背景技术:
免疫力是人体自身的防御机制,是人体识别和消灭外来侵入的任何异物(病毒、细菌等)。处理衰老、损伤、死亡、变性的自身细胞以及识别和处理体内突变细胞和病毒感染细胞的能力。正常机体在免疫系统的作用下,突变的细胞会被抑制和分解。但对于免疫力低下的人群来说,由于免疫活性细胞不能识别突变细胞,致使突变细胞无序繁殖,最终导致疾病产生。特别是儿童,由于其生理结构和功能发育的不完善,更容易受到环境病原体及家长为了“预防感染”而给孩子使用抗生素所带来的不良影响,需要借助外来的免疫制剂来提高对某些疾病的抵抗能力。
炎症是具有血管系统的活体组织对损伤因子所发生的防御反应,在炎症过程中,一方面损伤因子直接或间接造成组织和细胞的破坏,另一方面通过炎症充血和渗出反应,以稀释、杀伤和包围损伤因子。同时通过实质和间质细胞的再生使受损的组织得以修复和愈合。因此可以说炎症是损伤和抗损伤的统一过程。炎症的原因有生物、物理、化学、变态反应等原因。细菌、病毒、立克次体、支原体、真菌、螺旋体和寄生虫等为炎症最常见的原因,儿童也因是易感人群而缕遭感染。尤其是抗生素的滥用,这种化学炎症因子对孩子的危害远远大于成人,且过早的使用抗生素会增加儿童患哮喘风险、耐药、易致肝肾受损或造成二重感染等一系列副反应或者死亡。物理、生物、变态反应等原因导致的感染性疾病在儿童群体的发病率也逐年上升。
因此,提高身体免疫力,预防生物性因子或变态反应感染,找到副作用少的较安全的抗炎微生态食品,已成为当今迫切解决的一个话题。
益生菌是利用葡萄糖等碳水化合物时能产生乳酸的一类细菌。益生菌有一个共同特征,就是在有氧和无氧条件下都能生长,即它们多是兼性厌氧菌。益生菌与人类关系十分密切,其是人体肠道中最重要的有益菌群,对人体的营养、免疫、保健,甚至抗衰老、长寿都有重要作用。人体肠道内益生菌的比例越高,微生态环境就越好,肠道就越显年轻有活力。正常情况下,儿童体内益生菌占优势,控制住坏菌处于健康状态,若生病、滥用药物等,益生菌的优势地位就要受到威胁,导致食欲下降、消化吸收功能差、体弱消瘦、抵抗力下降,若肠道内有益菌数量不能及时恢复,有害菌就会乘机繁殖,使儿童反复生病。因此,及时补充益生菌,可以帮助提高抵抗力。
虽然市面上有很多种儿童益生菌产品,但用以研发具有抗炎作用者尚少,尤其对于儿童群体。本发明研发思路以中医学整体观念、辩证论治的理念体系为支撑,结合中药现代药理作用,同时要符合药食同源前提下的一方益多人的食品特点,对待外来侵袭机体的病原菌从原有的杀灭消亡的方式转变为以菌治菌,以“平衡”之理念,遵循中医古方配伍原则,发扬先传承再创新精神,按照不同物料的作用机制,组成了长、中、短三效的混合配方,还人体原本的自然康健。
发明内容:
本发明要解决的技术问题是公开一种儿童抗炎益生菌固体饮料。是将益生菌、益生元、中药、真菌、免疫球蛋白、肽类物质、维生素等共同配伍,起到协同增效、标本兼治的目的。
本发明方案采用嗜酸乳杆菌、鼠李糖乳杆菌、两歧双歧杆菌、青春双歧杆菌、短双歧杆菌、植物乳杆菌、低聚异麦芽糖、低聚半乳糖、人参、甘草、蒲公英、乌梅、香菇、蛋黄球蛋白粉、豌豆肽粉、酵母β-葡聚糖、针叶樱桃粉、木糖醇、脱脂乳粉为原料组成,原料的重量百分比为:
以上原料均为干品。
其制备方法是将甘草、蒲公英、乌梅、香菇、人参通过水提取、浓缩、干燥、粉碎,过筛,备用;将嗜酸乳杆菌、两歧双歧杆菌、鼠李糖乳杆菌、青春双歧杆菌、短双歧杆菌、植物乳杆菌、低聚异麦芽糖、低聚半乳糖、酵母β-葡聚糖、蛋黄球蛋白粉、豌豆肽粉、针叶樱桃粉、木糖醇、脱脂乳粉与粉碎过筛后的备用粉混合均匀,制得本发明食品。
其中甘草、乌梅、蒲公英、人参提取方法分别如下:
甘草:加水回流提取3次(15重量倍数的水、2.5小时)
乌梅:加水回流提取3次(料液重量比1∶15、温度90℃、时间2小时)
蒲公英:加水回流提取3次(加水20重量倍数、15重量倍数、10重量倍数,每次时间3小时、2小时、2小时)
人参:加水回流提取3次(料液重量比1∶15、温度100℃、时间3小时),所得浸膏用离心喷雾干燥机组,加电加热,进风温度140-230℃,排风温度70-95℃,蒸发量150Kg/h,膏粉终水分3.5-4.0%。
所述的过筛为80目筛。
本发明食品剂型为固体饮料。
益生菌是一类对宿主有益的活性微生物,是定植于人体肠道、生殖系统内,能产生确切健康功效从而改善宿主微生态平衡、发挥有益作用的活性有益微生物的总称。
嗜酸乳杆菌作为益生菌的一种,可以调整肠道菌群平衡,抑制肠道不良微生物的增殖,分泌抗生物素类物质,对致病微生物具有拮抗作用。在人体整个胃肠道中,"从上而下"调节微生态环境。"下"是指在大肠中,它们也可以释放有益双歧杆菌等其他益生菌生长的物质,增加大肠内益生菌的数量、增强它们的生命力。
鼠李糖乳杆菌从属于乳杆菌属,是人体正常菌群之一,肠道黏着率高,定植能力强,并具有高效降胆固醇,促进细胞分裂,可起到调节肠道菌群、预防和治疗腹泻、排除毒素、预防龋齿、提高机体免疫力及抗癌等重要的生理保健功能。
短双歧杆菌是1899年由法国学者从母乳营养儿的粪便中分离出的一种厌氧的革兰氏阳性杆菌,末端常常分叉,故名双歧杆菌,具有调节人体肠道,治疗慢性腹泻与抗生素相关性腹泻、改善人体健康的功效。
青春双歧杆菌具有显著的抗衰老作用,可明显增加血中超氧化物歧化酶的活性与含量,从而减少自由基的氧化反应和对人体细胞的损伤,具有延年益寿的功效。还具有降低胆固醇和甘油三酯的作用,使胆固醇转化成人体不吸收的粪甾醇,从粪便排出体外。同时对于慢性腹泻及便秘具有一定的功效。
两歧双歧杆菌为革兰氏阳性菌,细胞杆状,单生、成对、V字或堆状排列,不产芽胞。严格厌氧,发酵葡萄糖产乳酸。两岐双歧杆菌可治疗慢性腹泻。通过用两岐双歧杆菌对慢性腹泻患者临床观察研究表明,在服药两周以后,患者大便次数、形状正常,临床症状消失,总有效率为90.3%,复发率低。许多国内医院已将两岐双歧杆菌制剂作为治疗慢性腹泻的首选药物。
植物乳杆菌属于同型发酵乳酸菌。此菌与其他乳酸菌的区别在于此菌的活菌数比较高,能大量的产酸,而且其产出的酸性物质能杀灭肠道内有害菌。由于此菌是厌氧细菌(兼性好氧),在繁殖过程中能产出特有的乳酸杆菌素,对致病菌有抑制作用。
六种所添加菌种均来自我国卫生部公布的《可用于食品的菌种名单》,因此可以放心食用。
本发明特别添加了益生元。益生元是指不易被消化的食品成分,通过选择性的刺激一种或几种细菌的生长与活性而对寄主产生有益的影响,从而改善寄主健康的物质。益生元给益生菌提供“食物”,能够被肠道内有益细菌分解吸收,促进有益细菌生长繁殖。
低聚异麦芽糖能有效的促进人体内有益细菌-双歧杆菌的生长繁殖,故又称为“双歧杆菌生长促进因子”,简称“双歧因子”。糖浆为无色或浅黄色,透明的粘稠液体,甜味柔和,无异味,无肉眼可见杂质;糖粉为无定型粉末,甜味柔和,无异味,无肉眼可见杂质,温水溶解为无色或浅黄色。
低聚半乳糖是一种具有天然属性的功能性低聚糖,其分子结构一般是在半乳糖或葡萄糖分子上连接1~7个半乳糖基。在自然界中,动物的乳汁中存在微量的GOS,而人母乳中含量较多,婴儿体内的双歧杆菌菌群的建立很大程度上依赖母乳中的GOS成分,对益生菌有很显著的增殖效果。
两种益生元均为食品级安全可用。
食用真菌:在中国以真菌作为药材治疗人类疾病有着悠久的历史。它们在生长、发育的代谢活动中,能于菌丝体、菌核或子实体内产生酶、蛋白质、脂肪酸、氨基酸、肽类、多糖、生物碱、甾醇、萜类、苷类以及维生素等具有药理活性或对人体疾病有抑制或治疗作用的物质,临床上或是直接利用菌丝体、菌核或子实体,或是利用从菌体中分离出来的有效物质。
香菇是一种食用真菌。一般食用的部分为香菇子实体,香菇中还含有丰富的食物纤维,经常食用能降低血液中的胆固醇,防止动脉粥样硬化,对防治脑溢血、心脏病、肥胖症和糖尿病都有效。近年来,科学家发现香菇中含有一种“β-葡萄糖苷酶”,这种物质有明显的加强机体抗癌的作用,因此,人们把香菇称为“抗癌新兵”。香菇还能抗感冒病毒,因香菇中含有一种干扰素的诱导剂,能诱导体内干扰素的产生,干扰病毒蛋白质的合成,使其不能繁殖,从而使人体产生免疫作用。
酵母多糖是一种吸附病原菌,抗肿瘤,抗病毒,减轻辐射损伤,激发、增进免疫功能的高分子多糖物质。酵母葡聚糖是第一个被发现具有免疫活性的葡聚糖。国际上天然酵母葡聚糖被誉为"超级灵芝"、"定向清毒,免疫先锋"、"病毒细胞的追捕者”。2009年11月北海道大学的研究小组发现黑酵母β-葡聚糖具有增强人体对新型流感的免疫力,以及阻止重症化的效果。酵母β-葡聚糖是存在于酵母细胞壁中的一种具有增强免疫力活性的多糖,它的免疫增强活性更强,并具有改善血脂、抗辐射、改善肠道功能的作用,完全从酵母中提取得到,天然安全,同时具有较高的生物活性。
两种多糖类物质为药食两用食材,均具有提高免疫力的功效。
维生素是一系列有机化合物的统称。它们是生物体所需要的微量营养成分,而一般又无法由生物体自己生产,需要通过饮食等手段获得。维生素不能像糖类、蛋白质及脂肪那样可以产生能量,组成细胞,但是它们对生物体的新陈代谢起调节作用。
维生素C又叫L-抗坏血酸,是一种水溶性维生素。食物中的维生素C被人体小肠上段吸收。白细胞含有丰富的VC,当机体感染时白细胞内的VC急剧减少。VC可增强中性粒细胞的趋化性和变形能力,提高杀菌能力。同时VC参与免疫球蛋白的合成,促进淋巴母细胞的生成,提高机体对外来和恶变细胞的识别和杀灭。促进干扰素的产生,干扰病毒mRNA的转录,抑制病毒的增生。提高机体的应急能力。
针叶樱桃为金虎尾科植物巴西针叶樱桃、西印度樱桃的果实,是世界上已知含维生素C最丰富的水果。其100克果实中VC含量达2445毫克,远远高于柠檬的40mg,柑橘的68mg和猕猴桃的100mg,是名副其实的“维生素C之王”,针叶樱桃粉富含天然VC成分,儿童应用更安全。
免疫球蛋白指具有抗体活性的动物蛋白。主要存在于血浆中,也见于其他体液、组织和一些分泌液中。人血浆内的免疫球蛋白大多数存在于丙种球蛋白(γ-球蛋白)中。免疫球蛋白可以分为IgG、IgA、IgM、IgD、IgE五类。
免疫球蛋白IgY:普遍存在于鸟类、爬行动物和两栖类,在功能上等价于哺乳动物IgG,但其许多生物学性质仍未被发现。近来许多研究者分析了IgY的基因组成和生物学功能,而且证实其为哺乳动物IgG和IgE的直接起源。卵黄抗体IgY(蛋黄球蛋白粉):通过免疫注射产蛋鸡,即可由其生产的蛋黄中提取相应的抗体,并可用于相应疾病的预防和治疗。特定病原菌的卵黄抗体能直接黏附于病原菌的细胞壁上,改变病原细胞的完整性,直接抑制病原菌的生长;卵黄抗体可黏附于细菌的菌毛上,使之不能黏附于肠道黏膜上皮细胞而失去致病性。蛋黄球蛋白粉是鸡蛋中特有的高生物活性蛋白,经冷冻干燥而成的新产品,含有丰富的IgY,是一种优质的球蛋白资源。
此种免疫球蛋白,为鸡蛋以物理方式精炼而成,安全放心。
肽是一个氨基酸的氨基与另一个氨基酸的羧基可以缩合成肽,形成的酰胺基在蛋白质化学中称为肽键。两个或以上的氨基酸脱水缩合形成若干个肽键从而组成一个肽,分子量段在1000~50之间的称为小肽、寡肽、低聚肽,也称为小分子活性多肽。生物学家将小分子活性多肽统称为"生物活性肽"。具有直接吸收、不需消化、直接作用于细胞、激活细胞活性,快速吸收等特点。
豌豆肽粉是以天然豌豆蛋白为原料采用现在生物酶解技术分离纯化而成。对鼠李糖乳杆菌和嗜酸乳杆菌有明显的促进生长的作用。
此种肽为食品级原料,安全放心食用。
中药是我国的民族瑰宝,随着医源性疾病和药源性疾病的日益增加、健康观念的变化以及医学模式的转变,中药养生模式逐渐深入人心。相较于辩证论治的中药单方精准模式,药食同源的普方预防模式更以天然安全、早期干预更易受到儿童群体的欢迎。选择以下几种药食两用中药,是经过儿科古方严格筛选,通过对古方用药用量的参照,结合现代临床经方实践,得以确定。
甘草是一种补益中草药,以根茎入药,既能调和诸药,又能补气和中、泻火解毒。现代药理表明甘草还有抗炎,抗过敏作用。“弘景曰∶此草最为众药之主,经方少有不用者,犹如香中有沉香也。”即为帝师之称的国老。
蒲公英别名黄花地丁、婆婆丁、华花郎等。菊科多年生草本植物。含有蒲公英醇、蒲公英素、胆碱、有机酸、菊糖等多种健康营养成分。性味甘,微苦,寒。归肝、胃经。有消炎利尿、缓泻、退黄、解毒等功效。
乌梅别名酸梅、黄仔、合汉梅、干枝梅,为蔷薇科落叶乔木植物梅的近成熟果实,归肝、脾、肺、大肠经。敛肺,涩肠,生津,安蛔。含柠檬酸19%、苹果酸15%、琥珀酸、碳水化合物、谷甾醇、蜡样物质及齐墩果酸样物质,亦含有强杀菌性及提高肝脏功能的成分苦味酸及具有热镇痛作用的苦扁桃甙,具有良好的抗病原微生物作用。
人参在我国已有上千年的历史。别名地精、棒棰,为五加科植物,人参为药食同源物品,在《神农本草经》里面列为上等佳品。人参具有补五脏、安精神、定魂魄、止惊吓、除邪气、明目、开心、益智等多种功效,久服延年益寿。因产量稀少,并且滋补效果显著,而在世界享有极高的声誉,故被誉为“百草之王”。
本品在质量标准研究中尤其注重免疫因子的活性及真菌多糖含量,所以把免疫球蛋白IGY含量及真菌中粗多糖的含量作为除益生菌活菌数以外的重要检测指标。
免疫球蛋白IGY含量的检测:蛋白质具有极性的性质,在碱性溶液中PH值高于其等电点时,使其带负电荷,通电后则向正极移动,由于各类蛋白质所负电荷不同而移动速度不同,在通电一定时间后即行分开。以醋酸纤维素薄膜为支撑物,以鸡蛋血清IGY为基准物,在相同条件下进行电泳,在醋酸纤维素薄膜上分离出不同区带,经染色、脱色、洗脱和比色,可计算出样品中免疫球蛋白的含量。本品中免疫球蛋白(以IGY计)的含量在10%以上。
粗多糖的检测执行NY/T1676,以多糖在硫酸作用下,先水解生成单糖,并迅速脱水生成糖醛衍生物,与苯酚反应生成橙黄色溶液,在490nm处有特征吸收,与标准系列比较定量即得多糖含量结果。本品经检验,粗多糖含量在0.8%以上。

本发明将益生菌、益生元、中药、真菌、免疫球蛋白、肽类物质、维生素有机地结合到一起,建立了一种增强人体免疫力的新方法,开发出了具有消炎作用的儿童益生菌新产品。益生菌可促进肠道消化,加速新陈代谢,以菌治菌。益生元可促进益生菌增殖,从整体上提高人体免疫力。免疫球蛋白、真菌及肽类物质三者起效快,具有免疫活性,精准调节免疫系统。中药是重要的免疫调节剂,能够有效的预防及治疗机体气血不足或脏器虚损所引起的各种症状,“治未病”的保健长效机能充分显现出来。加之维生素的协同增效作用,相得益彰,大大提高了人体免疫力,消除炎症因子所带给身体的不良反应,提高身体抵抗能力。具有消炎解毒、调节胃肠功能、提高身体免疫力等效果。

例1、将甘草2公斤、人参5公斤、蒲公英3公斤、乌梅3公斤、香菇10公斤分别经水提、浓缩、干燥、粉碎,过80目筛,备用;将嗜酸乳杆菌3公斤、鼠李糖乳杆菌2公斤、两歧双歧杆菌3公斤、青春双歧杆菌2公斤、短双歧杆菌1公斤、植物乳杆菌1公斤、低聚异麦芽糖15公斤、低聚半乳糖2公斤、蛋黄球蛋白粉10公斤、豌豆肽粉5公斤、酵母β-葡聚糖5公斤、针叶樱桃粉3公斤、脱脂乳粉15公斤、木糖醇10公斤与备用的甘草、人参、香菇、乌梅、蒲公英提取物混合均匀,制得本发明食品。

例3、将甘草1公斤、人参1公斤、蒲公英1公斤、乌梅5公斤、香菇10公斤分别经水提、浓缩、干燥、粉碎,过80目筛,备用;将嗜酸乳杆菌0.5公斤、鼠李糖乳杆菌3公斤、两歧双歧杆菌0.5公斤、青春双歧杆菌0.5公斤、短双歧杆菌1公斤、植物乳杆菌1公斤、低聚异麦芽糖20公斤、低聚半乳糖1公斤、蛋黄球蛋白粉10公斤、豌豆肽粉1公斤、酵母β-葡聚糖5公斤、针叶樱桃粉1公斤、脱脂乳粉27公斤、木糖醇10公斤与备用的甘草、人参、香菇、乌梅、蒲公英提取物混合均匀,制得本发明食品。
