Description
Verfahren zur Herstellung neuer sekundärer Amine
Gegenstand der Erfindung ist ein Verfahren zur Herstellung von sekundären Aminen der Formel

worin R einen niederen aliphatischen Kohlenwasserstoffrest bedeutet, oder ihren Salzen.
Der niedere aliphatische Kohlenwasserstoffrest R ist z. B. ein niederer, gerader oder verzweigter, in beliebiger Stelle verbundener Alkyl- oder Alkenylrest, z. B. Allyl, n-Propyl, Isopropyl, Butyl, sek.-Butyl, Amyl, Isoamyl oder Hexyl, vor allem aber Methyl oder Äthyl.
Die neuen Verbindungen und ihre Salze besitzen ausgesprochen symp athicolytische Wirkung mit einer sedativen Wirkungskomponente. Sie können dementsprechend als Medikamente, vor allem als Sedativa, verwendet werden.
Besonders wertvoll ist das N-[fl-(cMethoxy- phenoxy)-äthyl]-tetrahydrofurfuryl-amin der Formel

worin R die oben angegebene Bedeutung hat, reduziert. Die Reduktion kann in üblicher Weise, z. B. durch katalytische Hydrierung, erfolgen.
Je nach der Arbeitsweise erhält man die neuen Verbindungen in Form der Basen oder ihrer Salze.
Aus den Salzen können in an sich bekannter Weise die freien Aminbasen gewonnen werden. Von letzteren wiederum lassen sich durch Umsetzung mit Säuren, die zur Bildung therapeutisch verwendbarer Salze geeignet sind, Salze gewinnen, wie z. B. Halogenwasserstoffsäuren, beispielsweise Salzsäure oder Bromwasserstoffsäure, Perchlorsäure, Salpetersäure oder Thiocyansäure, Schwefel-oder Phosphorsäuren, oder organische Säuren, wie Ameisensäure, Essigsäure, Propionsäure, Glykolsäure, Milchsäure, Brenztraubensäure, Oxalsäure, Malonsäure, Bernsteinsäure, Maleinsäure, Fumarsäure, Äpfelsäure, Weinsäure, Zitronensäure, Ascorbinsäure, Hydroxymaleinsäure, Dihydroxymaleinsäure, Benzoesäure, Phenylessigsäure, 4-Amino-benzoesäure, 4-Hydroxy-benzoesäure, Anthranilsäure, Zimtsäure, Mandelsäure, Salicylsäure, 4-Amino-salicylsäure,
2-Phenoxy-benzoesäure, 2-Acetoxy-benzoesäure, Methansulfons äure, 2ithansulfon- säure, Hydroxyäthansulfonsäure, Benzolsulfonsäure, p-Toluol-sulfonsäure, Naphthalinsulfonsäure oder Sulfanilsäure, oder Methionin, Tryptophan, Lysin oder Arginin.
Die Salze der neuen Verbindungen können auch zur Reinigung der erhaltenen Basen dienen, indem man die Basen in die Salze überführt, diese abtrennt und aus den Salzen wiederum die Basen freisetzt.
Die Ausgangsstoffe sind bekannt oder können nach an sich bekannten Methoden gewonnen werden.
Die neuen Verbindungen können als Heilmittel, z. B. in Form pharmazeutischer Präparate Verwendung finden, welche sie oder ihre Salze in Mischung mit einem für die enterale oder parenterale Applikation geeigneten pharmazeutischen organischen oder anorganischen, festen oder flüssigen Trägermaterial enthalten.
In dem folgenden Beispiel sind die Temperaturen in Celsiusgraden angegeben.
Beispiel
24,7 g (0,1 Mol) N-[A-(o-Methoxyphenoxy)- äthyl]-turfuryl-amin werden in 150cm3 Äthanol gelöst und in Gegenwart von 5 g Raneynickel im Autoklaven bei 100 atü Wasserstoffdruck und 50-800 katalytisch hydriert. Das Hydrierungsprodukt wird im Hochvakuum destilliert. Man erhält das N-[fl-(o- Methoxyphenoxy)-äthyl]-tetrahydrofurfuryl-amin der Formel

als farblose Flüssigkeit vom Kp. 0,07: 128 . Das mit Chlorwasserstoff in Essigester hergestellte Hydrochlorid schmilzt nach dem Umkristallisieren aus Essigester bei 102-104 .
Das als Ausgangsstoff verwendete N-[ss-(o-Methoxyphenoxy)-äthyl]-furfu ryl-amin wird z. B. wie folgt erhalten:
18,7 g (0,1 Mol) vMethoxyphenoxy-äthylchlorid und 38,4 g (0,4 Mol) Furfuryl-amin werden nach Zugabe von 50cm3 Isopropanol 15 Stunden unter Rückfluss gekocht. Die dunkelgefärbte Lösung wird im Wasserstrahlvakuum eingedampft, der Rückstand in Methylenchlorid aufgenommen und mit 100cm3 2n Natronlauge und anschliessend zweimal mit 50 cm3 Wasser ausgeschüttelt. Die Methylenchloridlösung trocknet man über wasserfreiem Natriumsulfat und destilliert das Lösungsmittel und überschüssiges Furfuryl-amin ab. Der Rückstand wird in wenig Benzol gelöst und auf eine Chromatographiersäule von 400 g Aluminiumoxyd (Aktivität II) aufgezogen.
Durch Elution mit Benzol-Methylenchlorid erhält man das N¯ [ss-(o-Methoxyphenoxy)-äthyl]-furfuryl-amin als hellgelbes, viskoses Ö1.
Das in üblicher Weise hergestellte Hydrochlorid bildet aus Alkohol-Essigester leicht bräunlich gefärbte Kristalle vom F. 112-1 13 .

The lower aliphatic hydrocarbon radical R is z. B. a lower, straight or branched alkyl or alkenyl radical connected in any position, e.g. B. allyl, n-propyl, isopropyl, butyl, sec-butyl, amyl, isoamyl or hexyl, but especially methyl or ethyl.
The new compounds and their salts have a pronounced sympatholytic effect with a sedative component. Accordingly, they can be used as drugs, especially as sedatives.

The inventive method for the preparation of the new amines is characterized in that compounds of the formula

Depending on the procedure, the new compounds are obtained in the form of the bases or their salts.
The free amine bases can be obtained from the salts in a manner known per se. From the latter, in turn, salts can be obtained by reaction with acids which are suitable for the formation of therapeutically useful salts, e.g. B. hydrohalic acids, for example hydrochloric acid or hydrobromic acid, perchloric acid, nitric acid or thiocyanic acid, sulfuric or phosphoric acids, or organic acids such as formic acid, acetic acid, propionic acid, glycolic acid, lactic acid, pyruvic acid, oxalic acid, malonic acid, succinic acid, maleic acid, fumaric acid , Citric acid, ascorbic acid, hydroxymaleic acid, dihydroxymaleic acid, benzoic acid, phenylacetic acid, 4-amino-benzoic acid, 4-hydroxy-benzoic acid, anthranilic acid, cinnamic acid, mandelic acid, salicylic acid, 4-amino-salicylic acid,

The starting materials are known or can be obtained by methods known per se.
The new compounds can be used as remedies, e.g. B. in the form of pharmaceutical preparations are used which contain them or their salts in a mixture with a pharmaceutical organic or inorganic, solid or liquid carrier material suitable for enteral or parenteral administration.

18.7 g (0.1 mol) of methoxyphenoxyethyl chloride and 38.4 g (0.4 mol) of furfurylamine are refluxed for 15 hours after addition of 50 cm3 of isopropanol. The dark colored solution is evaporated in a water jet vacuum, the residue is taken up in methylene chloride and extracted with 100 cm3 of 2N sodium hydroxide solution and then twice with 50 cm3 of water. The methylene chloride solution is dried over anhydrous sodium sulfate and the solvent and excess furfurylamine are distilled off. The residue is dissolved in a little benzene and drawn up on a chromatography column of 400 g of aluminum oxide (activity II).
Elution with benzene-methylene chloride gives the N¯ [ss- (o-methoxyphenoxy) ethyl] -furfurylamine as a light yellow, viscous oil.
The hydrochloride produced in the usual way forms slightly brownish colored crystals from alcohol / ethyl acetate with a mp of 112-113.
