本发明提供一种含银高分子的封填材料,包含:一卜特兰水泥;一银高分子,该银高分子可溶于水;一化合物,结构式如下式(I)、(II)或(III)所示: 其中,M2+为二价金属离子,A‑、B‑、C2‑、D3‑各自独立为且相异的C1~C15有机酸根离子或无机酸根离子,x介于0至2,y介于0至4,且z介于0至6,该银高分子可提高该封填材料的一辐射遮蔽性、一机械物性、及一抑菌功能,可用于一牙部的根管填补或封填,改善封填材料的临床操作性质。
Description
技术领域
本发明关于一种封填材料,更详细地说是关于一种用于牙齿填补且含银高分子的封填材料。
现有技术
于临床上,龋齿窝洞及根管区域为常见的牙科问题。根管治疗是指对于病变的齿髓及其根尖周围组织,采用牙齿覆髓、断髓剂、或根尖充填剂,以封闭患部,避免细菌感染发炎,并促进牙根周围组织修补愈合的方法。
目前使用的根管治疗材料中,以Dentsply公司所发展的MTA(mineral trioxide aggregate)最为广泛的应用于牙科领域中。MTA为一种矿物氧化物的聚合物,最早于1993年提出并进行一连串的研究与改良,MTA的主要成分为氧化钙与硅酸钙化合物的tricalcium complex粉末颗粒,加入少量的多种矿物氧化物目的在于调整材料的化学物理性质。最初的MTA材料为灰色三氧矿化物(grey MTA),主要成分为75%的第I型卜特兰水泥(Portland cement)、5%石膏(gypsum)、20%氧化铋(bismuth oxide)。然灰色MTA于硬化/固化后为灰绿色,影响修复的齿部外观,因此Dentsply公司研发一种白色MTA(去除卜特兰水泥中具深绿色泽的铁铝酸四钙)以取代灰色MTA。
虽然白色MTA已具有良好封填效果,然在操作上仍有其缺点。白色MTA具有较差的砂状手感及较差的操作性,以及需要3至4小时的硬化时间,使手术过程中,患部易受到组织液或血液的冲刷,影响封闭性与治疗效果,且于拌合初期粉体呈现颗粒状不易聚集等问题。
理想的牙科封填材料应具备,良好封闭能力、生物相容性、良好临床操作性质、较短的固化时间外,同时须具备辐射遮蔽性及优异的机械物性,以提高x光诊断的显影程度,或是较佳的弹性避免材料固化后脆裂。为此,若能缩短现有MTA固化时间、提高辐射遮蔽性及机械物性,则可达到改善此等缺失的目的。
发明内容
本发明的一目的为改善现有MTA固化时间、提高现有MTA辐射遮蔽性及其机械物性,使封填材料的临床操作性质获得改善。
为达成上述目的,本发明提供一种含银高分子的封填材料,包含:一卜特兰水泥;一银高分子,该银高分子可溶于水;一化合物,结构式如下式(I)、(II)或(III)所示:
其中,M2+为正二价金属离子,A-、B-、C2-、D3-各自独立且相异的有机酸根离子或无机酸根离子,x介于0至2,y介于0至4,且z介于0至6,该银高分子可提高该含银高分子的封填材料的一辐射遮蔽性、一机械物性、及一抑菌功能。
根据本发明的一较佳实施例,其中该银高分子以溶液形式存在。
根据本发明的一较佳实施例,其中该银高分子溶液中该银高分子浓度为4000ppm。
根据本发明的一较佳实施例,其中该卜特兰水泥为第一型卜特兰水泥。
根据本发明的一较佳实施例,其中M2+选自铍离子、镁离子、钙离子、锶离子、及钡离子所组成的群组。
根据本发明的一较佳实施例,其中该无机酸根离子选自硝酸根、硫酸根、氯离子、及磷酸根所组成的群组;该有机酸根离子选自葡萄糖酸根、乳酸根、丙酮酸根、乙醇酸根、氯乙酸根、二氯乙酸根、三氯乙酸根、氯乙酸根、酒石酸根、琥珀酸根、戊二酸根、顺丁烯二酸根、反丁烯二酸根、丙二酸根、柠康酸根、邻苯二甲酸根、间苯二甲酸根、对苯二甲酸根、柠檬酸根、丙三甲酸根、苯三甲酸根、及1,3,5-戊烷三羧酸根所组成的群组。
根据本发明的一较佳实施例,其中A-为乳酸根、B-为葡萄糖酸根、C2-为硫酸根。
根据本发明的一较佳实施例,其中该化合物为乳酸葡萄糖酸钙、乳酸硫酸钙、硫酸葡萄糖酸钙。
一种含银高分子的封填材料的用途,用于制备填补或封填牙部的材料。

本发明将搭配图式,为本发明的较佳实施例进行更详细的说明。然而,此说明书中所使用任何形式的实施例仅为说明用,无法以此限制本发明。更准确地说,实施例的提供是为了使本发明更彻底和完整地说明,并对本发明所属领域中技术人员充分传达。
本发明提供一种含银高分子的封填材料,可用于一牙部的填补或封填,例如根管封填材料,包含:一卜特兰水泥、一银高分子、一化合物结构式如下式(I)、(II)或(III)所示:
于一较佳实施方式中,M2+为二价金属离子,选自铍离子、镁离子、钙离子、锶离子、及钡离子所组成的群组。
于一较佳实施方式中,A-、B-、C2-、D3-各自独立为且相异的C1~C15有机酸根离子或无机酸根离子,例如:A-为乳酸根、B-为葡萄酸根、C2-为硫酸根,但不限于此。无机酸根离子选自硝酸根、硫酸根、氯离子、及磷酸根所组成的群组;有机酸根离子选自葡萄糖酸根、乳酸根、丙酮酸根、乙醇酸根、氯乙酸根、二氯乙酸根、三氯乙酸根、氯乙酸根、酒石酸根、琥珀酸根、戊二酸根、顺丁烯二酸根、反丁烯二酸根、丙二酸根、柠康酸根、邻苯二甲酸根、间苯二甲酸根、对苯二甲酸根、柠檬酸根、丙三甲酸根、苯三甲酸根、及1,3,5-戊烷三羧酸根所组成的群组。
该化合物的制备实施例将详述于下列实施例1至3。
实施例1:乳酸葡萄糖酸钙(calcium lactate gluconate)
将27g乳酸及36g葡萄糖酸内酯分别加入100mL去离子水中混合均匀后配置成溶液,加入14g氧化钙,经混合搅拌1小时反应后过滤,所收集的滤液,即为乳酸葡萄糖酸钙溶液,其pH值约9.8,经干燥即为乳酸葡萄糖酸钙粉,此粉末于25℃水中的溶解度为35.7至41g/100mL。
实施例2:乳酸硫酸钙(calcium lactate sulfate)
将2.45g硫酸及1.8g乳酸加入50mL去离子水中混合均匀后配置成溶液,加入2g的氧化钙,经混合搅拌反应1小时后过滤,所收集的滤液即为乳酸硫酸钙粉,该粉末在25℃水中的溶解度为4.6至5g/100mL。
实施例3:硫酸葡萄糖酸钙(calcium sulfate gluconate)
将2.45g硫酸及3.92g葡萄糖酸内酯加入50mL去离子水中混合均匀后配置成溶液,加入2g的氧化钙,经混合搅拌反应1小时后过滤,所收集的滤液即为硫酸葡萄糖酸钙溶液,其pH值为9.3,经干燥即为硫酸葡萄糖酸钙粉末,该粉末在25℃水中的溶解度为30.1至31.7g/100mL。
实施例4:添加化合物的封填材料
以实施例1所制备的乳酸葡萄糖酸钙配置为浓度20wt%的乳酸葡萄糖酸钙溶液,接着将此乳酸葡萄糖酸钙溶液与第一型卜特兰水泥粉末以水粉比1:4的重量比例混合,以形成一含化合物的封填材料。
实施例5:含银高分子的封填材料
本发明选用新型具水溶性的银高分子,例如丙烯酸钠和丙烯酸银的共聚物,其化学式如式(IV)所示,但不限于此。由于所选用的银高分子的分子量高达500万以上,是世界上第一个可以溶于水的含银高分子,既具有银离子广效的杀菌效果及水溶的便利加工特点,且由于其分子量为超过500万的巨大分子尺寸,是一般小分子抗菌剂的数万倍,故不易穿透过皮肤或粘膜进入人体内,影响人体健康,是非常安全的抗菌剂。
以实施例1所制备的乳酸葡萄糖酸钙及银高分子浓度4000ppm的银高分子混合为银高分子溶液,其中乳酸葡萄糖酸钙的浓度为20wt%,接着将此银高分子溶液与第一型卜特兰水泥粉末以水粉比1:4的重量比例混合,以形成一含银高分子的封填材料。
比较例1:
将去离子水与卜特兰水泥粉末以水粉比1:4的重量比例混合,以形成一封填材料。
固化时间分析
当卜特兰水泥与水溶液混合后即发生水合作用,其固化时间依国际标准方法(ISO 1566)进行测试,以测试针头顶端直径1mm(Vicat needle),以300g的荷重垂直下压样品表面,测试针头刺穿样品的深度少于1mm的时间,定义为初始固化时间(initial setting time);而测试针头与样品接触后,样品上不再有明显可见的压痕的时间,定义为完全固化时间(final settingtime)。
将实施例5所制备的含银高分子的封填材料及实施例4的含化合物的封填材料,与比较例1进行比较。其结果如表1所示,实施例4的封填材料约需20分钟,而实施例5所制备的含银高分子的封填材料约需30分钟,与比较例1需约2个半小时相比,皆大幅缩短等待固化的时间,因此于操作上的方便性也有所提升。

辐射不通透性分析
为分析待测品的「辐射不通透性」(Radiopacity)能力,亦即待测品的降低或完全阻隔X射线穿透的能力。依实施例4及实施例5所制备的封填材料,以水粉比1:4的比例调拌均匀。将调拌好的样品填入铁氟龙模具(直径10±0.1mm,高度1±0.1mm)中,于顶部及底部盖上盖板,确保样品厚度为1mm,每种待测物制备6个圆型锭片(N=6)。依据样品的使用说明等待其固化完成。并将样品放在温度37℃且湿度100%的烘箱使的固化24小时。将固化好的样品(含模具与盖板)与标准品(含与样品相同的盖板)一同放在感光底片(Imaging Plate REF 1340074,Kodak)上,使用的标准品为铝板铝纯度98%以上、铜含量0.1%以下、铁含量1%以下;长度32mm、宽度25mm、厚度从2mm–16mm,铝梯之间相邻间距为2mm,厚度精确到10μm。将X射线产生器(Portable dental x-ray system PORX)的照射口固定于感光底片上方300mm处,确认输出电压为65±5kV,照射时间为0.25秒,对样品及标准品进行照射。X射线照射后,以X射线摄影显影系统(ScanX Duo Digital Imaging System,Air Techniques)对感光底片进行影像显影。
以影像量化分析软体ImageJ对影像进行量化。依照标准品的各阶厚度所得的量化数值绘制减量线,并且依照样品的量化数值与减量线公式回推样品的「等效铝板厚度」,其结果如图1所示,实施例5的辐射不通透性为5.82±0.32mm Al,高于实施例4的辐射不通透性5.17±0.39mm Al,其p<0.05统计上具有显著差异,显示实施例5的含银高分子的封填材料具有较佳的辐射不通透性,换句话说,加入银高分子后可提升封填材料的辐射遮蔽性。
径向拉伸强度分析
机械物性可由众多方式检测,其中的一为「径向拉伸强度」(Diametral Tensile Strength,DTS)。为建立内部自行分析机械物性的能力并规范之,特定此「径向拉伸强度分析方法」。
以实施例4及实施例5所制备的封填材料为待测物,依水粉比1:4的比例调拌均匀。将调拌好的待测物填入铁氟龙模具(直径6±0.5mm,高度5±0.5mm)中,于顶部及底部盖上盖板,确保样品厚度为5±0.5mm,每种待测物制备6个圆柱型样品(N=6)。依据样品的使用说明等待其固化完成。并将样品放在温度37℃且湿度100%的烘箱使的固化24小时。将固化好的样品由模具上取下,实际量测每个样品的尺寸(直径与高度)并纪录之。开启质地分析仪(Texture analyzer TA.XTplus),选用P/25探头,调整施力速率为0.6mm/min,施力方向为对径向压缩。将圆柱型样品以横摆的方式置于探头的正下方进行压缩测试。纪录样品开始被施力至样品破裂的数据,其断裂伸长量(elongation at break)如表2所示。将破裂瞬间的最大力量数值(N),带入下列公式将其换算成DTS(MPa),并将所有结果计算出平均值与标准差作为最终结果:

结果如图2所示,实施例5的径向拉伸强度分析结果为2.23±0.29MPa,高于实施例4的径向拉伸强度1.61±0.3MPa,其p<0.05统计上具有显著差异。实施例5的含银高分子的封填材料在固化后,具有较佳的径向拉伸强度,也就是说,加入银高分子后可提升封填材料的机械物性,使封填材料在固化后弹性提升,降低物性脆度。
综上所述,本发明所提出的实施例1、2、3的化合物,以及添加银高分子的实施例5,皆可有效地缩短白MTA固化时间;而添加银高分子的实施例5还可提高辐射遮蔽性(辐射不通透性分析)及机械物性(径向拉伸强度分析),可用于一牙部的填补或封填,有助于改善临床操作便利性及诊断。
惟以上所述者,仅为本发明的较佳实施例,但不能以此限定本发明实施的范围;故,凡依本发明申请专利范围及发明说明书内容所作的简单的等效改变与修饰,皆仍属本发明专利涵盖的范围内。
Claims (10)
1.一种封填材料,其特征在于,包含:
一卜特兰水泥;
一银高分子,该银高分子可溶于水;以及
一化合物,结构式如下式(I)、(II)或(III)所示:
其中,M2+为二价金属离子,A-、B-、C2-、D3-各自独立为且相异的C1~C15有机酸根离子或无机酸根离子,x介于0至2,y介于0至4,且z介于0至6。
2.如权利要求1所述的封填材料,其特征在于,该银高分子以溶液型式存在。
3.如权利要求1所述的封填材料,其特征在于,该银高分子为丙烯酸钠与丙烯酸银的共聚物。
4.如权利要求1所述的封填材料,其特征在于,该银高分子溶液中该银高分子浓度为4000ppm。
5.如权利要求1所述的封填材料,其特征在于,该卜特兰水泥为第一型卜特兰水泥。
6.如权利要求1所述的封填材料,其特征在于,M2+选自铍离子、镁离子、钙离子、锶离子、及钡离子所组成的群组。
7.如权利要求1所述的封填材料,其特征在于,该无机酸根离子选自硝酸根、硫酸根、氯离子、及磷酸根所组成的群组;该有机酸根离子选自葡萄糖酸根、乳酸根、丙酮酸根、乙醇酸根、氯乙酸根、二氯乙酸根、三氯乙酸根、氯乙酸根、酒石酸根、琥珀酸根、戊二酸根、顺丁烯二酸根、反丁烯二酸根、丙二酸根、柠康酸根、邻苯二甲酸根、间苯二甲酸根、对苯二甲酸根、柠檬酸根、丙三甲酸根、苯三甲酸根、及1,3,5-戊烷三羧酸根所组成的群组。
8.如权利要求1所述的封填材料,其特征在于,A-为乳酸根、B-为葡萄糖酸根、C2-为硫酸根。
9.如权利要求1所述的封填材料,其特征在于,该化合物为乳酸葡萄糖酸钙、乳酸硫酸钙、或硫酸葡萄糖酸钙。
10.一种如权利要求1的封填材料的用途,其特征在于,用于制备填补或封填牙部的材料。
