本发明涉及一种新生婴儿免疫配方奶粉及制备方法,尤其是全面提升新生婴儿的免疫系统,属乳品加工技术领域。
背景技术
0-6个月的新生婴儿最理想的食品是乳汁,但由于各种原因不能哺乳时或哺乳量不足时,高质量的免疫配方奶粉是婴儿所首选的。
发明内容
本发明的目的在于克服现有新生婴儿配方奶粉的不足,而提供一种新生婴儿免疫配方奶粉,从而增强新生婴儿的免疫力。本发明根据母乳的营养成份,针对0-6个月新生婴儿配制,模拟初乳中主要的几大免疫物质,即:核苷酸等,全面提升宝宝的免疫系统,从而使宝宝少生病或不生病,从而促进宝宝的健康发育。

本发明对照现有技术的有益效果:
(1)本发明除添加了新生婴儿成长发育所必须的多种维生素和微量元素、矿物质元素,使其营养成份更趋合理,更有效促进0-6个月婴儿的全面生长发育,通过添加核苷酸的含量,使其提高0-6个月婴儿的免疫力,从而促进婴儿的生长发育。核苷酸是核糖核酸及脱氧核糖核酸的基本组成单位,核苷酸的来源有两种途径,一种是由机体的细胞合成,一种是从食物中获得,核苷酸的生理机能是能维持免疫系统的正常功能,能提高0-6个月婴儿对细菌、真菌感染的抵抗力,增加抗体产生,增强细胞免疫能力,刺激淋巴细胞增生等功能。此外,核苷酸能维持胃肠道正常功能,抑制有害细菌的生长,减少婴儿腹泻的发生。
(2)本发明通过添加酪蛋白磷酸肽(CPP)的含量,能有效促进婴儿骨骼和牙齿对钙的吸收和利用,它除了促进钙的吸收外,还促进铁、锌等二价矿物质元素的吸收,从而提高了铁、锌的吸收利用率,从而促进新生婴儿的健康发育。CPP是以牛乳酪蛋白为原料,通过生物技术制得的具有生物活性的多肽,它还具有免疫功能,能帮助婴儿提高免疫力,防止疾病发生。
(3)本发明通过添加牛磺酸的含量,它能促进新生婴儿脑组织和智力发育,牛磺酸在脑内的含量丰富,分布广泛,能明显促进神经系统的生长发育,从而促进婴儿智力生长发育,而牛磺酸与婴儿的中枢神经及视网膜的发育有密切关系,能促进垂体激素分泌活化胰腺功能,从而改善机体内分泌的状态,提高神经传导和视觉机能,具有促进有机体免疫力的增强和抗疲劳的作用。
(4)本发明通过添加复合益生元能够提高0-6个月婴儿机体细胞免疫力和体液免疫两方面的功能,复合益生元是指能够选择的刺激肠内一种或几种有益菌生长繁殖,而且不被宿主消化的物质,研究证明,复合益生元能够帮助提高体内免疫球蛋白IGG的水平,帮助0-6个月婴儿更好的建立自身免疫系统,增强抗病能力。
(5)亚油酸、亚麻酸是公认的必须脂肪酸,具有改善记忆、改善睡眠、提高智力、提高免疫力等保健功能,配方中的二十二碳六烯酸(DHA)也称脑黄金;二十碳四烯酸(ARA)又称花生四烯酸,它们是大脑营养不可缺少的不饱和脂肪酸,能提高智力等作用,是婴儿大脑发育成长的重要物质之一。而亚油酸、亚麻酸也是合成DHA、ARA的前提,0-6个月婴儿大脑发育系统不完善,单靠亚油酸、亚麻酸合成DHA、ARA远远不能满足,因此需另外补充DHA、ARA才能使婴儿大脑发育和视神经系统发育达到最佳状态。
本发明专为0-6个月的婴儿研制,具有原料配比新颖,制品质量稳,各项指标符合国家婴幼儿食品标准。结合0-6个月婴儿生长发育的营养要求,合理强化钙铁锌复合维生素,并特别添加了亚油酸、亚麻酸、DHA、ARA、左旋肉碱、肌醇、胆碱、核苷酸、复合益生元、牛磺酸等,经先进工艺而成。全面促进0-6个月婴儿的生长发育,使婴儿迅速建立更好的免疫系统,全面提升免疫配方奶粉的质量。
具体实施方式
本发明的生产方法:收奶——贮存——配料(在鲜奶中加入脱盐乳清粉、乳糖、胆碱、复合维生素及复合微量元素、矿物质元素、复合益生元、酪蛋白磷酸肽、核苷酸、亚油酸、亚麻酸、肌醇、左旋肉碱、二十二碳六烯酸、二十碳四烯酸、牛磺酸)——预热(加入无水奶油、大豆色拉油)——均质——杀菌——浓缩——干燥——包装。
免疫配方奶粉的功能性评价:
免疫调节作用:指肌体接触抗原性异物或异已的一种特异性生理反应。具有增强免疫调节作用的保健品,是指那些能增强机体对疾病的抵抗力、抗感染、抗肿瘤以及维持自身生理平衡的食品。在一组实验中,样品对免疫系统各方面的功能的实验具有增强作用,而对其他实验无抑制作用,就能认定该样品具有此方面的免疫调节功能。在细胞免疫功能、体液免疫功能、单核-巨噬细胞功能及NK细胞功能检测中,如果有两个(含两个)功能检测结果呈阳性,即可判定该受试物具有免疫调节功能。
实验材料
样品:A:免疫配方奶粉 B:普通奶粉
此样品有香港许记集团力维康营养食品有限公司提供。
实验动物和喂养方法:
小鼠购自江西省实验大小鼠生产基地,6周龄,体重20±3g,雌雄兼有,随机分两大组。
1组为正常组,Ⅱ组为免疫低下组
此后将每一大组分为三组,一组喂免疫配方奶粉;一组喂普通奶粉;另一组直接喂水。所有实验小鼠均正常喂食。喂养30天,在实验前一天禁止喂食。
实验方法:
参照卫生部卫生监督司颁发《保健食品功能性评价程序和实验方法》
免疫功能实验:
1、对小鼠免疫器官重量的影响
将分组的小鼠解剖,取出脾脏、胸腺称重,并按下列方式计算脾指数和胸腺指数。
2、单核-巨噬细胞功能测定
将每组的实验小鼠的腹腔注射浓度5×106/ml鸡红血球悬液(1ml/只)30分钟后,收集小鼠腹腔液,涂片以1∶1丙酮-甲醛液固定,染色,用油镜计数100个巨噬细胞中吞噬鸡红血球的巨噬细胞数目及吞噬鸡红血球的总数,并按下列公式计算吞噬百分比和吞噬指数。
3、细胞免疫功能的测定——小鼠脾淋巴细胞转化试验
将小鼠处死,制备脾细胞悬液,用淋巴细胞分离液获取淋巴细胞,用HanKS洗涤三次,用台盼蓝染色液柒色计数,活细胞数>95%,调整细胞浓度为2×106/ml。将细胞液分两组加入24孔培养板中,每孔1ml;一组加ConA50ul,另一组作对照,置5%CO2,37℃饱和湿度培养箱中。培养结束4h,每孔吸去上清液0.8ml,加入不含小牛血清的RPMI-1640培养液,同时加入MTT50ul/孔,培养至72h。培养结束后,每孔加入1ml的酸性异丙醇,吹打混匀,用721分光光度计,570nm波长测定OD值。
4、数据处理及结果判断:
将数据进行处理,并用t检验进行显著性检验。P<0.05表示差异显著,P>0.05差异不显著。
实验结果:
小鼠免疫器官重量的影响:
免疫配方奶粉对小鼠脾脏、胸腺的影响见表1:
注:P1为免疫配方奶粉和普通奶粉比较,P2为免疫配方奶粉和水对照比较。由表1可见,配方奶粉对脾脏、胸腺的作用有促进增长的趋势,喂食配方奶粉后正常实验组的脾脏指数及胸腺指数与喂食普通奶粉实验组以及喂食饮用水的实验组相比较略有提高;但除喂食配方奶粉组与喂饮用水组的胸腺指数在统计学意义上有明显的差异外,配方奶粉组和普通奶粉组的脾指数和胸胸指数在统计学意义上没有显著差异。
而免疫低下组小鼠经喂食配方奶粉后,其脾指数和胸腺指数也有增长趋势并恢复至正常水平。与同组的喂食普通奶粉组相比较,其脾指数和胸腺指数均无显著差异。
但实验发现免疫低下组小鼠喂食配方奶粉一组的存活率明显比喂食普通奶粉组及饮用水的实验组的存活率高。喂食免疫配方奶粉的小鼠的死亡率10%,而喂食普通奶粉的死亡率50%,饮用水的死亡率40%。可见,喂食配方奶粉后,使小鼠恢复了免疫活性,提高了机体的免疫力,延长其生存时间。
小鼠腹腔巨噬细胞吞噬功能实验
配方奶粉对小鼠腹腔巨噬细胞吞噬鸡红细胞的数量及其吞噬活性的影响见表2:
注:P1为免疫配方奶粉和普通奶粉比较,P2为免疫配方奶粉和水对照比较。巨噬细胞是机体免疫的关键细胞,测定巨细胞的吞噬功能在一定程度上可了解机体的免疫情况。表2表明:正常小鼠在喂食配方奶粉后,其巨噬细胞吞噬鸡红细胞的能力和对照组相比有显著性提高,每100个巨噬细胞能吞噬鸡红细胞的细胞从35.6个提高至51.40个,提高了44%,每100个巨噬细胞内吞噬鸡红细胞数量由84个提高至93个,提高了10.7%,而喂食普通奶粉的实验组则在激活单核巨噬细胞方面无变化,免疫低下组中的小鼠在喂食配方奶粉后,其吞噬指数和吞噬百分比与同一实验中的喂食普通奶粉与饮用水的两组相比均有显著提高,并基本上恢复正常水平。表明配方奶粉可使免疫低下的小鼠单核-巨噬细胞的吞噬功能恢复至正常水平。本实验表明:配方奶粉对小鼠的非特异性免疫功能有显著的增强作用。
淋巴细胞转化实验

注:P1为配方奶粉和普通奶粉比较;P2为配方奶粉和水比较。脾脏能产生大量的淋巴细胞,淋巴细胞是机体内最为复杂的一个细胞系。淋巴细胞以B细胞为主,约占总数60%左右。B细胞通过产生抗体发挥其体液免疫功能。表3可知,正常小鼠在喂食配方奶粉后其淋巴细胞的转化率与普通奶粉相比较,都有显著提高;而普通奶粉在这一性能上与饮用水的实验组没有明显差异,表明配方奶粉有一定的促进B淋巴细胞转化作用。免疫低下组的小鼠经喂食配方奶粉后,其淋巴细胞转换率也恢复至正常水平,此实验表明配方奶粉在细胞免疫方面具有调节效应。

Claims (6)
2.按照权利要求1所述的一种新生婴儿免疫配方奶粉,其特征在于所述的复合维生素包括:(克/吨)
维生素A 4.2g 维生素D 0.07g 维生素E 60g
维生素K1 0.3g 维生素B 15g 维生素B2 7g
维生素B6 3.5g 维生素B12 0.0106g 维生素C 650g。
3.按照权利要求1所述的一种新生婴儿免疫配方奶粉,其特征在于所述复合微量元素种类及含量:(克/吨)
生物素 0.105g 叶酸 0.65g 烟酸 26g 泛酸 28g。
4.按照权利要求1所述的一种新生婴儿免疫配方奶粉,其特征在于所述复合益生元种类及含量:(克/吨)
低聚果糖(FOS) 3000g 低聚半乳糖(GOS) 5000g。
5.按照权利要求1所述的一种新生婴儿免疫配方奶粉,其特征在于所述矿物质元素种类及含量:(克/吨)
钠 1500g 钾 4500g 铜 2.4g 镁 400g
铁 32g 锌 36g 锰 0.8g 钙 3650g
磷 2400g 碘 0.65g 氯 3420g 硒 0.128g。
6.按照权利要求1所述的一种新生婴儿免疫配方奶粉,其特征在于其生产方法:收奶——贮存——配料(在鲜奶中加入脱盐乳清粉、乳糖、胆碱、复合维生素及复合微量元素、矿物质元素、复合益生元、酪蛋白磷酸肽、核苷酸、亚油酸、肌醇、左旋肉碱、二十二碳六烯酸、二十碳四烯酸、亚麻酸、牛磺酸)——预热(加入无水奶油、大豆色拉油)——均质——杀菌——浓缩——干燥——包装。
