本发明涉及宝乐果在经皮透皮吸收中的用途,属于护肤的化妆品技术领域。
背景技术
宝乐果(Borojoa sorbilis Cuter.)是茜草科林果属植Borojoa patinoiCuatrec.和Borojoa sorbilis Cuatrec.的成熟果实,产于拉丁美洲厄瓜多尔气候的热带雨林,呈棕褐色。当地民间称宝乐果具有消除疲劳,稳定血糖水平,维持正常血压,增强免疫,抗炎及抗氧化等功效。宝乐果水果浓缩粉富含的多糖、蛋白质、钾、钠、钙、磷、铁、镁及水溶性B族维生素等物质,有增强细胞活力的作用;宝乐果还含有人体不能合成的亚油酸及亚麻酸,可作为维持细胞膜流动的重要物质,有利于细胞膜的酶促反应,同时对降低脂质过氧化有着积极的作用。宝乐果水果浓缩粉富含的倍半萜类(环烯醚萜苷)、多酚类和VE可能是其抗氧化作用产生的重要原因之一。宝乐果水果浓缩粉中、高剂量组能显著提高小鼠血清和肝脏中SOD活力、降低MDA含量;宝乐果水果浓缩粉高剂量组能显著提高衰老小鼠血清和肝脏中GSH-Px活力。目前,尚未见文献对宝乐果的透皮情况作深入的研究。
皮肤是人类具有的天然抵抗外来物质入侵的人体屏障,具有对皮肤有特异保护性能、良好的透皮效果、适当的透皮剂量和作用是功效护肤型化妆品的基础条件。在经皮给药系统的开发中,药物透皮速率的研究是关键,是药物、经皮透过吸收促进剂和高分子材料等处方筛选的依据。目前文献中报道的经皮给药制剂体外透皮扩散试验采用的主要方法是扩散池法。该法是将离体皮肤去除皮下脂肪后,剪成合适大小夹在扩散池中,离体皮肤将扩散池隔离成供给室和接受室,角质层朝向供给室,真皮层朝向接受室,并使接受池中的接受液恰好与真皮层接触。药物应用于供给室紧贴角质层,然后在设定的时间点取一定体积的接受液来分析不同时间点药物在接受液中的浓度,从而分析药物的经皮吸收情况。目前应用于体外透皮扩散试验的扩散池主要为改良型franz扩散池(也称Keshary-Chien扩散池),由于K-C池使用简便并具有良好的应用性,现已成为经皮给药制剂体外透皮扩散研究中最常用的仪器。人体皮肤是体外透皮扩散试验的最佳皮肤,但较难获得,故目前在国内文献报道中,体外透皮扩散研究所用的皮肤大部分来自健康大鼠、小鼠和家兔等。在接收液的选择上,人体血液中大部分是水,只有小部分电解质和蛋白质等物质。理想的接受液应在满足漏槽条件的基础上尽可能接近人体皮肤微环境。目前国内、外常用的接受液有生理盐水(NS)、林格液、等渗磷酸盐缓冲液(IPBS)。
体外透皮实验是考察药物透过皮肤的重要方法,是保证疗效和质量稳定可控的重要手段,本申请发明人选择大鼠皮肤作为介质,生理盐水为接收液,以改良型Frans扩散池方法进行含宝乐果护肤品的透皮实验,以宝乐果中特异性活性物质宝乐果8,9-环烯醚萜苷作为参比标志物,用UPLC-MS/MS方法检测活性物经皮吸收的透皮释放量,来研究宝乐果护肤品的透皮情况。

本发明的目的在于克服上述现有护肤品技术不能透过皮肤发挥功效作用的不足之处而提供宝乐果在透皮吸收中的用途。

作为本发明所述用途的优选实施方式,所述宝乐果的透皮效果同护肤品中宝乐果添加的量呈剂量效应关系。即:护肤品中添加宝乐果的量多,透过皮下粘液中的宝乐果-8,9-环烯醚萜苷的量就多;相反,护肤品中添加宝乐果的量少,透过皮下粘液中的宝乐果-8,9-环烯醚萜苷的量就少。宝乐果8,9-环烯醚萜苷能在短时间内渗入皮肤深层发挥药效,且在皮下的作用时间可以持续24h。
与现有技术相比,本发明的有益效果为:本发明的宝乐果具有透皮吸收的效果,其活性成分能透过皮肤角质层,渗入皮肤深层发挥功效作用,能用于制备经透皮吸收的护肤型化妆品。

具体实施方式
为更好地说明本发明的目的、技术方案和优点,下面将结合附图和具体实施例对本发明作进一步说明。需要指出的是,实施例中用到的护肤品的成分含量,本领域技术人员可以根据需要进行常规选择,本申请只是对宝乐果的透皮吸收效果进行研究。
实施例1宝乐果透皮吸收的效果试验
1、试验材料
(1)供试品材料
本实施例以宝乐果活性物质8,9-环烯醚萜苷作为标志物,对不含宝乐果的化妆品基质、宝乐果防晒乳、宝乐果美白精华霜、宝乐果修护睡眠面膜、宝乐果柔肤水的透皮吸收效果进行测试。各供试品相关信息如表1所示,表1中的结果代表各组动物使用相应配方的护肤品后,皮下粘液中所含活性物指标的量。

各供试品的成分组成及制备方法如下:
化妆品基质:
水、甘油、苯基苯并咪唑磺酸、硫酸镁、甘草酸二钾、二氧化钛/氢氧化铝/氧化锌/三乙氧基辛基硅烷、聚二甲基硅氧烷PEG-10/15交联聚合物/聚二甲基硅氧烷,等。
制备方法:准确称量配方中各种原料,用干净并消毒好的器皿盛放;将原料分别加入搅拌锅中,充分搅拌均匀;开启真空,将搅拌锅中物料抽入真空乳化锅中,搅拌均匀混合锅中原料,均质乳化2~3分钟后充分搅拌均匀,恒温搅拌消泡10分钟后降温。对标样合格过100目出料,检测合格后灌装;灌装后再抽检,检测合格入库。
宝乐果防晒乳:
成分组成:水、甘油、苯基苯并咪唑磺酸、精氨酸、硫酸镁、甘草酸二钾、透明质酸钠、二氧化钛/氢氧化铝/三乙氧基辛基硅烷、甲氧基肉桂酸乙基己酯、辛基聚甲基硅氧烷、环五聚二甲基硅氧烷、丁基辛醇水杨酸酯、水杨酸乙基己酯、氧化锌/三乙氧基辛基硅烷、聚二甲基硅氧烷PEG-10/15交联聚合物/聚二甲基硅氧烷、丁二醇二辛酸/二癸酸酯、环五聚二甲基硅氧烷/聚二甲基硅氧烷/乙烯基聚二甲基硅氧烷交联聚合物、环五聚二甲基硅氧烷/二硬脂二甲铵锂蒙脱石/碳酸丙二醇酯、双-乙基己氧苯酚甲氧苯基三嗪、聚甲基丙烯酸甲酯、丙烯酸(酯)类/聚二甲基硅氧烷共聚物/环五聚二甲基硅氧烷、PEG-10聚二甲基硅氧烷、聚甲基硅倍半氧烷、三甲基硅烷氧基硅酸酯、氯氧化铋、HDI/三羟甲基己基内酯交联聚合物/硅石、聚羟基硬脂酸、二甲基甲硅烷基化硅石、宝乐果果汁、生物糖胶-1/岩藻糖/水、水/马齿苋(PORTULACA OLERACEA)提取物/丁二醇、苯氧乙醇/乙基己基甘油、水/木糖醇/脱水木糖醇/木糖醇基葡糖苷、3-o-乙基抗坏血酸。
制备方法:准确称量配方中各种原料,用干净并消毒好的器皿盛放;将原料分别加入搅拌锅中,搅拌升温至80~85℃后,充分搅拌均匀,保温备用;开启真空,将搅拌锅中物料抽入真空乳化锅中,搅拌均匀混合锅中原料,均质乳化2~3分钟后充分搅拌均匀,恒温搅拌消泡10分钟后降温;降温至45℃以下时,加入事前预溶的宝乐果果汁按0.5~5%配置比例(宝乐果汁在加入之前,按照在总体系中的含量先用纯净水溶解成液体,然后再加入到体系中),充分搅拌均匀后出料、静置;对标样合格过100目出料,检测合格后灌装;灌装后再抽检,检测合格入库。
宝乐果美白精华霜:
成分组成:水、双丙甘醇、羟乙基脲、赤藓醇、丙烯酸(酯)类/C10-30烷醇丙烯酸酯交联聚合物、卡波姆、羟苯甲酯、透明质酸钠、泛醇、氢氧化钠、水/马齿苋(PORTULACAOLERACEA)提取物/丁二醇、宝乐果果汁、烟酰胺、α-熊果苷、甘草类黄酮/双丙甘醇、PEG-40氢化蓖麻油、丙二醇、香精、神经酰胺2/聚甘油-10油酸酯/丁二醇/苯氧乙醇、水、苯甲醇/乙基己基甘油、水/丁二醇/苯氧乙醇、CI 40800。
制备方法:准确称量配方中各种原料,用干净并消毒好的器皿盛放;将原料加入搅拌锅中充分搅拌均匀后升温至85~90℃,保温搅拌15~30分钟降温;充分搅拌均匀后出料、静置;降温至45℃时以下,加入事前预溶的宝乐果果汁按0.2~3%配置比例,充分搅拌均匀后出料、静置;检验合格后,灌装,包装,喷码,入库。
宝乐果柔肤水:
成分组成:水、丙二醇、双丙甘醇、赤藓醇、泛醇、宝乐果果汁、异壬酸乙基己酯、丙烯酸(酯)类共聚物钠、氢化聚癸烯、PPG-1十三醇聚醚-6、水解葡糖氨基聚糖、甘油、丙烯酸/MA共聚物钠、丁二醇、辛甘醇、1,6-己二醇、EDTA二钠、丙烯酸(酯)类/C10-30烷醇丙烯酸酯交联聚合物、三乙醇胺、金黄洋甘菊(CHRYSANTHELLUM INDICUM)提取物、透明质酸钠、燕麦(AVENA SATIVA)仁提取物、银耳(TREMELLA FUCIFORMIS)提取物、扭刺仙人掌(OPUNTIASTREPTACANTHA)茎提取物、己二醇、刺云实(CAESALPINIA SPINOSA)胶、山梨酸钾、苯甲醇、乙基己基甘油、羟苯甲酯、苯氧乙醇、香精、CI40800。
制备方法:准确称量配方中各种原料,用干净并消毒好的器皿盛放;将原料加入真空乳化锅中,搅拌升温至80~85℃后,充分搅拌均匀,保温备用;加入事前预溶的宝乐果果汁按0.01~0.5%配置比例,开启真空,均质乳化5~8分钟后充分搅拌均匀,恒温搅拌消泡10分钟后降温;降温至45℃以下时,充分搅拌均匀后出料、静置;对标样合格过100目出料,检测合格后灌装;灌装后再抽检,检测合格入库。
宝乐果修护睡眠面膜:
成分组成:水、甘油、丙二醇、赤藓醇、羟乙基脲、聚丙烯酸酯交联聚合物-6、皱波角叉菜(CHONDRUS CRISPUS)提取物、丁二醇、野蔷薇(ROSA MULTIFLORA)果油、生育酚乙酸酯、丙烯酸羟乙酯/丙烯酰二甲基牛磺酸钠共聚物、三乙醇胺、卡波姆、鲸蜡硬脂基葡糖苷、山梨坦硬脂酸酯、甘油硬脂酸酯、鲸蜡硬脂醇、丙烯酸(酯)类共聚物钠、氢化聚癸烯、PPG-1十三醇聚醚-6、宝乐果)果汁、丙烯酸(酯)类/C10-30烷醇丙烯酸酯交联聚合物、尿囊素、丙烯酸/MA共聚物钠、辛甘醇、1,6-己二醇、小核菌(SCLEROTIUM ROLFSSII)胶、透明质酸钠、燕麦(AVENA SATIVA)仁提取物、银耳(TREMELLA FUCIFORMIS)提取物、扭刺仙人掌(OPUNTIASTREPTACANTHA)茎提取物、己二醇、苯甲醇、乙基己基甘油、羟苯甲酯、苯氧乙醇、香精、CI40800。
制备方法:准确称量配方中各种原料,用干净并消毒好的器皿盛放;将原料加入真空乳化锅中,搅拌升温至80~85℃后,充分搅拌均匀,保温备用;加入事前预溶的宝乐果果汁按0.1~2%配置比例,开启真空,均质乳化5~8分钟后充分搅拌均匀,恒温搅拌消泡10分钟后降温;降温至45℃以下时,充分搅拌均匀后出料、静置;对标样合格过100目出料,检测合格后灌装;灌装后再抽检,检测合格入库。
(2)仪器
超高效液相串联质谱(UPLC-MS/MS,美国Waters,XEVO TQ-S,四元泵);ACQUICTYUPLC HSS T3色谱柱(2.1×100mm,1.8μm);5804R型冷冻离心机(德国Eppendorf公司);分析天平(型号:BT-125D);TP-6智能型透皮扩散仪(天津市精拓仪器科技有限公司)。
(3)药品与试剂
8,9-环烯醚萜苷对照品(自制),芹菜素标准品(中国食品药品检定研究院,纯度≥99%,批号:111901-201603);乙腈、甲醇、甲酸(色谱级,默克股份两合公司,4L);超纯水(默克密理博有限公司,ELIX超纯水系统)。
(4)动物
SPF级雄性SD大鼠15只,体重(250±20)g,由广东省医学实验动物中心提供,合格证号SCXK(粤)2013-0002;均在SPF实验室内饲养与实验操作,湿度(40%~70%),温度为(20℃~24℃)。所有实验操作按照广东省中医院动物伦理委员会的要求进行。
2、试验方法
SD大鼠麻醉后,腹部剃毛,剥离腹部皮肤,剔除皮下脂肪,立即进行透皮试验。接受室加入生理盐水,供给室加入表1所述样品各1g,再在供给室上盖上保鲜膜,体外透皮试验装置如图1所示,将处理好的皮肤置于接受室与供给室之间(透皮面积为3.14cm2,接收室容积为15ml,实验温度保持为37±0.5℃,搅拌速度为300±10r/min)。分别于1、2、4、6、8、10、12、24h从接收液中取样1mL,同时补充相同体积的接收液,得到的样品取出400μL于EP管中,并加入100μL芹菜素内标溶液,涡旋3min,14000rpm离心30min,取上清进样分析,计算活性物:宝乐果8,9-环烯醚萜苷的含量。
3、试验结果
按照公式1计算单位面积累积透过量:
公式1中Cn为第n个取样点药物的校正浓度;
Ci是第i个取样点测得的药物浓度;
Vn和Vi分别是接收池的体积(15mL)和取样体积(1mL);
A是扩散池的扩散面积(A=3.14cm2)。
以Qn对时间t作图,并对曲线中的直线部分线性回归,得出直线斜率即稳态渗透速率Js(μg·cm-2·h-1)。直线截距为滞后时间tlag,渗透系数(P)=Js/Cd(Cd为供试品溶液中药物的质量浓度)。
宝乐果8,9-环烯醚萜苷在样品中的渗透参数如表1所示,渗透曲线如图2所示。

由表1和图2可知,不含宝乐果的化妆品基质样品中未检出宝乐果8,9-环烯醚萜苷。宝乐果防晒乳的稳态渗透速率(6.37±2.35ng.cm-2.h-1)显著高于宝乐果美白精华霜(3.69±1.01ng.cm-2.h-1)、宝乐果修护睡眠面膜(0.56±0.12ng.cm-2.h-1)和宝乐果爽肤水(0.32±0.11ng.cm-2.h-1)。宝乐果的四个供试品均有透皮吸收效果,其中,宝乐果防晒乳透过皮肤角质层的量最多,高于其他宝乐果化妆品。可以看出,宝乐果的透皮效果同护肤品中宝乐果的量呈剂量效应关系。即:护肤品中添加宝乐果的量多,透过皮下粘液中的宝乐果-8,9-环烯醚萜苷的量就多。爽肤水中使用的较少,因此皮下粘液中宝乐果-8,9-环烯醚萜苷的量就少。宝乐果8,9-环烯醚萜苷能在短时间内渗入皮肤深层发挥药效,且,在皮下的作用时间可以持续24h。
实施例2
1、试验材料
(1)供试品材料
本实施例以宝乐果活性物质环烯醚萜苷作为标志物,对使用不含宝乐果的抗敏精华液、阳性抗敏精华液对照、含宝乐果的抗敏精华液、含宝乐果和阳性成分的抗敏精华液的四组动物进行透皮吸收效果测试。各供试品的成分组成及制备方法如下:
不含宝乐果的抗敏精华液:
水、甘油、苯基苯并咪唑磺酸、硫酸镁、甘草酸二钾、二氧化钛/氢氧化铝/氧化锌/三乙氧基辛基硅烷、聚二甲基硅氧烷PEG-10/15交联聚合物/聚二甲基硅氧烷,等。
制备方法:准确称量配方中各种原料,用干净并消毒好的器皿盛放;将原料分别加入搅拌锅中,充分搅拌均匀;开启真空,将搅拌锅中物料抽入真空乳化锅中,搅拌均匀混合锅中原料,均质乳化2~3分钟后充分搅拌均匀,恒温搅拌消泡10分钟后降温。对标样合格过100目出料,检测合格后灌装;灌装后再抽检,检测合格入库。
抗敏精华液阳性对照:
水、甘油、苯基苯并咪唑磺酸、硫酸镁、甘草酸二钾、二氧化钛/氢氧化铝/氧化锌/三乙氧基辛基硅烷、聚二甲基硅氧烷PEG-10/15交联聚合物/聚二甲基硅氧烷,4-氨基吡啶0.02%(阳性对照):等。
制备方法:准确称量配方中各种原料,用干净并消毒好的器皿盛放;将原料分别加入搅拌锅中,充分搅拌均匀;开启真空,将搅拌锅中物料抽入真空乳化锅中,搅拌均匀混合锅中原料,均质乳化2~3分钟后充分搅拌均匀,恒温搅拌消泡10分钟后降温。对标样合格过100目出料,检测合格后灌装;灌装后再抽检,检测合格入库。
含宝乐果的抗敏精华液:
水、甘油、苯基苯并咪唑磺酸、硫酸镁、甘草酸二钾、二氧化钛/氢氧化铝/氧化锌/三乙氧基辛基硅烷、聚二甲基硅氧烷PEG-10/15交联聚合物/聚二甲基硅氧烷,宝乐果粉2%,等。
制备方法:准确称量配方中各种原料,用干净并消毒好的器皿盛放;将原料分别加入搅拌锅中,充分搅拌均匀,待温度下降到40℃时,加入宝乐果粉,搅拌均匀。开启真空,将搅拌锅中物料抽入真空乳化锅中,搅拌均匀混合锅中原料,均质乳化2~3分钟后充分搅拌均匀,恒温搅拌消泡10分钟后降温。对标样合格过100目出料,检测合格后灌装;灌装后再抽检,检测合格入库。
含宝乐果和阳性成分的抗敏精华液:
水、甘油、苯基苯并咪唑磺酸、硫酸镁、甘草酸二钾、二氧化钛/氢氧化铝/氧化锌/三乙氧基辛基硅烷、聚二甲基硅氧烷PEG-10/15交联聚合物/聚二甲基硅氧烷,4-氨基吡啶0.02%(阳性对照)、宝乐果粉2%,等。
制备方法:准确称量配方中各种原料,用干净并消毒好的器皿盛放;将原料分别加入搅拌锅中,充分搅拌均匀,待温度下降到40℃时,加入宝乐果粉,搅拌均匀。开启真空,将搅拌锅中物料抽入真空乳化锅中,搅拌均匀混合锅中原料,均质乳化2~3分钟后充分搅拌均匀,恒温搅拌消泡10分钟后降温。对标样合格过100目出料,检测合格后灌装;灌装后再抽检,检测合格入库。
仪器、试剂材料
1.1.1实验动物SPF级KM小鼠,60只,雌雄各半,由中山大学(大学城)实验动物中心提供,体重19.0±1.7g,合格证号为:SCXK(粤)-2011-0029。
1.1.2主要仪器BSA224S电子分析天平(赛多利斯科学仪器(北京)有限公司),恒温水浴锅(上海精密实验设备有限公司),酶标仪Model680(伯乐生命医学产品有限公司),高速冷冻离心机KDC-140HR(安徽中科中程科学仪器有限公司),FJ200-S数显高速分散机(上海标本模型厂),CP-8000剃毛器深圳科德士电器有限公司,Seriesn170剃毛器德国BRAUN。
1.1.3药品与试剂
1.1.3.1试验样品:
①C13738化妆品基质;②C13737宝乐果;③C13739阳性成分;④C13740宝乐果+阳性成分。
1.1.3.2小鼠阳性致敏物
4-氨基吡啶:Merck公司产品,批号:54914223。使用前用生理盐水配成0.02%溶液0.01mL/g。
1.1.3.3组胺试验试剂盒
小鼠HIS Elisa试剂盒,E-20451北京永辉科技有限公司生产,批号:201306。

1.2.1实验方法
A小鼠皮肤4-AP致痒模型对致痒阈影响实验
按体重将小鼠随机分为空白对照、基质组、宝乐果组、阳性对照组,阳性对照+宝乐果组。使用剃毛器在其背部剃除2cm×3cm面积毛发,受试液均匀涂于剃毛区皮肤上,每天1次,100μL/只,连续涂抹7d,空白对照组不给任何药物。末次给药40min后于颈背部涂药部位的皮下注射4-氨基吡啶(生理盐水配成0.02%)溶液0.01mL/g,空白对照组注射生理盐水。记录各组在10min内的舔体次数,并计算反应抑制率。
B小鼠皮肤4-AP致痒实验后皮下粘液透皮实验
致痒实验后,分别将各组小鼠处死,在背部剃毛部位开一切口,揭开皮肤表层,用注射器抽取皮下粘液至试管,冷链即时送精密仪器室做HPLC-MS/MS实验,测试透皮液中宝乐果-8,9环烯醚萜苷的含量。
3、试验结果
宝乐果-8,9环烯醚萜苷成分、多酚在各样品中的透过率分别如表2、3所示。

由表2可知,不含宝乐果的抗敏精华液和抗敏精华液阳性对照中未检出环烯醚萜苷成分,而含宝乐果的抗敏精华液和含宝乐果和阳性成分的抗敏精华液中均检测到表格环烯醚萜苷的透皮吸收;从表3可以看出,含宝乐果的抗敏精华液和含宝乐果和阳性成分的抗敏精华液中宝乐果营养成分中的多酚也存在透皮吸收效果。
最后所应当说明的是,以上实施例仅用以说明本发明的技术方案而非对本发明保护范围的限制,尽管参照较佳实施例对本发明作了详细说明,本领域的普通技术人员应当理解,可以对本发明的技术方案进行修改或者等同替换,而不脱离本发明技术方案的实质和范围。

1.宝乐果在透皮吸收中的用途。
2.如权利要求1所述的用途,其特征在于,所述透皮吸收是指宝乐果的活性成分透过皮肤角质层,渗入皮肤深层参与新陈代谢发挥功效作用。
3.如权利要求2所述的用途,其特征在于,所述活性成分为宝乐果8,9-环烯醚萜苷。
4.宝乐果在用于可经透皮吸收的护肤型化妆品中的用途。
5.如权利要求4所述的用途,其特征在于,所述经透皮吸收是指宝乐果的活性成分能透过皮肤角质层,渗入皮肤深层参与新陈代谢发挥功效作用。
6.如权利要求5所述的用途,其特征在于,所述活性成分为宝乐果8,9-环烯醚萜苷。
7.如权利要求4所述的用途,其特征在于,所述化妆品为防晒乳、防晒霜、防晒水、精华液、爽肤水、面膜、眼霜、面霜或乳液。
8.如权利要求4所述的用途,其特征在于,所述化妆品的剂型为水剂、乳剂、膏剂、粉剂或霜剂。
9.如权利要求4~8任一项所述的用途,其特征在于,所述宝乐果在化妆品中的质量百分含量为0.01~5%。
10.如权利要求4~8任一项所述的用途,其特征在于,所述宝乐果的透皮效果同护肤品中宝乐果添加的量呈剂量效应关系。
