技术领域
本发明属于复合生物材料领域,尤其涉及一种再生纤维素伤口敷料及其制备方法。
背景技术
当皮肤损伤后,需要使用皮肤修复材料作为受损皮肤的临时替代物来保护伤口,防止伤口感染和重度脱水,为伤口愈合提供相对湿润的环境,促进组织再生和创面愈合。在众多皮肤修复材料中,自体皮是一种最佳的修复材料,因其不会产生免疫反应且含有各种促进修复愈合的组织细胞和活性生长因子。由于自体皮的来源有限,只能用作小范围的皮肤损伤,所以当大面积皮肤损伤时,需要借助其他皮肤修复材料,如医用伤口敷料、皮肤支架材料、人工皮肤等作为损伤皮肤的临时替代物来实现损伤修复,促进新生组织的再生和伤口的快速愈合。
随着组织工程学的发展,皮肤修复材料经历了从传统敷料到新型敷料再到皮肤组织工程支架材料的发展历程,利用天然活性高分子或人工合成高分子制成的皮肤组织工程支架材料已在临床上得到广泛的应用。尽管各种伤口敷料以其不同的特点被应用于临床不同的创面,至今为止还没有一种理想的敷料能够满足各种创面修复愈合的要求,现有技术中的伤口敷料的组织修复能力还有待进一步提升。
发明内容
为了解决现有技术中存在的问题,本发明提供了一种可以用于各种创面修复愈合的、具有优良的组织修复能力的再生纤维素伤口敷料。
本发明采用以下技术方案:
一种再生纤维素伤口敷料,包括以下重量份数的原料:棉短绒60-70份、壳聚糖溶液5-10份、藻酸盐溶液5-10份、蚕丝蛋白溶液5-10份;
所述的壳聚糖溶液采用以下方法制备:称取0.2g壳聚糖,溶于40ml去离子水中,配制质量体积比为20%的壳聚糖溶液,另加入0.32g氯化钠晶体及0.5ml冰醋酸,调节pH至5.0混合搅拌均匀得壳聚糖溶液;
所述的藻酸盐溶液采用以下方法制备:称取0.2g藻酸盐,溶于40ml去离子水中,配制质量体积比为20%的藻酸盐溶液,另加入0.32g氯化钠晶体,搅拌至充分溶解得藻酸盐溶液。
优选的,所述的蚕丝蛋白溶液采用以下方法制备:将2L去离子水烧热至沸腾,加入4g碳酸钠及5g家蚕生丝,煮沸30min后,用去离子水清洗干净并烘干,再用500ml、9.3mol/L的LiBr溶液溶解,待完全溶解后将溶液装入透析袋,去离子水透析72h,9000r/min离心20min,重复两次,得蚕丝蛋白溶液。
优选的,所述的再生纤维素伤口敷料,还包括中药提取物3-5份。
优选的,所述的中药提取物为淫羊藿提取物、骨碎补提取物、川芎提取物、巴戟天提取物、土鳖虫提取物、丹参提取物中的一种或一种以上的组合。
优选的,所述的中药提取物采用以下方法制备:称取中药20-25g,粉碎成粗粉,加入其重量10-15倍的蒸馏水浸泡1-2h,回流提取1-1.5h,抽滤得滤液1;向滤渣中加入8-10倍滤渣重量的蒸馏水,回流提取1-1.5h,抽滤得滤液2;再向滤渣中加入8-10倍滤渣重量的蒸馏水,回流提取1-1.5h,抽滤得滤液3;将滤液1、滤液2、滤液3合并,在温度为40-50℃、真空度为0.05-0.08Pa条件下减压浓缩至中药重量的4-6倍,用三层纱布过滤,即得中药提取液。

本发明的有益效果在于:
1)纤维素作为一种生物材料,具有良好的生物适应性和生物活性,能作用于创面修复的多个环节,促进创面的快速愈合。以纤维素为原料制成的伤口敷料柔软透明、舒适;有良好的通透性,能为创面提供湿润、微酸的修复环境;能调节创面氧张力,促进毛细血管形成,从而加速伤口的愈合。
2)蚕丝蛋白生物相容性良好,降解速度缓慢、且降解产物对组织具有低免疫原性,对皮肤、牙周等组织有营养和修复能力,在体内或体外培养细胞时,多孔结构的丝素蛋白能适应细胞的黏附、铺展和增殖,对组织的修复和重建极为有利,是一种性能优异的生物材料。
3)壳聚糖和藻酸盐均被视为细胞支架材料并广泛运用在组织工程领域。壳聚糖是一种来源于海洋蟹类生物的天然多糖,生物相容性好,可降解,无毒性而且有其对耐甲氧西林金黄色葡萄球菌和大肠杆菌等均具有一定的抗菌效果。藻酸盐除了良好生物相容性,可降解性及无毒性之外,还对细胞的粘附、迁移有促进作用。
4)本发明制得的再生纤维素伤口敷料可以用于各种创面的修复愈合、且具有优良的组织修复能力。
具体实施方式
实施例1
一种再生纤维素伤口敷料,包括以下重量份数的原料:棉短绒60份、壳聚糖溶液5份、藻酸盐溶液5份、蚕丝蛋白溶液5份。
所述的壳聚糖溶液采用以下方法制备:称取0.2g壳聚糖,溶于40ml去离子水中,配制质量体积比为20%的壳聚糖溶液,另加入0.32g氯化钠晶体及0.5ml冰醋酸,调节pH至5.0混合搅拌均匀得壳聚糖溶液;
所述的藻酸盐溶液采用以下方法制备:称取0.2g藻酸盐,溶于40ml去离子水中,配制质量体积比为20%的藻酸盐溶液,另加入0.32g氯化钠晶体,搅拌至充分溶解得藻酸盐溶液。
所述的蚕丝蛋白溶液采用以下方法制备:将2L去离子水烧热至沸腾,加入4g碳酸钠及5g家蚕生丝,煮沸30min后,用去离子水清洗干净并烘干,再用500ml、9.3mol/L的LiBr溶液溶解,待完全溶解后将溶液装入透析袋,去离子水透析72h,9000r/min离心20min,重复两次,得蚕丝蛋白溶液。
一种再生纤维素伤口敷料的制备方法,包括以下步骤:
步骤一:称取棉短绒,将其溶解在氢氧化锂/尿素水溶液中,配制成质量浓度4%的纤维素溶液,所述的氢氧化锂/尿素水溶液采用氢氧化锂8.76g、尿素12g及水79.24g配制而成,充分搅拌均匀、过滤、静置24h,于20kV条件下静电纺丝得纤维膜;
步骤二:将壳聚糖溶液、藻酸盐溶液、蚕丝蛋白溶液混合均匀,涂抹于纤维膜表面,即得敷料。
实施例2
一种再生纤维素伤口敷料,包括以下重量份数的原料:棉短绒70份、壳聚糖溶液10份、藻酸盐溶液10份、蚕丝蛋白溶液10份、中药提取物3份。
所述的壳聚糖溶液采用以下方法制备:称取0.2g壳聚糖,溶于40ml去离子水中,配制质量体积比为20%的壳聚糖溶液,另加入0.32g氯化钠晶体及0.5ml冰醋酸,调节pH至5.0混合搅拌均匀得壳聚糖溶液;
所述的藻酸盐溶液采用以下方法制备:称取0.2g藻酸盐,溶于40ml去离子水中,配制质量体积比为20%的藻酸盐溶液,另加入0.32g氯化钠晶体,搅拌至充分溶解得藻酸盐溶液。
所述的蚕丝蛋白溶液采用以下方法制备:将2L去离子水烧热至沸腾,加入4g碳酸钠及5g家蚕生丝,煮沸30min后,用去离子水清洗干净并烘干,再用500ml、9.3mol/L的LiBr溶液溶解,待完全溶解后将溶液装入透析袋,去离子水透析72h,9000r/min离心20min,重复两次,得蚕丝蛋白溶液。
所述的中药提取物为淫羊藿提取物、骨碎补提取物、川芎提取物、巴戟天提取物、土鳖虫提取物、丹参提取物按重量比1:1:1:1:1:1的混合物。
所述的中药提取物采用以下方法制备:称取中药20-25g,粉碎成粗粉,加入其重量10-15倍的蒸馏水浸泡1-2h,回流提取1-1.5h,抽滤得滤液1;向滤渣中加入8-10倍滤渣重量的蒸馏水,回流提取1-1.5h,抽滤得滤液2;再向滤渣中加入8-10倍滤渣重量的蒸馏水,回流提取1-1.5h,抽滤得滤液3;将滤液1、滤液2、滤液3合并,在温度为40-50℃、真空度为0.05-0.08Pa条件下减压浓缩至中药重量的4-6倍,用三层纱布过滤,即得中药提取液。
一种再生纤维素伤口敷料的制备方法,包括以下步骤:
步骤一:称取棉短绒,将其溶解在氢氧化锂/尿素水溶液中,配制成质量浓度4%的纤维素溶液,所述的氢氧化锂/尿素水溶液采用氢氧化锂8.76g、尿素12g及水79.24g配制而成,充分搅拌均匀、过滤、静置24h,于20kV条件下静电纺丝得纤维膜;
步骤二:将壳聚糖溶液、藻酸盐溶液、蚕丝蛋白溶液及中药提取物混合均匀,涂抹于纤维膜表面,即得敷料。
选择12只小白鼠,随机分成3组,每组4只,在每只白鼠的脊柱上制造出1个2×2cm的全层皮肤缺损的创面模型,又分成空白对照组和两个实验组(实施例1、实施例2)。在试验后每天观察创面愈合的大体情况和愈合时间;同时在创面中心取创面组织,福尔马林溶液固定,石蜡包埋,软组织切片,行HE染色,在显微镜下观察空白对照组和实验组皮肤全层缺损创面的愈合情况。第7、14、21、28、35天分别对实验组和空白对照组的创面照相采图,用计算机图像分析软件计算各组的创面愈合面积,测定各组的创面愈合百分率,按公式创面愈合率(%)=[(创面原始面积-创面未愈合面积)/创面原始面积]×100%,结果见下表1。

结果:在对比空白对照组和敷用敷料的实验组的愈合时间及创面愈合率可知:创面一般情况实验组明显好于空白对照组,实验组所用的敷料对皮肤缺损具有良好的促愈合作用。

1.一种再生纤维素伤口敷料,其特征在于,包括以下重量份数的原料:棉短绒60-70份、壳聚糖溶液5-10份、藻酸盐溶液5-10份、蚕丝蛋白溶液5-10份;
所述的壳聚糖溶液采用以下方法制备:称取0.2g壳聚糖,溶于40ml去离子水中,配制质量体积比为20%的壳聚糖溶液,另加入0.32g氯化钠晶体及0.5ml冰醋酸,调节pH至5.0混合搅拌均匀得壳聚糖溶液;
所述的藻酸盐溶液采用以下方法制备:称取0.2g藻酸盐,溶于40ml去离子水中,配制质量体积比为20%的藻酸盐溶液,另加入0.32g氯化钠晶体,搅拌至充分溶解得藻酸盐溶液。
2.根据权利要求1所述的一种再生纤维素伤口敷料,其特征在于,所述的蚕丝蛋白溶液采用以下方法制备:将2L去离子水烧热至沸腾,加入4g碳酸钠及5g家蚕生丝,煮沸30min后,用去离子水清洗干净并烘干,再用500ml、9.3mol/L的LiBr溶液溶解,待完全溶解后将溶液装入透析袋,去离子水透析72h,9000r/min离心20min,重复两次,得蚕丝蛋白溶液。
3.根据权利要求1所述的一种再生纤维素伤口敷料,其特征在于,包括以下重量份数的原料:棉短绒60份、壳聚糖溶液5份、藻酸盐溶液5份、蚕丝蛋白溶液5份。
4.根据权利要求1所述的一种再生纤维素伤口敷料,其特征在于,包括以下重量份数的原料:棉短绒70份、壳聚糖溶液10份、藻酸盐溶液10份、蚕丝蛋白溶液10份。
5.根据权利要求1所述的一种再生纤维素伤口敷料,其特征在于:还包括中药提取物3-5份。
6.根据权利要求5所述的一种再生纤维素伤口敷料,其特征在于:所述的中药提取物为淫羊藿提取物、骨碎补提取物、川芎提取物、巴戟天提取物、土鳖虫提取物、丹参提取物中的一种或一种以上的组合。
7.根据权利要求5所述的一种再生纤维素伤口敷料,其特征在于,所述的中药提取物采用以下方法制备:称取中药20-25g,粉碎成粗粉,加入其重量10-15倍的蒸馏水浸泡1-2h,回流提取1-1.5h,抽滤得滤液1;向滤渣中加入8-10倍滤渣重量的蒸馏水,回流提取1-1.5h,抽滤得滤液2;再向滤渣中加入8-10倍滤渣重量的蒸馏水,回流提取1-1.5h,抽滤得滤液3;将滤液1、滤液2、滤液3合并,在温度为40-50℃、真空度为0.05-0.08Pa条件下减压浓缩至中药重量的4-6倍,用三层纱布过滤,即得中药提取液。
8.一种如权利要求1-4任一项所述的再生纤维素伤口敷料的制备方法,其特征在于,包括以下步骤:
步骤一:称取棉短绒,将其溶解在氢氧化锂/尿素水溶液中,配制成质量浓度4%的纤维素溶液,所述的氢氧化锂/尿素水溶液采用氢氧化锂8.76g、尿素12g及水79.24g配制而成,充分搅拌均匀、过滤、静置24h,于20kV条件下静电纺丝得纤维膜;
步骤二:将壳聚糖溶液、藻酸盐溶液、蚕丝蛋白溶液混合均匀,涂抹于纤维膜表面,即得敷料。
9.一种如权利要求5-7任一项所述的再生纤维素伤口敷料的制备方法,其特征在于,包括以下步骤:
步骤一:称取棉短绒,将其溶解在氢氧化锂/尿素水溶液中,配制成质量浓度4%的纤维素溶液,所述的氢氧化锂/尿素水溶液采用氢氧化锂8.76g、尿素12g及水79.24g配制而成,充分搅拌均匀、过滤、静置24h,于20kV条件下静电纺丝得纤维膜;
步骤二:将壳聚糖溶液、藻酸盐溶液、蚕丝蛋白溶液及中药提取物混合均匀,涂抹于纤维膜表面,即得敷料。
