本发明涉及一种稳定性好、效果突出的肉桂醛衍生物,尤其涉及该类衍生物在日用化学品、药品、食品以及饲料中的应用。
背景技术
肉桂醛(Cinnamaldehyde)是一种醛类有机化合物,为黄色黏稠状液体,大量存在于肉桂等植物体内。自然界中天然存在的肉桂醛均为反式结构,该分子为一个丙烯醛上连接上一个苯基,因此可被认为是一种丙烯醛衍生物,是传统中药肉桂挥发油的主要活性成分。肉桂为樟科常绿乔木植物肉桂的干燥树皮,其主要分布在广东、广西、云南和福建等,其中两广的肉桂油生产量约占世界产量的80%左右,在四川、贵州、湖南、江西、浙江也有少量的栽培。肉桂醛还天然存在于风信子、玫瑰、广蕾香等植物精油中。
肉桂醛有顺式和反式两种异构体,现商用的桂醛无论是天然的或者合成的桂醛,都是反式异构体。

肉桂醛的抑菌、防腐等性能突出,因而其应用非常广泛。谭龙飞、谢小梅等分别对其杀菌消毒防腐性能进行研究,特别是对真菌具有显著的抑制活性,对大肠杆菌、枯草杆菌、金黄色葡萄球菌、白色葡萄球菌、伤寒和副伤寒甲杆菌、肺炎球菌、产气杆菌、变形杆菌、炭疽杆菌、肠炎沙门氏菌霍乱弧菌等都具有很好的抑制活性,且对革兰氏阳性菌杀菌效果显著,可用于治疗多种因细菌感染而引起的疾病。
据报道,肉桂醛具有抗溃疡、加强胃肠道运动,其作用机制是由于溃疡活性因素的抑制与防御因素的加强,以及抑制胃粘膜电位降低和对粘膜保护作用所致。除此之外,肉桂醛能降低胰酶活性。肉桂醛系芳香性健胃驱风剂,对肠胃有缓和的刺激作用,可促进唾液及胃液分泌,增强消化功能,解除胃肠平滑肌痉挛,缓解肠道痉挛性疼痛。
肉桂醛被美国FDA公认为安全、无毒的物质(21CFR 182.60),美国香味料和萃取物制造者协会(FEMA)认为是安全无毒的食品香料成分(FEMA,1965),食品添加剂联合专家委员会(JECFA)也认为肉桂醛作为食品添加剂是安全的(JECFA No.656)。
农林专家研究发现,肉桂醛以其独有的抑菌和杀菌作用,能在饲料添加剂中对动物营养物质消化产生积极作用。研究表明,肉桂醛能增加动物胆汁的分泌,日粮中添加肉桂醛已被证明可以刺激肠粘膜和胰腺消化酶的活性。有学者在肉鸡日粮中添加肉桂醛,能改善饲料适口性,提高饲料消化利用率,促进和改善动物生长性能以及动物产品质量,另外,能有效防止饲料变质腐败,而延长产品的保质时间。
肉桂醛可以用于食品起防腐保鲜作用,黑龙江八一农垦大学食品学院用肉桂醛对肉制品的防腐作用进行平行比较研究后发现,肉桂醛对不同菌种均有抑制作用,对黄曲霉的抑制作用尤其明显,相同剂量下,使用肉桂醛比山梨酸钾使肉制品保鲜期延长1周左右,更为重要的是,肉桂醛本身作为一种食品用香料,对人体完全无毒无害,国标GB/T 2076-2011规定的允许使用其作为食品添加剂,可用于制备肉类、调味品、口腔护理用品、口香糖、糖果用香精。
肉桂醛性能较好、应用领域广泛,但由于其结构原因,决定其具有易挥发、易氧化、气味刺激、变色、水溶性差等特点,严重影响其应用的广度与深度,集中表现在如下几点:
1. 在空气中,极易氧化成肉桂酸而变质;
2. 具有黄色或浅绿黄色,影响其在配方中的应用,尤其是无色透明体系;
3. 具有强烈刺激性气味,具有辣感并有灼烧感;
4. 具有溶解性差,难溶于水和甘油,严重影响配伍性与利用度;
5. 具有极易挥发性大,效果持续性差。
为克服以上存在的缺点,科研人员对肉桂醛进行衍生物与包裹处理,以期提高其对光、热、空气等稳定性和降低刺激性气味;通过对肉桂醛的包裹,提高稳定性与溶解度;通过形成纳米乳液与微胶囊技术,提高储存稳定性。
张园园等对肉桂醛进行衍生物的合成,以肉桂醛为原料合成了无刺激性气味的邻硝基肉桂醛、α-溴代肉桂醛和含有亲水性的羟基和磺酸基官能团的肉桂醛羟基磺酸钠,显著降低了气味问题,但是由于醛基依然存在,其稳定性并没有得到提升,并且引进了硝基、卤素、磺酸基等基团,而这些基团存在刺激性大、毒性高、副产物多等特点,因此该改进并没有较大的实际应用价值。
钱亮亮等通过β-环糊精对肉桂醛进行包合来提高其稳定性,该包合物应用于产品后,缓慢释放肉桂醛,而达到在食品表面发挥抗菌的作用,试验证明具有良好的防霉功效,可提高其安全性并延长货架期。从某种程度上说,提高肉桂醛的稳定性与溶解度,有较大的进步,但是由于大分子的介入,与体系的相容性较差,另外随着肉桂醛的释放与消耗,效果会逐步消失,因此其保存周期还不够理想,仍有较大的改进空间。
谢帆等通过微胶囊技术,将肉桂醛包裹于微胶囊中,其稳定性得到一定的提高,并起到良好的控制释放作用,结果表明:在室温条件储藏过程中,肉桂醛的保留率会随着时间延长缓慢下降,3个月后其保留率仍达到97%,表明微胶囊肉桂醛在室温下贮存稳定。该方法与环糊精处理方法相似,微胶囊对温度、配方体系存在稳定性问题,影响其应用范围。
以上研究,相对直接使用肉桂醛已有较大的改进,可以某种程度上解决某一个或几个问题,但均不能同时解决易氧化、水溶性、挥发性、刺激性以及色泽等问题。
发明内容
本发明的目的在于提供一种稳定性好、效果突出的肉桂醛衍生物;
本发明的另一目的在于提供该类衍生物在食品中的应用;
本发明的另一目的在于提供该类衍生物在药品中的应用;
本发明还提供一种用于提供该类衍生物在日用化学品中的应用;
本发明还有一目的是提供该类衍生物在饲料的应用。
本发明采用的技术方案如下:
一种肉桂醛衍生物,该衍生物具有如式I所示的结构:

其中X可以为-H、烷基、-OH、-COOH、-CH2COOH、-CH2OH、ph-CH=CH-CH=N-;
其中Y可以为-H、烷基、-OH、-COOH、-CH2COOH、-CH2OH、ph-CH=CH-CH=N-;
其中Z可以为-H、烷基、-OH、-COOH、-CH2COOH、-CH2OH、ph-CH=CH-CH=N-;
其中n=2~15的任意整数。
所述的烷基为所述的烷基可以为C3~C16的直链烷基或取代苯基,可以为甲基、乙基、丙基或丁基。
本发明肉桂醛衍生物的制备方法,参照现有技术与方法制得:
将含有氨基的六元氧杂环与肉桂醛在合适的溶剂中,缩合,使用合成方式可以为:固相研磨法、微波催化法、超声合成法;使用的纯化分离方法,包括柱层析法与重结晶法,获得目标化合物。

本发明所产生的有益效果:
本发明衍生物,除了保留肉桂醛本身的特性,如抑菌防腐、抗病毒、抗肿瘤、保鲜等性能,还具有肉桂醛不具有的优点与特性:
1)优良的稳定性能,对氧气、光、热、重金属等耐受性好;
2)无挥发性,不会因为挥发而其活性成分流失,使得活性降低;
3)低刺激性与致敏性;
4)水溶性好,不会因为水溶性问题而影响产品的性能;
5)配伍性佳,便于在配方体系中使用;
6)衍生物为白色固体,几乎无味;
7)该衍生物防腐性能较肉桂醛有所提升。
由于该系列化合物具有如上这些特点,因此,可广泛应用于日用化学品、食品、药品、饲料等产品。
本发明所述的肉桂醛衍生物可作为防腐剂,广泛应用于日用化学品中,可用于洗发膏、洗发精、护发素、定型剂、烫发和染发剂、发乳、洗面乳、沐浴乳、液体皂、美容皂、婴儿皂,对皮肤和毛发作用温和,具有良好的抑菌杀菌性能,能防止产品的变质等;可作为防腐剂可用于口腔产品,如漱口水、牙膏、牙粉,具有杀菌、抑菌、护理口腔的功能;作为防腐剂应用于化妆品护肤产品,如护理化妆水、乳液、面霜、凝胶、洁面乳、防晒霜、护手霜等;可作为去屑剂,如在洗发护发产品中,添加了此类衍生物后,可以增加对毛发的亲和力,具有保护头发的功能,同时具有防腐杀菌能力,本发明衍生物还可以作为协同增效剂,与传统去屑剂复配使用,可以降低传统区去屑剂的用量,减少对皮肤的刺激性,降低致敏几率。
本发明肉桂醛衍生物可用于食品,作为防霉剂,防止食品的霉变,该类衍生物易溶于水,可广泛用作火腿、香肠、蛋糕、饼干的保鲜剂和月饼、蛋糕的防霉剂,月饼中加入适当比例的该类化合物, 能使其保持清香、新鲜、保存期可达4周以上,在蛋糕中添加可延长保存期2~5天,另外可应用于强化幼儿食品、老年食品、方便食品、功能食品、奶油、奶粉、炼乳、人造奶油、植物油、火腿、香肠、巧克力、口香糖等各类食品。
另外可将本发明肉桂醛衍生物应用于药品中,可以起到抗病毒作用,对流感病毒表现出超强的抑制活性。
根据本发明的肉桂醛衍生物,具有很好的抗霉杀菌作用,添加于饲料中,能使饲料保存很长时间不发霉、不变色或变味。
具体实施方式
实施例1 衍生物(1a)的制备
R:=CH-CH=CH-Ph
室温下,将7.8g肉桂醛(0.06mol)和5.1g(0.01mol)化合物a加入干燥的研钵中, 在研钵中混合均匀后,快速研磨20分钟,再于室温密闭保存12小时,薄层析监测反应完成后,加入5.0ml乙醇(75%),研磨15分钟,抽滤,无水乙醇洗涤三次,固体真空干燥后, 得白色粉末状固体,收率83%(以原料a计),熔点175℃(分解)。
实施例2 衍生物(1b)的制备
R:=CH-CH=CH-Ph
室温下,将1.0g去离子水,7.8g肉桂醛(0.06mol)和5.0g(0.01mol)化合物a加入微波反应器中, 于60℃微波辐射10分钟,冷却至室温,薄层析监测反应完成后,加入7.5ml无水乙醇,继续研磨10分钟,抽滤,无水乙醇洗涤三次,固体真空干燥后, 得白色粉末状固体,收率75%(以原料b计),熔点161℃(分解)。
实施例3 衍生物(1c)的制备
参照实施例2进行制备,将1.5g水,3.9g肉桂醛(0.03mol)和5.1g(0.01mol)化合物c加入反应器。经过后处理后,得白色粉末状固体,收率69%。
实施例4-7 衍生物(1d-1f,1i)的制备
参照实施例1进行制备,投料比按照氨基:肉桂醛=1:3(物质量比)进行及收率情况见表1。
实施例8-10 衍生物(1g-1h,1j)的制备
将各物料按照表1提供的反应条件,投料按照氨基:肉桂醛=1:3(物质量比)进行,投入超声反应器,收率见表1。
实施例11 衍生物1a-j溶解度实验
将衍生物1a-j样品分别进行溶解度测量,结果如下表2:
由上表可见,肉桂醛衍生化后溶解度大幅度提升,可以很好的溶于水并添加至配方中。
实施例12 常温稳定性研究
将衍生物1a-j样品进行稳定性研究,分别称量0.5g样品于称量瓶中,置于25℃恒温箱中,放置6个月,定期进行HPLC含量检测,结果见表3:
结果表明,肉桂醛含量大幅降低,而衍生物均没有明显的降低。
实施例13 加速稳定性研究
将衍生物1a-j样品进行加速稳定性研究,于48℃条件下,放置3个月,使用HPLC进行含量检测,结果见表4:
根据上表4,对该系列肉桂醛衍生化进行加速稳定性考察,结果表明,各衍生物均具有很好的稳定性,含量降低百分比均很小。
实施例14 衍生物1a-j最小抑菌浓度(MIC)测量
将衍生物1a-j进行最小抑菌浓度检测(MIC)测定,采用琼脂稀释法进行试验,各衍生物MIC检测数据如下表5:
由表中数据可见,该类衍生物的对金色葡萄菌、大肠杆菌、绿脓杆菌、洋葱假单胞菌、白色念珠菌、黑曲霉、马拉色菌等的抑制能力与肉桂醛相比优于肉桂醛。
实施例15 月饼防霉试验
霉菌分离:取普通PDA培养基,按国标GB5009.15霉菌酵母菌计数进行优势霉菌的分离鉴定。污染严重的样品采用10点点种法,从霉变月饼上采样,置于28℃温箱培养。
将市售月饼4块表面分别喷染分离到的霉菌混合孢子悬液0.5mL(约含100个/mL回收霉菌孢子),置于普通聚丙烯包装袋(500mL)中,同时放入RQA纸片一片(每片分别载有0mg、20mg的肉桂醛、20mg 1b、20mg 1j),封口后置于28℃温箱培养,肉眼观察月饼长霉情况,共观察20天,结果见表6。
结果可见,肉桂醛衍生物的保鲜能力优于肉桂醛。
实施例16 在中药饮品中的应用
将桑叶、余甘子各10g及0.1g菊花加入20g水中,加热提取,并浓缩至2.0mL,再加入0.5g蜂蜜、0.1g薄荷、0.01g化合物1a或1e混合均匀,100mL溶解稀释,拌匀过滤即可。
观察样品浑浊或沉淀析出情况,见表7。
实施例17 在宠物饲料中的应用
将18.0g砂糖、18.0g酱油、2.0g鸡油加入950.0g水中,强力搅拌成调味液,备用。再分别将550.0g鸡肉绞成馅;等量土豆、芋头、豌豆(30.0g)、45.0g红薯及50.0g胡萝卜混合并切成粒状(粒<3mm),与60.0g玉米淀粉、25.0g海藻胶、21.0g小麦粉、16.0g砂糖、15.0g复合维生素粉、12.0g食盐、3.0g酶化血粉、1.2g叶素、1.5g化合物1i及80.0g水混合,搅拌均匀制成馅状,置于盘中蒸20min,冷却,再于0~2℃冰水浴中恒温1.0h,取出切成20mm左右的小块,可保存12月份以上不变质。
实施例18 在乳制品中的应用
将10.0g高果糖玉米糖浆、9.0g乳制品基质、0.29g乳蛋白、0.19g果胶、0.15g乳酸、0.035g化合物1d、0.67mg 食用氧化镁,加入100g纯净水,混合均匀即可。
将以上混合物巴氏杀菌,并且暴露于光下8小时,并且将其与未添加1d样品做比较,外观表现出其具有更好的稳定性。
实施例19 在O/W型护肤霜中的应用
将两相分别加热到65~70℃,搅拌下将水相加到油相中。搅拌冷却,40℃下添加香料。
实施例20 在保湿霜中的应用
将A、B分别加热至70~75℃,然后将B在搅拌下加到A中后,搅拌至40~45℃,再加入C。继续搅拌15~20分钟,冷却至室温包装。
实施例21 在滋养弹力素中的应用
将A、B相分别加热到75℃,把B相加入A相,乳化3分钟(7500rpm),然后搅拌降温至45℃,加入C相,搅拌混合均匀,出料。
实施例22 在皂表洁面乳中的应用
将A相原料,搅拌加热至80-85℃,称取B相原料,搅拌加入至A相中,温度控制在80-85℃,继续搅拌约30-60分钟,使皂化完全;再称取C相原料,搅拌加入,温度控制在80℃左右,温度降至50℃时,加入D相原料,继续搅拌,冷却至室温,出料。

其中L可以为-H、烷基、-OH、-COOH、-CH2COOH、-CH2OH;
其中M可以为-H、烷基、-OH、-COOH、-CH2COOH、-CH2OH;
其中N可以为-H、烷基、-OH、-COOH、-CH2COOH、-CH2OH;
其中m=2~10的任意整数;
其中X可以为-H、烷基、-OH、-COOH、-CH2COOH、-CH2OH、ph-CH=CH-CH=N-;
其中Y可以为-H、烷基、-OH、-COOH、-CH2COOH、-CH2OH、ph-CH=CH-CH=N-;
其中Z可以为-H、烷基、-OH、-COOH、-CH2COOH、-CH2OH、ph-CH=CH-CH=N-;
其中n=2~15的任意整数。
2.根据权利要求1所述的肉桂醛衍生物,所述的烷基可以为C3~C16的直链烷基或取代苯基。
3.根据权利要求1所述的肉桂醛衍生物,其特征在于:所述的n为2~15的任意整数,优选3-8。
4.权利要求1所述的肉桂醛衍生物在日用化学品中的应用。
5.权利要求1所述的肉桂醛衍生物在药品中的应用。
6.权利要求1所述的肉桂醛衍生物在食品中的应用。
7.权利要求1所述的肉桂醛衍生物在饲料中的应用。
