本发明公开一种可保持五谷虫分泌排泄物活性的收集液,是Tween80的磷酸盐缓冲液或牛血清白蛋白的磷酸盐缓冲液,Tween80与磷酸盐缓冲液的质量体积比为10克/升,牛血清白蛋白与磷酸盐缓冲液的质量体积比为1克/升。可保持所收集的五谷虫分泌排泄物生物活性成分的稳定性,保证了收集物的质量,即使在温度出现较大变化(反复冻融达10余次)的情况下仍能保持其稳定性,可长期保存,利于五谷虫分泌排泄物的临床应用及治疗功效的进一步研究。

技术领域
本发明涉及一种收集五谷虫分泌排泄物的收集液,尤其是一种可保持分泌排泄物中生物活性成分的稳定性,利于保存的五谷虫分泌排泄物的收集液。
背景技术
由于五谷虫(蛆虫)能够吞噬/啃食腐肉促进创伤愈合,为此五谷虫活体已长期用于治疗因创伤引起的坏死和感染严重的伤口。目前,人们已发现五谷虫口中分泌物和排泄物中含有大量生物活性成分,如蛋白质和多肽类化合物等,这些物质在抗菌、消炎和促细胞增殖等方面均发挥重要作用。现有收集五谷虫分泌排泄物的方法是:将清洗过的五谷虫放入生理盐水(20条/10ml)中约5~10分钟,使其吐出和排泄体内物质。用生理盐水作为收集液难以保持五谷虫分泌排泄物中的蛋白质和其它生物活性物质的稳定性,因而影响了所收集的五谷虫分泌排泄物整体质量且不利于长期保存,制约了五谷虫分泌排泄物的临床应用及治疗功效的进一步研究。
发明内容
本发明是为了解决现有技术所存在的上述技术问题,提供一种可保持分泌排泄物中生物活性成分的稳定性,利于保存的五谷虫分泌排泄物的收集液。
本发明的技术解决方案是:一种可保持五谷虫分泌排泄物活性的收集液,是Tween 80的磷酸盐缓冲液或牛血清白蛋白的磷酸盐缓冲液,Tween 80与磷酸盐缓冲液的质量体积比为10克/升,牛血清白蛋白与磷酸盐缓冲液的质量体积比为1克/升。
本发明与现有技术相比,可保持所收集的五谷虫分泌排泄物生物活性成分的稳定性,保证了收集物的质量,即使在温度出现较大变化(反复冻融达10余次)的情况下仍能保持其稳定性,可长期保存,利于五谷虫分泌排泄物的临床应用及治疗功效的进一步研究。

具体实施方式
实施例1:
将10克Tween 80溶于1升磷酸盐缓冲液,即制得收集液。
所述Tween 80和磷酸盐缓冲液(PBS)均为现有技术。
收集过程同现有技术,即将清洗过的五谷虫放入本实施例1收集液中(20条/10ml)中约5~10分钟,取出五谷虫,则收集液中含有五谷虫吐出和排泄的分泌排泄物。
实施例2:
将1克牛血清白蛋白(BSA)粉末溶于1升磷酸盐缓冲液,即制得收集液。
所述牛血清白蛋白(BSA)粉末和磷酸盐缓冲液(PBS)均为现有技术。
收集过程同现有技术,即将清洗过的五谷虫放入本实施例2收集液中(20条/10ml)中约5~10分钟,取出五谷虫,则收集液中含有五谷虫吐出和排泄的分泌排泄物。
实验:
取本发明实施例1、实施例2及现有技术(生理盐水)为收集液所得到的含有五谷虫分泌排泄物的收集物,在同等条件下反复冻融10次后进行凝胶电泳,电泳图如图1所示:1为本发明实施例1;2为本发明实施例2;3为现有技术。结果表明:实施例1、实施例2中的蛋白成分即是在反复冻融达10次的情况下也仍能保持稳定, 而现有技术中的蛋白液则大部分降解。因此,采用本发明制备的五谷虫分泌排泄物十分适合各种条件下的使用、长期保存和长途运输。

1.一种可保持五谷虫分泌排泄物活性的收集液,其特征在于是Tween 80的磷酸盐缓冲液或牛血清白蛋白的磷酸盐缓冲液,Tween 80与磷酸盐缓冲液的质量体积比为10克/升,牛血清白蛋白与磷酸盐缓冲液的质量体积比为1克/升。
