Abstract
本发明公开了一株经过常压室温等离子体诱变的具有芽孢高产的解淀粉芽孢杆菌(Bacillus amyloliquefaciens)KN‑527,其保藏编号为CCTCC NO:M2015709。该菌株发酵芽孢数比诱变前提高161%,芽孢形成率高,耐热性好,对甜瓜白粉病有显著的抗病作用,防效达到95%以上,可应用于黄瓜、西葫芦、南瓜、苦瓜和西瓜等瓜类作物。通过甜瓜田间试验验证,用该菌株生产的微生物菌剂加上常规施肥后,植株生长趋旺,生发侧枝能力强,叶色鲜绿。

技术领域
本发明属于微生物技术领域,具体涉及一株解淀粉芽孢杆菌(Bacillusamyloliquefaciens),本发明还涉及一种抗瓜类作物白粉病的微生物菌剂。
背景技术
白粉病是瓜类作物发生较为普遍的一种病害,白粉病菌有3个属6个种,瓜单囊壳菌和二孢白粉菌是主要病原,而瓜单囊壳菌又有报道发现多个生理小种。白粉病主要危害叶片,也侵染叶柄、瓜蔓和瓜。叶片发病初在叶面上产生近圆形白色小霉点(即病原菌菌丝和分生孢子),后扩大成霉斑,条件适宜,霉斑迅速扩大,彼此连片,白粉状物会布满整个叶片,致叶片变脆,后期病斑上霉状物变灰白色,其上长出黑色小粒点,即病菌的闭囊壳。发病重的,叶片出现不规则黄褐病斑,病斑连片致整个叶片褐色干枯,但不脱落。叶柄、瓜蔓和瓜染病,亦产生白色粉斑,发展后,病部布满白粉。白粉病可减少植株冠层,降低单株座果率,使果实变小,从而导致减产失去市场价值,给栽培者造成严重的经济损失。
我国是农业大国,农药使用品种多,用量大,其中70~80%的农药直接散落在环境中。应用化学农药后,不仅植物本身吸附大量农药,而且通过渗透作用进入植物内部,造成严重残留,进而对农产品产生污染。生物从环境介质或食物中不断吸收低剂量的有毒物质,并逐渐在体内积累浓缩,在整个生态系统中,农药不断地通过生物富集与食物链的传递,逐级浓缩、逐级放大,人类处于食物链顶端,受害最为严重。化学农药的持续使用,造成土壤肥力下降,板结等,会引起土壤微生物体系的剧烈改变,进而影响土壤中酶的活性,营养物质的转化,改变农业人生态系统营养循环的效率和速度,使土地持续生产力下降。针对病害的生理特点,适时对症下药,选择高效、安全、生物活性高的生物农药,就显得尤为重要。
目前白粉病多采用化学防治方法,但由于白粉病病原菌分化快,亚种多,化学试剂很难长期有效地保证作物的无公害生产,因此,新的安全、高效、经济的防控手段成为热门。近年来已经研究报道了大量的拮抗微生物,包括细菌、酵母菌、放线菌等。然而普通细菌、酵母菌及放线菌抗逆性较差,生长环境要求高,无法批量推广应用。芽孢作为整个生物界中抗逆性最强的生命体,在抗热,抗化学药物和抗辐射等方面,十分突出。解淀粉芽孢杆菌作为一种好氧产芽孢的革兰氏阳性杆状细菌,目前尚无报道对人畜致病,其营养要求简单、易于存活、定植与繁殖,稳定性好,不仅能提高作物的产量,增加抗逆性,而且还大大地增加了土壤肥力,当其在土壤中繁殖为优势菌群后,还可以达到抑制病原菌的效果,很适合作为生防菌和生物肥料大量生产应用。
发明内容
本发明的目的是为了弥补解淀粉芽孢杆菌在瓜类作物白粉病防治应用领域的空白,提供一株具有芽孢高产的解淀粉芽孢杆菌及其微生物菌剂。
本发明以解淀粉芽孢杆菌为出发菌株,经过常压室温等离子体诱变,获得一株具有芽孢高产的解淀粉芽孢杆菌(Bacillus amyloliquefaciens)KN-527,已于2015年11月26日在中国武汉武汉大学内的中国典型培养物保藏中心(CCTCC)保藏,其保藏编号CCTCC NO:M2015709。
本发明提供了一种抗瓜类作物白粉病的微生物菌剂,包含上述解淀粉芽孢杆菌KN-527。
所述微生物菌剂能用作促进瓜类作物生长的生物肥料。
本发明的有益效果:
本发明的解淀粉芽孢杆菌经过离子体诱变筛选后,产芽孢数可以达到1.19E+10CFU/克,比诱变前的出发菌株7.37E+09CFU/克,提高了161%。田间试验结果表明,施药14天后,KN-527对甜瓜白粉病的防效达到95%以上,明显优于化学农药多菌灵。所述菌株能有效预防瓜类白粉病的发生,实验组黄瓜、南瓜、西瓜的病情指数远远低于对照组,0.1亿cfu/g解淀粉芽孢杆菌菌剂稀释3000倍菌液(施药14天)能有效防治瓜类白粉病,病指控制在6%以下,防效达到90%以上;施药14天后,对照组病指均达到了97%以上,而对草莓、小麦、辣椒等非瓜类作物的白粉病防效较低。
具体实施方式
以下通过实施例对本发明进行详细说明。
实施例1
(1)解淀粉芽孢杆菌出发菌株的纯化培养
挑取出发菌株解淀粉芽孢杆菌KN-52(从武汉蔬菜种植基地土壤中自然筛选而得)单菌落,接入由蛋白胨1%、酵母浸粉0.5%,氯化钠1%,pH7.2-7.5组成的简单培养基中,在温度32℃与180rpm的条件下恒温摇床上培养16-20小时,取10ml发酵液,4℃,6000r/min条件下离心,收集菌浆待用;
(2)初筛:常温常压等离子诱变法
取步骤(1)制备的菌体用10ml无菌生理盐水溶解震荡均匀,取10μl涂于在已灭菌的金属载片上,将载片置于载物台上,调整载物台高度,设定好处理时间后对解淀粉芽孢杆菌进行照射。氦气为载体,照射功率150W,工作空气流量10SLPM,照射距离2mm,照射时间60~240s。照射处理后将载片转移到装有1ml无菌生理盐水的EP管中,振荡1~2min,使附着在载片上的菌体洗脱到生理盐水中,形成新的敏感菌悬液。将敏感菌悬液稀释至10-4、10-5、10-6共3个梯度。
(3)甜瓜白粉病病原菌的收集,制备压力筛选平板
田间采集严重感染白粉病的甜瓜叶片,叶片表面喷洒无菌水,4℃冰箱过夜。在超净工作台中,用洗耳球吹入水琼脂平板,放入培养箱,20摄氏度培养7d。
配置压力筛选平板固体培养基,蔗糖5%-10%、酵母抽提物0.1-0.5%、氯化钠0.02%-0.05%、硫酸镁0.02%-0.05%、碳酸钙0.05%-0.2%、琼脂粉2%,pH7.2-7.5组成,灭菌后与45度左右加入2%白粉病病原菌;
将稀释的敏感菌悬液的每个梯度涂布3块压力筛选平板。将涂布后的平板放置培养箱中30℃进行培养16h。挑取抑菌圈透明带大的单菌落接入斜面培养基,在30℃恒温培养箱中培养全部形成芽孢后收入4℃冰箱保存备用;
所述的斜面培养基是由蛋白胨1%、酵母浸粉0.5%、氯化钠1%、琼脂粉1.5%-2%,pH7.2-7.4组成;
(4)抗性菌株高产株的选择
取步骤(3)中斜面菌种,用25ml无菌水洗下装入无菌带玻璃珠的100ml三角瓶中,震荡混合均匀制成菌悬液后放入70℃水浴锅中水浴30min,取出用冷水立即降至室温后接入发酵种子摇瓶32℃与180rpm的条件下培养16-24h后转入发酵摇瓶,同样在32℃与180rpm的条件下培养40-48h;取摇瓶发酵液,常规涂片,用碱性石炭酸复红染色,用奥林巴斯显微镜油镜(16×100倍)观察发酵后芽孢形成率。
选择具有典型解淀粉芽孢杆菌芽孢形态,芽孢形成率高的菌株。根据上述结果,综合判断诱变菌株的抗病能力和芽孢形成率,得到一株芽孢形成率高的菌株KN-527。
经发酵后CFU对比,KN-527芽孢数可达1.19E+10,比诱变前的出发菌株CFU 7.37E+09,提高161%,见表1。

实施例2解淀粉芽孢杆菌KN-527对甜瓜白粉病的田间抑制效果验证。
1.材料
供试药剂:解淀粉芽孢杆菌KN-527、化学农药选用传统白粉病防治药剂。
供试作物:甜瓜
防治对象:白粉病
试验共设置3个处理:
(1)对照组:清水处理;
(2)处理组:0.1亿CFU/g所述解淀粉芽孢杆菌稀释1000、2000和3000倍
(3)50%多菌灵可湿性粉剂;
以上处理在科研基地甜瓜大棚内进行。供试甜瓜施药前摘除瓜的侧蔓,于发病初期第一次施药,隔7d后再喷药1次,将药液均匀喷洒在植株叶片上。
2.方法与结果
定点定株调查,每片区选10株,每株视生长情况调查5片叶长5cm以上的叶片,药前调查病情级数,药后7d、14d各调查一次,按叶片发病程度分级计数。
分级标准:按0~9级病情分级指数标准
0级:叶健,无病斑
1级:病斑占全叶叶面积的10%以下;
3级:病斑占全叶叶面积的11%~25%;
5级:病斑占全叶叶面积的16%~40%;
7级:病斑占全叶叶面积的41%~65%;
9级:病斑占全叶叶面积的65%以上;
药效计算方法:
病情指数(%)=∑(各级病叶数×相应病级数)×100%/调查总叶数×最高病级数;
病指增长率=(药后病指-药前病指)/药前病指×100%;
防效=[1-(处理区药后病指×对照区药前病指)/(处理区药前病指×对照区药后病指)]×100%。

从表2的数据中可以看出,0.1亿cfu/g解淀粉芽孢杆菌菌剂稀释1000、2000和3000倍菌液(施药14天)能有效防治甜瓜白粉病,病指控制在5%以下,防效达到95%以上;施药14天后,对照组病指达到了98.2%,50%多菌灵可湿性粉剂对甜瓜白粉病的防效为65.4%,而稀释了3000倍的菌液防治甜瓜白粉病的防效就可以达到95.9%。
实施例3解淀粉芽孢杆菌KN-527作为生物肥料对甜瓜的田间效果验证。
供试药剂:解淀粉芽孢杆菌KN-527
供试作物:甜瓜
营养液:以色列Polyfood全元素营养液
试验方法:
将定植一周后的甜瓜分为两组,A组施用营养液+清水,B组施用营养液+解淀粉芽孢杆菌可湿性粉剂。
在抽蔓期、座瓜期、膨瓜期对植株生长情况、果实生长情况及产量、品质进行考察。

试验结果表明,解淀粉芽孢杆菌KN-527可作为生物肥料,对甜瓜植株生长具有明显促进作用,生发侧枝能力增强,植株粗壮。
实施例4解淀粉芽孢杆菌KN-527对瓜类白粉病和其他作物白粉病的田间抑制效果验证。
1.材料
供试药剂:解淀粉芽孢杆菌CCTCC NO:M2015709
供试作物:黄瓜、南瓜、西瓜、草莓、小麦、辣椒
防治对象:白粉病
试验共设置2个处理:
(1)处理组:清水处理(CK);
(2)0.1亿CFU/g所述解淀粉芽孢杆菌3000倍液
以上处理在科研基地各大棚内进行。供试作物于发病初期第一次施药,隔7d后再喷药1次,将药液均匀喷洒在植株叶片上。
2.方法与结果
定点定株调查,每片区选20株,每株视生长情况调查5片叶长5cm以上的叶片,药前调查病情级数,药后7d、14d各调查一次,按叶片发病程度分级计数。
分级标准:按0~9级病情分级指数标准
0级:叶健,无病斑
1级:病斑占全叶叶面积的10%以下;
3级:病斑占全叶叶面积的11%~25%;
5级:病斑占全叶叶面积的16%~40%;
7级:病斑占全叶叶面积的41%~65%;
9级:病斑占全叶叶面积的65%以上;
药效计算方法:

防效=[1-(处理区药后病指×对照区药前病指)/(处理区药前病指×对照区药后病指)]×100%。

白粉病菌有3个属6个种,瓜单囊壳菌和二孢白粉菌是主要病原。黄瓜、南瓜、西瓜等瓜类白粉病的病原菌主要是瓜单囊壳菌,而草莓、小麦、辣椒白粉病的病原菌主要是羽衣草单囊壳菌、禾本科布氏白粉菌、鞑靼内丝白粉病。从表4的数据中可以看出,0.1亿cfu/g解淀粉芽孢杆菌菌剂稀释3000倍菌液(施药14天)能有效防治瓜类白粉病,病指控制在6%以下,防效达到90%以上;施药14天后,对照组病指均达到了97%以上,对草莓、小麦、辣椒白粉病的防效分别为81.3%、50.2%和63.8%,防治效果不理想。

2.一种抗瓜类作物白粉病的微生物菌剂,其特征在于:所述微生物菌剂的活性成分为权利要求1所述的解淀粉芽孢杆菌。
