Abstract
Patente de Invenção: <B>"SISTEMA DE TRANSFORMAçãO EM CANDIDA UTILIS"<D>. A presente invenção fornece um sistema de transformação útil para expressar proteínas heterólogas na levedura Candida utilis, com base na obtenção de mutantes auxotróficos dessa espécie bem como no isolamento de diferentes genes, a partir de uma biblioteca genómica, que complementam as referidas auxotrofias. O sistema de transformação emprega como hospedeiros novos mutantes auxotróficos obtidos da cepa NRRL Y-1084 de Candida utilis, os quais são defeituosos principalmente nas vias biossintéticas da uracil e da histidina, os quais são transformados com plasmídios que contêm como marcadores de seleção os genes URA3 e HIS3 de Candida utilis. Outro aspecto da invenção é o isolamento do gene que codifica a enzima sacarose invertase ou <225>-frutofuranosidase de Candida utilis, bem como a identificação de seq³ências para promoção, sinalização de secreção e terminação do referido gene INV1. Estas seq³ências podem ser vantajosamente utilizadas para obter a expressão de proteínas heterólogas na referida levedura.Invention Patent: <B> "TRANSFORMATION SYSTEM IN CANDIDA UTILIS" <D>. Another aspect of the invention is the isolation of the gene that encodes the enzyme sucrose invertase or Candida utilis -fructofuranosidase, as well as the identification of sequences for promotion, secretion signaling and termination of said INV1 gene. These sequences can be advantageously used to obtain the expression of heterologous proteins in said yeast.
