Abstract
本发明涉及一种利用餐厨废液生产液态有机菌肥的方法。具体方法为:选择巨大芽孢杆菌、解脂假丝酵母和假单胞菌。制备假单胞菌斜面培养基、巨大芽孢杆菌斜面培养基、热带假丝酵母斜面培养基。将假单胞菌、解脂假丝酵母、巨大芽孢杆菌分别转入新鲜的斜面培养基,在30℃培养24h进行活化。将活化好的菌种分别放入去除琼脂的培养基,接种量均为2%,均在30℃培养18小时,摇床转速均为180转/分,获得活化菌液。将活化好的假单胞菌、巨大芽孢杆菌、解脂假丝酵母菌液以1∶1∶1的比例,接种到餐厨垃圾废液中,接种量为2%。加入60%的畜禽粪便辅料,制得扩培菌液。复配好的菌液在30℃下发酵72小时,即获得液态有机菌肥。

本发明涉及一种废物资源再生利用领域,特别涉及一种包含菌种扩培系统的利用餐厨废液生产液态有机菌肥的方法。
背景技术
餐厨废液COD最高可达100000mg/L以上,属于高浓度有机废水,如果排入水体,会造成严重的富营养化问题,而且处理达标排放则成本很高。因此,利用富含有机成分的餐厨废液作为营养液经过微生物扩培生产液态菌肥,不仅可以消除废水排放,而且实现了废液的高度资源化利用。菌肥是复合生物肥料的俗称,是采用人工方法培养某些有益微生物而制成的生物肥料。这种含有微生物的特定制品应用于农业生产中,能够获得特定的肥料效应,制品中活微生物的生命活动起关键作用。复合生物菌肥含有二种以上有益微生物,它们之间不拮抗,并能提高农作物一种或几种营养元素的供应水平。其肥料对改善土壤结构,增强土壤肥力,促进作物生长,增强作物抗病能力等方面具有重要作用。生物菌肥内的多种功能性微生物进入土壤后,在生长繁殖过程中产生大量的次生代谢产物,这些产物能够促进土壤团粒结构的形成。团粒结构的形成使土壤变的疏松、绵软,保水保肥性能增加,水、气、热更加协调,减少土壤板结,有利于保水、保肥、通气和促进根系发育,为农作物提供舒适的生长环境。土壤理化性状的改善,加强了土壤有益微生物的活动,从而最大限度促使有机物分解转化,产生多种营养物质和刺激性物质,促使微生物的生长发育,促进作物生长,最终达到增产增收的目的。另外,功能性微生物在作物根系周围形成优势菌群,抑制或拮抗有害微生物的生长繁殖,减轻了作物发生病害的程度。
在国际上,液体肥料在一些发达国家已经得到了较为普遍的应用,如美国、英国、澳大利亚、加拿大、德国、比利时、荷兰等国的液体肥料在整个肥料施用量中占有相当大的比例。我国对液体肥料的开发研究始于20世纪50年代,70年代在北京、浙江、山东、河北、广东、新疆等地开展示范和推广工作。国内生产的液体肥料除大面积根部施用外,一部分也用作叶面施肥,如对小麦、玉米、棉花、水稻、甜菜、油葵等农作物用液体肥进行根施灌溉,而对蔬菜、水果、园林、花卉等经济作物在生长期内经常喷施液体肥。烟草产区除施基肥外,还在烟草生长期内喷施或灌溉液体肥。目前,我国还没有液体肥料使用的详细统计数据,初步估计,每年施用量约为30-40万吨。
液体肥料的特点为,(1)生产费用低。化肥生产企业可以省去一部分生产固体肥料所必需的设备或工段,如造粒、干燥等,有利于降低生产装置投资和维修费用,减少燃料和动力消耗。(2)有利于环境保护。液体肥料在生产和运输过程中无粉尘、无烟雾,减少了因“三废”排放可能对环境造成的污染。由于去掉了部分设备,生产过程中的噪声污染也会相应降低。(3)施用方便。液体肥料可以直接施入土壤,也可以作为果树、蔬菜、花卉等各种作物的叶面肥直接喷施,还可以与农药结合起来喷洒。由于配方灵活,各种营养元素均匀合理,适用于作基肥、追肥。(4)直接被农作物吸收。液体肥料施入土壤后,营养成分可直达根系,可以降低某些营养成分被土壤固定的数量。液体肥料作为叶面肥直接喷施时,养分吸收比土壤施肥快,可以较快克服农作物养分缺乏症状,能使植物迅速从叶面部位吸收营养。
(5)降低施肥成本。由于液体肥料施用方便、有利于被农作物吸收,因而可以减少对农作物的施肥次数,节省人力支出和物力消耗。随着液体肥料的规模化生产和机械化施用的步伐加快,施肥成本将会进一步降低。(6)液体肥料在施用上也有一定的局限性,如运输费用相对较高,用于土壤施肥的液体肥料需要专门的贮存和运输设备,而且还需要配置专门的施用工具等。随着技术的发展和配套设施的不断完善,这些制约因素将逐渐被取消。对于一些小规模的、特种用途的液体复混肥目前则不存在这些问题。
总的看来,利用餐厨废液生产液态有机菌肥,既减少污染,又节省废水处理费用,同时变废为宝,促进相关产业发展,经济效益和环境效益显著。
发明内容
为了解决污水处理和废水资源化利用,实现餐厨垃圾处理的零排放问题,填补利用餐厨废液生产液态菌肥技术的空白,本发明提供一种利用餐厨废液生产液态有机菌肥的方法。本发明实现污水资源利用,转化率高,利用生物技术,能耗很低,无二次污染。
本发明通过下面的技术方案,实现利用餐厨废液生产液态有机菌肥的方法,该技术现污水资源利用,转化率高,利用微生物扩培技术,操作简便,能耗很低,无二次污染。
本发明的利用餐厨废液生产液态有机菌肥的方法如下:
整个方法包括如下几部分:菌种选择、菌种培育和活化、菌液复配,菌种扩培和液态有机菌肥制备。整个生产过程没有废气、废液和废渣产生,环境风险低。且废水中有机物再利用,不仅节省废水处理费用,且创造价值,环境效益和经济效益显著。
(1)菌种的选择
本发明所用的菌种均为农业部关于微生物肥料生物安全通用技术准则(NY11092006)附录中规定使用的生产菌种。包括但不限于芽胞菌和酵母菌,所说的芽胞菌为巨大芽孢杆菌或地衣芽胞杆菌、或枯草芽胞杆菌,优选巨大芽胞杆菌。所说的酵母菌为或乳酸酵母菌,优选解脂假丝酵母。另外,本发明的混合菌群中还包括假单胞菌。
(2)菌种培育和活化方法
本发明所说的菌种培育方法如下:
①假单胞菌斜面培养基制备方法(牛肉膏蛋白胨琼脂培养基)
牛肉膏3g;蛋白胨10g;NaCl5g;琼脂;水1000mL;pH7.4-7.6
②巨大芽孢杆菌斜面培养基制备方法(磷细菌培养基)-
葡萄糖10g;NaCI0.3g;MgSO40.3g;MnSO40.03g;CaCO31.0g(NH4)2SO40.5g;KCl0.3g;FeSO40.03g;卵磷脂0.2g;酵母粉0.5g;琼脂;水1000mL;pH7.0-7.2
③热带假丝酵母斜面培养基制备方法(豆芽汁培养基)
黄豆芽100g;葡萄糖20g;水1000mL;琼脂;pH7.2-7.4
配制方法如下:洗净黄豆芽,加水煮沸30min。用纱布过滤,滤液中加入琼脂,加热溶解后放入糖,搅拌使它溶解,补足水分到1000mL,pH7.2-7.4
④摇瓶培养基:斜面培养基去除琼脂作为摇瓶培养基
⑤菌种活化及菌液制备方法:将假单胞菌、解脂假丝酵母、巨大芽孢杆菌分别转入新鲜的斜面培养基,在30℃培养24h,得到活化的菌种。将活化好的菌种分别放入摇瓶培养基,接种量均为1%~3%,优选2%,均在20℃~40℃下培养10~30小时,优选地,均在
30℃培养18小时,摇床转速均为100转/分~300转/分,优选180转/分。获得活化后的菌液。
(3)混合菌液复配方法
将活化好的假单胞菌、巨大芽孢杆菌、解脂假丝酵母菌液以1∶1∶1的比例,添加到餐厨垃圾废液中,接种量为1%~3%,优选2%。然后,加入畜禽粪便辅料,加入量为40%~80%,优选60%。
(4)菌液的扩培和有机菌肥制备方法
复配好的菌液在20℃~40℃下进行发酵,优选30℃。发酵时间为48小时~96小时,优选72小时,即获得液态有机菌肥。

选择巨大芽孢杆菌、解脂假丝酵母和假单胞菌。利用牛肉膏3g、蛋白胨10g、NaCI5g、琼脂、水1000mL,制备假单胞菌斜面培养基;利用葡萄糖10g、NaCI0.3g、MgSO40.3g、MnSO40.03g、CaCO31.0g、(NH4)2SO40.5g、KCl0.3g、FeSO40.03g、卵磷脂0.2g、酵母粉0.5g、琼脂、水1000mL,制备巨大芽孢杆菌斜面培养基;利用黄豆芽100g、葡萄糖20g、水1000mL、琼脂,将黄豆芽洗净,加水煮沸30min。用纱布过滤,滤液中加入琼脂,加热溶解后放入糖,搅拌使它溶解,补足水分到1000mL,制备热带假丝酵母斜面培养基。将假单胞菌、解脂假丝酵母、巨大芽孢杆菌分别转入新鲜的斜面培养基,在30℃培养24h,得到活化的菌种。将活化好的菌种分别放入去除琼脂的培养基,接种量均为2%,均在30℃培养18小时,摇床转速均为180转/分。获得活化后的菌液。将活化好的假单胞菌、巨大芽孢杆菌、解脂假丝酵母菌液以1∶1∶1的比例,添加到餐厨垃圾废液中,接种量为2%。然后,加入60%的畜禽粪便辅料,制得扩培菌液。复配好的菌液在30℃下发酵72小时,即获得液态有机菌肥。发酵产物中假单胞菌、巨大芽孢杆菌、解脂假丝酵母活菌总数达108个/mL。另外,本发明所用的菌种均为农业部关于微生物肥料生物安全通用技术准则(NY11092006)附录中规定使用的生产菌种。
Claims (5)
1.一种利用餐厨废液生产液态有机菌肥的方法,其特征在于:
整个方法包括但不局限于如下几部分:菌种选择、菌种培育和活化、菌液复配,菌种扩培和液态有机菌肥制备,整个生产过程没有废气、废液和废渣产生,环境风险低,且废水中有机物再利用,不仅节省废水处理费用,且创造价值,环境效益和经济效益显著,所说的菌种,包括但不限于芽胞菌和酵母菌,所说的芽胞菌为巨大芽孢杆菌或地衣芽胞杆菌、或枯草芽胞杆菌,优选巨大芽胞杆菌,所说的酵母菌为或乳酸酵母菌,优选解脂假丝酵母,另外,所说的混合菌群中还包括假单胞菌。
2.权利要求1中所说的菌种培育和活化方法为,利用牛肉膏3g、蛋白胨10g、NaCl5g、琼脂、水1000mL,制备假单胞菌斜面培养基;利用葡萄糖10g、NaCI0.3g、MgSO40.3g、MnSO40.03g、CaCO31.0g、(NH4)2SO40.5g、KCl0.3g、FeSO40.03g、卵磷脂0.2g、酵母粉0.5g、琼脂、水1000mL,制备巨大芽孢杆菌斜面培养基;利用黄豆芽100g、葡萄糖20g、水1000mL、琼脂,将黄豆芽洗净,加水煮沸30min,用纱布过滤,滤液中加入琼脂,加热溶解后放入糖,搅拌使它溶解,补足水分到1000mL,制备热带假丝酵母斜面培养基。
3.将权利要求2所说的菌种转入新鲜的斜面培养基,在30℃培养24h,得到活化的菌种,活化好的菌种分别放入摇瓶培养基,接种量均为1%~3%,优选2%,均在20℃~40℃下培养10~30小时,优选地,均在30℃培养18小时,摇床转速均为100转/分~300转/分,优选180转/分,获得活化后的菌液。
4.权利要求1中所说的菌液复配方法为,将权利要求3中活化好的假单胞菌、巨大芽孢杆菌、解脂假丝酵母菌液以1∶1∶1的比例,添加到餐厨垃圾废液中,接种量为1%~3%,优选2%,然后,加入畜禽粪便辅料,加入量为40%~80%,优选60%,制得扩培菌液。
5.权利要求1所说的菌种扩培和菌肥制备方法为,将复配好的菌液在30℃下发酵72小时,进行菌种扩培,即获得液态有机菌肥。
