTA-Gmubi载体;按照In-Fusion HD Cloning Kit将该片段与pBIB-BASTA-Gmubi载体进行连接,构建流程如图3所示。转化感受态Ecoli DH5α,菌液PCR检测,阳性克隆测序。
1.3 pCAMBIA1302-Bar-MsGCHI-FLAG-Gmubi-MsADCS-HA载体的构建
用Hind III与EcoR I双酶切pCAMBIA1302-Bar-MsGCHI-FLAG载体和pBIB-BASTA-Gmubi-MsADCS-HA,设计包括Gmubi启动子、MsADCS-HA融合基因、pAnos终止子在内序列的引物,并分别在上下游引物的5′端加入一段与pCAMBIA1302-Bar-MsGCHI-FLAG载体末端互补的15bp扩展序列,引物设计如下:

纯化回收pCAMBIA1302-Bar-MsGCHI-FLAG载体大片段和pBIB-BASTA-Gmubi-MsADCS-HA小片段,依据In-Fusion HD Cloning Kit将Nos-Gmubi-MsADCS-HA与pCAMBIA1302-Bar-MsGCHI-FLAG载体连接,构建流程如图4所示。转化感受态Ecoli DH5α,挑取阳性克隆,菌液PCR检测后送华大测序。
2.农杆菌介导的遗传转化
2.1外植体的准备
培育百脉根无菌苗子叶(带叶柄)作为研究所用的外植体。
2.2菌液的制备
取10μL农杆菌菌液加入的LB液体培养基中(含有50mg/L Kan、25mg/L Rif),置于28℃恒温摇床中,200rpm培养16h使溶液呈明显的浑浊,并检测OD600(0.6-0.8);倒入离心管离心(4℃,4000rpm,10min);结束后倒掉上清液,向离心管中加入适量LB培养基重悬浮菌体,并用液体LB培养基将菌液OD600调节至0.5-0.6,以备侵染之用。
2.3遗传转化
(1)预培养:在超净工作台上,将培养7d的百脉根无菌苗子叶切两半置于分化培养基(B5+0.5mg·L-16-BA+5mg·L-1Basta)上,黑暗条件下预培养3d(24±2℃);
(2)菌液的制备:同“2.2”。
(3)侵染:将经过预培养的子叶放入装有重悬液(OD600=0.5)的三角瓶中,用真空泵抽真空(压力)处理10min,超声波处理1min,再抽真空10min;
(4)共培养:将外植体从菌液中取出,吸干外植体表面多余菌液,然后置于分化培养基上,在24±2℃,黑暗条件下共培养3d;
(5)抗性体细胞胚的筛选:在对抗性胚体进行筛选时,采用前期选择的方法,将共培养后的外植体转入5mg·L-1Basta和400mg·L-1头孢(Cef)的分化培养基上,在光照强度约为200μmol·m-2·s-1、24±2℃,16h·d-1条件下进行光培养,并进行筛选培养。每2周继代1次,直到大量成熟抗性芽点出现为止;
(6)生根培养:将成熟抗性芽点移到含5mg·L-1Basta和400mg·L-1Cef的生根培养基上生根成苗。
2.4转基因百脉根PCR检测
2.4.1百脉根基因组DNA的提取
具体步骤如下:
(1)取组织培养获得的转基因百脉根2片三出复叶于1.5mL无菌离心管中,并将离心管迅速放入液氮中冷冻;
(2)用研磨棒将叶片快速充分研磨成粉末状;
(3)加入350μL SDS提取缓冲液,振荡混匀后离心(10000rpm,10min),取250μL上清液移至新的离心管中;
(4)加入250μL的异丙醇混匀,室温放置10min(时间越长,DNA越多)后离心(10000rpm,10min),弃上清,吸出残液,留取沉淀;
(5)加入250μL 75%的乙醇,将沉淀轻轻弹起;
(6)室温12000rpm离心5min,弃上清,12000rpm离心40s,吸出残液;
(7)放置通风橱中使乙醇彻底挥发(10-20min),向沉淀物中加入35μL ddH2O溶解10min,测定浓度并保存。
2.4.2 PCR扩增
根据MsGCHI、MsADCS和Bar基因的核酸序列,利用DNAMAN 6.0软件和NCBI数据库中的Primer-Blast工具设计三对特异性引物(表2),为避免植物自身同源基因的干扰,在该基因的启动子上设计上游引物、基因上选取片段设计下游引物,所有引物均由陕西杨凌奥科公司合成。

PCR扩增时,以H2O和野生型百脉根DNA为模板的反应为阴性对照,以质粒DNA和空载体转基因百脉根DNA为模板的反应为阳性对照。体系如下:
PCR反应结束后,用1%的琼脂凝胶电泳检测是否扩增出目的条带。
3.转基因百脉根RT-PCR检测
3.1百脉根总RNA的提取及反转录
选取阳性的转基因百脉根株系,参考TaKaRa Mini BEST Plant RNA ExtractionKit试剂盒(TaKaRa,北京)的具体说明书,提取转基因百脉根株系和野生型植株的总RNA。用NANO-DROP1000核酸检测仪(伯乐,美国)检测RNA质量,并将总RNA样品保存在-80℃冰箱备用。
根据PrimeScriptTM II 1st Strand cDNA Synthesis Kit和PrimeScriptTM RTMaster Mix(Perfect Real Time)试剂盒(TaKaRa,北京)操作说明书反转录获得各株系cDNA。
3.2 RT-PCR扩增
根据MsGCHI和MsADCS基因的核酸序列,利用DNAMAN 6.0软件以及NCBI数据库中的Primer-Blast工具设计两对特异性引物(表3),并以百脉根Actin基因作为内参基因。
进行百脉根Actin基因RT-PCR扩增时,以野生型百脉根和转化空载体的转基因百脉根cDNA为模板作为阴性对照。根据百脉根Actin基因扩增的结果,对各系列的cDNA量进行调整,直到使各系列Actin基因扩增得到的产物量相对一致。然后根据各系列Actin基因扩增时加入的cDNA量进行MsGCHI和MsADCS基因的RT-PCR扩增,产物检测方法同上。

4.转基因百脉根qRT-PCR检测
4.1 qRT-PCR引物设计与扩增
使用DNAMAN 6.0和NCBI数据库中的Primer-Blast在线工具,分别设计MsGCHI和MsADCS的特异性引物(表4),以百脉根Actin基因作为内参。
以各样品稀释10倍的cDNA为模板,每个样品3个重复,进行qRT-PCR扩增,扩增体系为20μL:
轻轻混匀,盖膜,12000rpm离心3min,然后于ABI 7500荧光定量PCR仪(ABI,美国)中进行扩增,PCR扩增条件:95℃5s,60℃10s,72℃20s,40个循环。采用2-△△Ct法计算两个基因的相对表达量。

5.结果与分析
5.1 MsGCHI-MsADCS双价植物表达载体的构建
采用In-Fusion无缝克隆技术,将MsGCHI-FLAG融合基因,与携带有Bar基因的pCAMBIA1302线性载体相连接。菌液PCR检测,如图5所示,在1350bp左右出现一条清晰的条带,这与MsGCHI-FLAG融合基因1395bp的长度相符合。通过将测序结果与MsGCHI-FLAG融合基因序列比对。进一步证明MsGCHI-FLAG融合基因已构入pCAMBIA1302载体。
同样,采用In-Fusion方法将Gmubi启动子、MsADCS-HA融合基因与携带有Bar标记基因及MsGCHI-FLAG融合基因的pCAMBIA1302载体连接,菌液PCR检测及测序比对结果均正确,证明Gmubi启动子与MsADCS-HA融合基因已成功连接到pCAMBIA1302载体上。
5.2菌液PCR鉴定
将重组质粒转化农杆菌,并做菌液PCR进行检测,凝胶电泳结果如图6所示。可以看出菌液中可以检测出重组质粒中Bar、MsGCHI、MsADCS三个基因和Gmubi启动子,并且条带明亮,故可用作为侵染百脉根的工程菌。
5.3 pCAMBIA1302-Bar-MsGCHI-FLAG-Gmubi-MsADCS-HA对百脉根的遗传转化
实验所用的受体材料为百脉根无菌苗的子叶,将带柄的子叶在分化培养基(B5+0.5mg·L-16-BA+5mg·L-1Basta)上预培养3d后,经抽真空10min+超声处理1min+抽真空10min的侵染方式后,置于共培养基上(B5+0.5mg·L-16-BA)共培养3d后,可观察到子叶的边缘出现菌圈,如图7A所示。
随后将共培养的百脉根幼苗子叶,置于选择培养基(B5+0.5mg·L-16-BA+5mg·L- 1Basta+500mg·L-1Cef)上选择培养,如图7B/C所示。从图中可以看出大部分百脉根幼苗子叶在5mg·L-1Basta的作用下开始变成褐色,但是有一小部分子叶边缘开始分化,长出绿色的芽点。
将在选择培养基上长势良好的幼苗,置于分化培养基(B5+0.5mg·L-16-BA)上培养30d后发现百脉根幼苗子叶边缘开始有绿色芽点长出,如图7D/E所示。继续培养两周,大部分不定胚趋向成熟,如图7F所示。之后将百脉根幼苗子叶边缘绿色芽点切下后放置生根培养基(1/2MS+0.05mg/L NAA+5mg·L-1Basta)上。生长状况如图7G所示,炼苗后,移于装满蛭石的花盆中继续培养,如图7H所示。
5.4转基因百脉根PCR鉴定
以SDS法提取的抗性百脉根株系和野生型百脉根植株的叶片总DNA为模板,用目的基因MsADCS、MsGCHI和标记基因Bar的特异性引物进行PCR检测,电泳结果表明转基因百脉根株系中有13个系列目的基因MsADCS、MsGCHI和标记基因Bar的扩增片段大小与阳性质粒对照PCR的扩增片段大小一致(图8),分别为392bp、519bp和481bp。
5.5转基因百脉根半定量RT-PCR分析
为进一步确定MsGCHI和MsADCS在转基因百脉根株系中的表达水平,以各株系的cDNA为模板,分别用MsGCHI和MsADCS的特异性引物对其进行RT-PCR分析。结果如图9所示,不同株系间MsGCHI和MsADCS的表达丰度存在一定差异;综合来看,2、3、5、9、10、11、12和13号株系中2个目的基因的表达丰度相对较高。
5.6转基因百脉根qRT-PCR分析
为进一步挑选目的基因MsGCHI和MsADCS表达丰度相对一致的转基因株系,以RT-PCR检测出表达水平相对较高的2、3、5、9、10、11、12和13号株系的cDNA为模板,对其目的基因的表达进行qRT-PCR分析。通过qRT-PCR分析发现,发现2、3、5、10、11号株系中目的基因MsGCHI和MsADCS表达相对一致,且表达水平较高,如图10所示。因此,我们选定上述5个株系进行测定转基因百脉根叶酸含量及其主要衍生物含量。
实施例二、基因MsGCHI和MsADCS提高植物叶酸含量的作用
将实施例一筛选出的5个超表达转基因百脉根株系分别命名为GA-2、GA-3、GA-5、GA-10和GA-11,移栽至装有无菌蛭石的花盆中,用1/2Hoagland营养液浇灌培养。并利用扦插法对转基因株系GA-3、GA-11和野生型百脉根植株进行无性扩繁,每隔2天浇水一次,待扦插苗生根后移栽至装有无菌蛭石的花盆中,用1/2Hoagland营养液浇灌培养,植物培养室中温度为24±2℃、光照为16h·d-1、相对湿度为65%左右。
1.叶酸及其组分含量测定
1.1标准品配制
用分析天平称取0.01g的标准品,以100mL Na2CO3溶液(0.1mol/L,以1mol/L盐酸调节pH=7)充分溶解(其中:5-甲基四氢叶酸以甲醇溶解),再用起始流动相(5%乙腈+0.1%甲酸+94.9%水溶液)依次进行梯度稀释,上机检测,最终根据实验样品的检测数据,选择合适的浓度区间绘制“浓度-峰面积”标准曲线。
1.2样品叶酸及其组分提取及测定
具体步骤如下:
(1)分别称取0.5g茎和叶样品,加入液氮研磨成粉末,放于10ml离心管中,加入2.5mL提取液(0.5%氨水溶液,按1%的比例加入抗坏血酸)。同时设置空白样品及回收率检测样品。
(2)先超声30min,再低速涡旋振荡60min、离心混匀(4℃,12000rpm,20min),4℃浸提12h,再次离心后吸取上清。
(3)滤渣中加入2.5mL甲醇,冰上超声30min,混匀离心(4℃,12000rpm,20min),4℃浸提1h,再次离心后吸取上清,合并浸提液。
(4)将收集到的洗脱液干燥浓缩成粉末。
(5)每个样品用0.3mL起始流动相(5%乙腈+0.1%甲酸+94.9%水溶液)复溶,涡旋震荡混匀。再经0.22μm的尼龙膜过滤后上机检测。
(6)采用超高效液相色谱仪ACQUITYUPLCH-Class系统进行分析,进样量为1.0μL,流动相A(0.1%甲酸-水溶液);流动相B(0.1%甲酸-乙腈溶液)。
(7)采用Xevo TQ-S串联四级杆质谱检测系统,离子源为电喷雾源(ESI),脱溶剂温度(450℃),脱溶剂流速(1000L/hr),锥孔器流速(30L/hr),毛细管电压(+3.5KV)。
(8)样本含量(ng/g)=检测浓度(ng/ml)×0.3(mL)÷0.5(g)
2.转基因百脉根生长性状的相关指标测定
用直尺直接测量植株的株高和根长,用排水法测量其根体积。
3.数据分析
用MassLynx V4.1软件进行操作和采集叶酸及其组分的数据,利用SPSS 26.0(IBM,美国)数据分析软件对所有数据进行单因素方差分析(ANOVA)和独立样本T检验分析,数据以平均值±标准误(Means±SE)表示,用SigmaPlot 12.5绘图。
4.结果分析
4.1温室条件下转基因百脉根株系中叶酸及前体含量分析
采用高效液相色谱-串联质谱技术(HPLC-MS/MS)对野生型和选定的5个转基因株系的叶酸及前体含量进行分析。结果表明,共表达MsGCHI和MsADCS基因对百脉根株系体内叶酸和前体含量产生了显著影响,转基因株系中叶酸和蝶呤含量均显著高于野生型,除转基因株系GA-10以外,其余转基因株系中p-ABA含量均显著高于野生型(图11)。
野生型植株叶中叶酸含量为1.08μg/100g FW,蝶呤含量为0.31μg/100g FW,p-ABA含量为0.03μg/100g FW。在5株转基因百脉根株系中,叶中叶酸、蝶呤和p-ABA含量与野生型相比平均提高了2.5倍、2.7倍和0.6倍;茎中叶酸、蝶呤和p-ABA含量与野生型相比平均提高了2.0倍、3.3倍和2.1倍;其中GA-3株系叶和茎中叶酸含量分别达到4.68μg/100g FW和2.99μg/100g FW,与野生型植株相比提高幅度最大,分别提高了3.3倍和2.9倍;与野生型植株相比,GA-5株系叶中蝶呤和p-ABA含量提高幅度最大,分别提高了3.7倍和1.6倍,GA-3株系茎中蝶呤含量提高幅度最大,提高了6.1倍,GA-10株系中p-ABA含量提高幅度最大,提高了3.7倍。
进一步分析野生型和转基因百脉根茎和叶中叶酸、蝶呤和p-ABA含量发现,无论是野生型植株还是转基因株系,其叶中叶酸含量显著高于茎,除GA-5株系外,各株系茎中蝶呤和p-ABA含量显著高于叶(如图12所示)。
4.2温室条件下叶酸生物强化对百脉根生长影响的评价
为了排除代谢工程对植株发育可能产生的不利影响,并进一步明确叶酸生物强化对植物的影响,我们分析了野生型和2个转基因株系的株高、1级分枝数、根长和根体积。如图13所示,培养4周龄后,野生型和2个转基因株系长势良好,转基因株系GA-3长势明显优于野生型和转基因株系GA-11。
进一步比较发现,转基因株系GA-3株高显著高于野生型植株和转基因株系GA-11,与野生型相比提高了29.2%;转基因株系的1级分枝数多于野生型,其中转基因株系GA-11比野生型增多了31.4%;转基因株系GA-3根体积显著大于野生型植株和转基因株系GA-11,GA-11根长显著长于野生型植株和GA-3,与野生型相比增长了12.9%(如图14所示);
综上所述,本发明提供了基因MsGCHI和MsADCS在促进植物生长中的应用,在温室条件下,转基因株系叶中叶酸及前体含量均显著高于野生型,且转基因株系叶中叶酸含量显著高于茎,茎中前体含量普遍高于叶。说明MsGCHI和MsADCS的共表达能够显著提高百脉根中叶酸及前体含量,并且百脉根中前体的转运也是影响叶酸合成的重要因素之一。同时,转基因百脉根株系的生长速度明显优于野生型。说明叶酸生物强化对百脉根生长没有不利影响,且叶酸生物强化合成的内源叶酸还能促进转基因百脉根株系的生长。
