Abstract
Se proporcionan métodos para producir una proteína recombinante de interés en un cultivo de células de mamífero en medios que carecen de IGF-1. También se proporcionan métodos para producir células de mamífero capaces de crecer en medios que carecen de IGF-1. Reivindicación 1: Un método para producir una proteína de interés a partir de un cultivo de células de mamífero que comprende: (a) cultivar una célula de mamífero que expresa una proteína de interés en un segundo medio de cultivo celular que tiene 0.05 mg/l o menos de factor de crecimiento insulínico (IGF-1) para expresar la proteína de interés, en donde la célula de mamífero se ha adaptado directamente a crecer en un primer medio de cultivo celular que tiene 0.03 mg/l o menos de IGF-1 y comprende un ácido nucleico heterólogo que codifica la proteína de interés; y (b) recuperar la proteína de interés producida por la célula de mamífero. Reivindicación 16: Un método para adaptar directamente una célula de mamífero a medios IGF⁻ que comprende: a) cultivar una población de células de mamífero en un medio de cultivo celular que comprende 0.03 mg/l o menos de IGF-1; b) obtener células individuales de la población de células de mamífero mediante clonación de una sola célula; c) expandir y someter a pases las células individuales hasta conseguir una recuperación del 90% o más y un tiempo de duplicación inferior a 30 horas.
