Abstract

ESTABELECIMENTO DE LINHAGEM DE CÉLULAS-TRONCO EMBRIONÁRIAS HUMANAS USANDO CÉLULAS DE MAMÍFEROS, onde são reveladas preparações purificadas de células-tronco embrionárias humanas com determinadas características específicas da população. Esta preparação é caracterizada por uma expressão positiva dos seguintes marcadores de superficie celular pluripotentes: SSEA-1 (-); SSEA-4 (+); TRA-1-60 (+); TRA-1-81 (+); fosfatase alcalina (+), bem como um conjunto de marcadores celulares ES incluindo Oct-4, Nanog, Rex1, Sox2, Thy1, FGF4, ABCG2, Dppa5, UTF1, Cripto1, hTERT, Conexina-43 e Conexina-45. As células da preparação são negativas para marcadores específicos da linhagem como Queratina 8, Sox- 1, NFH (ectoderme), MyoD, brachyury, actina cardíaca (mesoderme), HNF-3 beta, albumina e PDX1 (endoderme). As células da preparação são células-tronco embrionárias humanas, possuem cariótipos normais, exibem alta atividade da telomerase e continuam a proliferar em um estado indiferenciado após cultura contínua de mais de 40 passagens. A linhagem de células-tronco embrionárias Relicell® Hes 1 também possui a habilidade, ao longo da cultura, de diferenciar os tipos de tecidos e células derivados de todas as três camadas de origem embrionárias (endoderme, mesoderme e ectoderme). Também são revelados métodos para isolar a linhagem de células-tronco embrionárias humanas.
