Abstract

技术领域
本发明属于生物技术领域,主要涉及葡聚糖修饰几个Hrps功能多肽组合片段的技术方法,以及通过该技术方法,将葡聚糖和几个Hrps功能多肽片段不同程度的共价复合,生成完全和部分不完全共价三聚肽,以及共价三聚肽产物在多领域的应用。
背景技术
所有生物的细胞表面都由许多不同类型的糖链所包被,而且在细胞内也存在各种类型的糖基化。寡糖链与蛋白质多肽链中的氨基酸以多种形式共价连接,构成糖蛋白的糖肽连接链。糖蛋白是参与许多生物过程的基本物质,这些过程包括细胞生长、细胞与细胞的粘着、免疫应答、受精、凝血块的降解、病毒增殖、寄生虫感染和炎症反应等。随着蛋白质组技术的不断发展,糖基化蛋白质组的研究将越来越受到广泛的重视。
植物在长期进化中,在同周围各种生存和生长环境条件(包括生物的和非生物的)相互作用中,形成了一种共同的适应能力,这就是进化中形成的共同防御系统和生长系统,这是植物共有的一类防御机制和生长机制,并由多条信号通路和代谢途径来实现。
研究表明,Hrp蛋白类作为植物的上游信号物质,与植物细胞壁中广泛存在的受体Hrp结合蛋白(Harpin-binding proteins)结合并激活植物中的多条信号通路,这些信号通路的激活,将实现对植物固有的细胞防御和生长多条代谢途径和机制的启动和调控,这就是Hrp蛋白类作为植物生长调节剂和生物农药的作用机制。目前,称为超敏蛋白的Hrp蛋白类产品已被广泛研究、开发和应用,具有广阔的前景。
随着基因工程技术的发展,越来越多的多肽、酶、信号分子、细胞因子等蛋白类制品被研究开发,这些蛋白类制品具有专一性强、时效高、应用广泛、安全性好等优点,但同时也发现了几个显著的不足之处,比如(1)可溶性和均一度较差;(2)抗原性及免疫原性;(3)容易从循环系统中被快速清除;(4)不稳定,容易被体内外的酶降解;(5)外用时,容易被外界环境的微生物快速降解和光降解。
对于Hrps类蛋白来说,由于植物种类繁多,不同物种间、品种间生物学特性差异较大,内外环境也不同以及基因间互作的差异,不同来源和条件下获得的Hrps类蛋白及其制剂的性质在使用条件和效果上存在明显的选择性差异,单一种类的Hrps蛋白在功效上已经明显表现出不足;Hrps类蛋白作为应用于植物的一类上游功能性信号蛋白质,其可溶性和稳定性较差,同时,在自然条件下使用(叶面喷施和根施时),很容易被外界环境的微生物快速降解和光降解,从而影响它们的时效和功效。
鉴于以上原因,需要对Hrps类蛋白进行结构改造和化学修饰。
结构改造和化学修饰法是指在分子水平上对蛋白质进行局部改造和修饰,即在体外将蛋白质的侧链基团通过人工方法与一些化学基团,特别是具有生物相容性的分子基团和大分子进行共价连接,形成完全和部分不完全的共价复合物,从而部分改变蛋白质的性质和结构,并显示出比各自独立存在时更优越的性能,该技术和方法已经在医药卫生和食品业的蛋白质产品的应用上引起越来越广泛的重视。
需要强调指出的是,对不同来源的Hrps类蛋白多肽进行组合、结构改造和化学修饰形成共价复合物,国内外目前均未见报道。
常见用于蛋白质修饰的分子基团和大分子很多,主要有聚乙二醇类(包括PEG和mPEG等)、多糖类,以及同源蛋白质及人工合成的多肽类、聚氨基酸等。对蛋白质的结构改造和化学修饰技术近几年来发展很快,针对不同的酶、多肽或蛋白质的化学修饰形成了多种技术和方法,但是,都有其适用范围和局限性,至今还没有普适的通用技术和方法可供选择和利用,随着蛋白质修饰技术研究的深入和发展,必将进一步促成能提高对各种不同蛋白质的修饰特异性好、修饰率高、操作简便、产物稳定性和功效性好的新技术和新方法的产生。
发明内容
本发明目的是提供一种葡聚糖修饰的Hrps共价三聚肽及其制备方法,使Hrps多肽乳化稳定性、均一性和结构稳定性更高、时效更长,功能更多,应用范围更广。
本发明的技术方案为:葡聚糖修饰的Hrps共价三聚肽,它由三种Hrps多肽和葡聚糖通过共价修饰形成共价三聚肽,其结构通式为:(3Hrps)n-(C6H10O5)n;其中,三种Hrps多肽为SEQ ID NO:1,SEQ ID NO:2,SEQ ID NO:3,SEQ ID NO:4所示的氨基酸序列中的任意三种。
进一步地,SEQ ID NO:1,SEQ ID NO:2,SEQ ID NO:3,SEQ ID NO:4的质量配比为3:2:3:2。
进一步地,三种Hrps多肽与葡聚糖的质量配比为10:1至1:10。
进一步地,三种Hrps多肽与葡聚糖的质量配比为1:2。
进一步地,葡聚糖为燕麦β-葡聚糖,分子质量为5300-257200。
进一步地,葡聚糖为燕麦β-葡聚糖,分子质量为32000-38000。
葡聚糖修饰的Hrps共价三聚肽的制备方法,制备步骤如下:
(1)从SEQ ID NO:1、SEQ ID NO:2、SEQ ID NO:3、SEQ ID NO:4所示的四种Hrps多肽中任意选择三种Hrps多肽;
(2)向步骤(1)选择的三种Hrps多肽中加入葡聚糖,三种Hrps多肽与葡聚糖的质量配比为:10:1至1:10;加入磷酸缓冲液,使体系pH值为2-10;静置0-5小时,在温度为15-80℃、搅拌速度为30-300r/min条件下反应3-25小时;
(3)反应完成后,低温真空连续干燥,收集产物即得共价三聚肽,(3Hrps)n-(C6H10O5)n。
进一步地,步骤(2)中,磷酸缓冲液的浓度为10-200mmol/L,磷酸缓冲液加入量为三种Hrps多肽和葡聚糖总质量的10-100倍。
优选地,制备方法如下:
(1)从SEQ ID NO:1、SEQ ID NO:2、SEQ ID NO:3、SEQ ID NO:4所示的四种Hrps多肽中任意选择三种Hrps多肽;其中,SEQ ID NO:1、SEQ ID NO:2、SEQ ID NO:3、SEQ ID NO:4四种Hrps多肽的质量配比为3:2:3:2。
(2)向步骤(1)选择的三种Hrps多肽中加入葡聚糖,葡聚糖为燕麦β-葡聚糖,分子质量为32000-38000;三种Hrps多肽与葡聚糖的质量配比为:1:2;加入浓度为50mmol/L的磷酸缓冲液,磷酸缓冲液加入量为三种Hrps多肽和葡聚糖总质量的40倍,使体系pH值为7,静置1小时;在温度为46℃、搅拌速度为60r/min条件下反应12小时;
(3)反应完成后,低温真空连续干燥,收集产物即得共价三聚肽,(3Hrps)n-(C6H10O5)n。
四个Hrps功能多肽片段分别是:
SEQ NO ID:1所示的Hrps多肽片段来源于由356个氨基酸组成的HrpsNEccs基因编码蛋白所含的Harpin家族蛋白质高度保守区域片段,由200个氨基酸组成,位于蛋白的C-端(氨基酸157-356)。
SEQ NO ID:2所示的Hrps多肽片段来源于由370个氨基酸组成的HrpsZpst基因编码蛋白所含的Harpin家族蛋白质高度保守区域片段,由312个氨基酸组成,位于蛋白的N-端及C-端(氨基酸1-270和329-370)。
SEQ NO ID:3所示的Hrps多肽片段来源于由370个氨基酸组成的HrpsNEcb基因编码蛋白所含的Harpin家族蛋白质高度保守区域片段,由200个氨基酸组成,位于蛋白的C-端(氨基酸171-370)。
SEQ NO ID:4所示的Hrps多肽片段来源于由339个氨基酸组成的HrpsNEch基因编码蛋白所含的Harpin家族蛋白质高度保守区域片段,由326个氨基酸组成,位于蛋白的中段(氨基酸9-334)。
这些功能多肽片段都是专利发明人自主完成的基因及其表达产物研究成果,已由专利发明人在GenBank注册,它们注册号分别是:AY999002.1,AY999004,DQ355519.1及AY999001;按照这些功能多肽片段的大小、在保守区域位置和序列差异,以及Hrp蛋白基因来源细菌的特征,互补性地选择了这些片段,配置了四个组合,每种组合含有3个不同的多肽片段,将要进行的修饰多肽的四个片段组合分别是,I:1号、2号、3号,II:1号、2号、4号,III:1号、3号、4号和IV:2号、3号、4号,它们的表达通式是(3Hrps)n,四个片段组合的表达式分别为(3Hrps)n(I)、(3Hrps)n(II)、(3Hrps)n(III)和(3Hrps)n(IV)。
对以上4个Hrps功能多肽片段作进一步的说明:
1、几个Hrps功能多肽来源的源头基因编码的Harpin蛋白,它们的大小和氨基酸序列组成都差异较大。1号HrpNEccs蛋白由356个氨基酸组成;2号HrpZpst蛋白由370个氨基酸组成;3号HrpNEcb蛋白由370个氨基酸组成;而4号HrpNEch蛋白只有339个氨基酸。
2、四个基因编码的Harpin蛋白都含Harpin家族蛋白质高度保守区域(DOMAIN)。Harpin家族蛋白质高度保守区域是Harpin类蛋白的主要功能区域,其卓越的生物功效主要由所含Harpin家族蛋白质高度保守区域的序列,以及这些保守区域的多肽高级结构所决定。
3,四个Harpin蛋白及其所含之Harpin家族蛋白质高度保守区域间的氨基酸序列无论从序列大小、位置以及氨基酸组成,它们间的差异都极大。1号蛋白的Harpin家族蛋白质高度保守区域由200个氨基酸组成,位于蛋白的C-端(氨基酸157-356);2号蛋白的Harpin家族蛋白质高度保守区域由312个氨基酸组成,位于蛋白的N-端及C-端(氨基酸1-270和329-370);3号蛋白的Harpin家族蛋白质高度保守区域由200个氨基酸组成,位于蛋白的C-端(氨基酸171-370);4号蛋白的Harpin家族蛋白质高度保守区域由326个氨基酸组成,位于蛋白的中段(氨基酸9-334)。这4个Harpin家族蛋白质高度保守区域的多肽片段氨基酸序列以及保守区域的多肽高级结构决定了它们具有的卓越的生物功效。

燕麦中的β-(1→3,1→4)葡聚糖,简称燕麦β-葡聚糖,是一种非淀粉多糖。它由单体β-D-吡喃葡萄糖,通过β-(1→3)和β-(1→4)糖苷键连接起来形成的一种高分子聚合物。85%以上的燕麦β-葡聚糖分子中每2-3个β-(1→4)糖苷键间有1个β-(1→3)糖苷键连接,15%是由长链β-(1→4)糖苷键间隔1个β-(1→3)糖苷键组成,其长度可能有4、5个或8个葡萄糖残基。具有β-(1→3)键的直链状而无分支的多糖体结构,其中β-(1→4)和β-(1→3)糖苷键的比例约为2.4:1。在燕麦胚乳和糊粉层细胞壁成分中,β-葡聚糖占85%以上。燕麦β-葡聚糖是一种相对分子质量较小的短链葡聚糖,其相对分子质量的变化范围为5300-257200。

(3)多肽片段四个组合中的Hrps功能多肽片段和葡聚糖的质量配比,通常为10:1至1:10之间;
(4)反应浓度,Hrps多肽组合片段和葡聚糖混合,10-200mM磷酸缓冲液稀释为混合物质量的10至100倍;
(5)反应pH和静置处理时间,通常为10-200mM磷酸缓冲液pH2-10和0-5小时;

应用这类功能多肽共价聚合体时:(1)处理植物的适宜方法包括浸种、拌种、种子包衣、蘸根、叶面喷雾、根施、注射、花果喷涂等;(2)处理植物的适宜条件包括选择适宜的用药时间和用药时的环境气象条件;颗粒剂、可湿性粉剂或水剂应用无氯的河水、井水、地下水或自来水配成要求的使用浓度,切忌与酸、碱混合,药品应保存在阴凉干燥处;(3)处理植物的具体要求包括整个生长季节均适于用药,一季作物可施药3-6次,每次间隔10-20天,施用时间和次数视使用目的确定,使用时已配制好的药液建议在24小时内用完。
实施例4
葡聚糖修饰Hrps功能多肽片段制备的四个组合的(3Hrps)n-(C6H10O5)n(I-IV)共价三聚肽的稳定性检测:
我们按实施例1方法制备的(3Hrps)n-(C6H10O5)n(I-IV)共价三聚肽与对应的Hrps多肽组合的混合片段,在几种条件下的稳定性进行了比较,结果如下:
在室温下,均配制成含量3%的水溶液,Hrps多肽组合的混合片段水溶液静置20分钟后,出现明显的分层,1小时后,形成絮状沉淀;(3Hrps)n-(C6H10O5)n(I-IV)共价三聚肽水溶液静置6小时后,溶液仍均匀,未出现分层。
裸露在自然条件下,均配制成含量3%的水溶液,分别在培养皿注入一薄层配制的水溶液,Hrps多肽组合的混合片段水溶液3天后,取样电泳时Hrp蛋白谱带已经很弱了,5天后,Hrps蛋白谱带已经消失,表明Hrps多肽基本解体;(3Hrps)n-(C6H10O5)n(I-IV)共价三聚肽水溶液7天后,电泳时它们的谱带还清晰可见。
在100℃条件下,均配制成含量3%的水溶液,Hrps多肽组合混合片段水溶液加热25分钟后,注射不再引起烟草叶片过敏反应,Hrps多肽基本丧失活性;(3Hrps)n-(C6H10O5)n(I-IV)共价三聚肽水溶液加热30分钟后,注射引起烟草叶片过敏反应,共价三聚肽仍有活性,50分钟后还有较弱的活性保持。
暴露在30000Lux强光照条件下,均配制成含量3%的水溶液,分别在培养皿注入一薄层配制的水溶液,Hrps多肽组合混合片段水溶液强光照60分钟后,注射不再引起烟草叶片过敏反应,Hrps多肽丧失活性;(3Hrps)n-(C6H10O5)n(I-IV)共价三聚肽强光照120分钟后,注射引起烟草叶片过敏反应,仍有活性,150分钟后不再引起烟草叶片过敏反应,才丧失活性。

结果表明,(3Hrps)n-(C6H10O5)n(I-IV)共价三聚肽、单一HrpEcb蛋白和对应的Hrps多肽组合的混合片段都具有诱导供试植物抗性的能力和生物学活性,在诱导过敏反应的植物种类范围以及诱导抗性的能力和生物学活性上,(3Hrps)n-(C6H10O5)n(I-IV)共价三聚肽最优、最强,其次是对应的Hrps多肽组合的混合片段,再其次是单一HrpEcb蛋白,显然,(3Hrps)n-(C6H10O5)n(I-IV)共价三聚肽具有更广的使用范围和更强的诱抗能力。

(1)四川省德阳市2个市县的5个镇村进行了20余批次大田的共价三聚肽试验和示范,实施面积总量360余亩,主要供试作物为土豆、黄瓜、玉米、辣椒、西红柿、晒烟、红油菜、花椰菜、水稻、桑树、油桃等,增产幅度为8%至36%,表现出对病虫害和逆境的显著诱抗作用,提高了产品品质和商品性,并延长了产品的保存期。
(2)四川省成都市5个区县的7个村进行了三个生长季节10余批次大田的共价三聚肽试验和示范,实施面积总量280余亩,主要供试作物为土豆、黄瓜、辣椒、茄子、芹菜、白菜、茶叶(白茶)、草莓、果桑、葡萄、枇杷,薯蓣以及紫金花、仙客来、蝴蝶兰、三色堇、一品红、矮牵牛、非洲凤仙花、菊花、倒挂金钟、金鱼草、玻璃海棠等二十余种花卉,蔬菜和水果类增产幅度为12%至32%,表现出对病虫害和逆境的显著诱抗作用,提高了产品品质和商品性,并延长了产品的保存期;花卉表现出对病虫害和逆境的显著诱抗作用,塑型作用较强,提高了产品商品性,并延长了花期,增加了花数和色彩更为鲜艳;茶叶提高了品质,并可以多采摘1次,经济效益显著。

Claims (9)
1.葡聚糖修饰的Hrps共价三聚肽,其特征在于,它由三种Hrps多肽和葡聚糖通过共价修饰形成共价三聚肽,其结构通式为:(3Hrps)n-(C6H10O5)n;其中,三种Hrps多肽为SEQ IDNO:1,SEQ ID NO:2,SEQ ID NO:3,SEQ ID NO:4所示的氨基酸序列中的任意三种。
2.根据权利要求1所述的葡聚糖修饰的Hrps共价三聚肽,其特征在于,SEQ ID NO:1,SEQ ID NO:2,SEQ ID NO:3,SEQ ID NO:4的质量配比为3:2:3:2。
3.根据权利要求1所述的葡聚糖修饰的Hrps共价三聚肽,其特征在于,三种Hrps多肽与葡聚糖的质量配比为10:1至1:10。
4.根据权利要求1所述的葡聚糖修饰的Hrps共价三聚肽,其特征在于,三种Hrps多肽与葡聚糖的质量配比为1:2。
5.根据权利要求1所述的葡聚糖修饰的Hrps共价三聚肽,其特征在于,葡聚糖为燕麦β-葡聚糖,分子质量为5300-257200。
6.根据权利要求1所述的葡聚糖修饰的Hrps共价三聚肽,其特征在于,葡聚糖为燕麦β-葡聚糖,分子质量为32000-38000。
7.根据权利要求1-6任一项所述的葡聚糖修饰的Hrps共价三聚肽的制备方法,其特征在于,制备步骤如下:
(1)从SEQ ID NO:1、SEQ ID NO:2、SEQ ID NO:3、SEQ ID NO:4所示的四种Hrps多肽中任意选择三种Hrps多肽;
(2)向步骤(1)选择的三种Hrps多肽中加入葡聚糖,三种Hrps多肽与葡聚糖的质量配比为:10:1至1:10;加入磷酸缓冲液,使体系pH值为2-10;静置0-5小时,在温度为15-80℃、搅拌速度为30-300r/min条件下反应3-25小时;
(3)反应完成后,低温真空连续干燥,收集产物即得共价三聚肽,(3Hrps)n-(C6H10O5)n。
8.根据权利要求7所述的葡聚糖修饰的Hrps共价三聚肽的制备方法,其特征在于,步骤(2)中,磷酸缓冲液的浓度为10-200mmol/L,磷酸缓冲液加入量为三种Hrps多肽和葡聚糖总质量的10-100倍。
9.根据权利要求7所述的葡聚糖修饰的Hrps共价三聚肽的制备方法,其特征在于,制备方法如下:
(1)从SEQ ID NO:1、SEQ ID NO:2、SEQ ID NO:3、SEQ ID NO:4所示的四种Hrps多肽中任意选择三种Hrps多肽;其中,SEQ ID NO:1、SEQ ID NO:2、SEQ ID NO:3、SEQ ID NO:4四种Hrps多肽的质量配比为3:2:3:2;
(2)向步骤(1)选择的三种Hrps多肽中加入葡聚糖,葡聚糖为燕麦β-葡聚糖,分子质量为32000-38000;三种Hrps多肽与葡聚糖的质量配比为:1:2;加入浓度为50mmol/L的磷酸缓冲液,磷酸缓冲液加入量为三种Hrps多肽和葡聚糖总质量的40倍,使体系pH值为7,静置1小时;在温度为46℃、搅拌速度为60r/min条件下反应12小时;
(3)反应完成后,低温真空连续干燥,收集产物即得共价三聚肽,(3Hrps)n-(C6H10O5)n。
