Abstract
"MéTODOS PARA DETECçãO DA PRESENçA DE SANGUE EM UMA AMOSTRA BIOLóGICA PARA DETECçãO DE SANGRAMENTO GASTROINTESTINAL INFERIOR, PARA DETECçãO DE SANGRAMENTO NO TRATO GASTROINTESTINAL SUPERIOR, PARA DIAGNOSTICAR CONDIçõES DE DOENçA E PARA DETECçãO DA PRESENçA DE UM ANALITO EM UMA AMOSTRA BIOLóGICA" A presente invenção refere-se geralmente ao método de detecção da presença de um analito na amostra biológica. O primeiro método para detecção de sangue na amostra biológica envolve aplicar a amostra a primeira região da matriz de ensaio, detectar globina ou hemoglobina por meio da prova imunológica na segunda região da matriz de ensaio e detectar hemo por meio da prova de base em cromógeno na terceira região da matriz de ensaio. Este método é capaz de diferenciação entre o sangramento do trato gastrointestinal superior e inferior e é útil para o diagnóstico de doenças do trato gastrointestinal, em particular, doenças do trato gastrointestinal inferior tal como o câncer coloretal. The first method for detecting blood in the biological sample involves applying the sample to the first region of the test matrix, detecting globin or hemoglobin by means of the immunological test in the second region of the test matrix and detecting heme by means of the chromogen base test in the third region of the test matrix. This method is capable of differentiating between bleeding from the upper and lower gastrointestinal tract and is useful for the diagnosis of diseases of the gastrointestinal tract, in particular, diseases of the lower gastrointestinal tract such as colorectal cancer. The second method disclosed involves applying the sample to the first region of the test matrix, the sample is allowed to flow to the second region where it comes into contact with the anti-analyte immunointeractive molecule and the anti-analyte labeled immunointeractive conjugate. Any analyte-anti-analyte complex that is formed is immobilized and detected in this second region, any immunointeractive conjugate labeled non-complexed anti-analyte is allowed to flow to the third region where it is detected.
