Abstract
La presente se refiere al campo de la biotecnología, la biología molecular y la medicina; en particular, a la enzima nucleasa y su uso. Más específicamente, la presente se refiere a la enzima nucleasa PaCas9. La presente también se refiere a un ácido nucleico que codifica dicha nucleasa, un constructo genético, un vector de expresión, un vector de administración que comprende dicho ácido nucleico, un liposoma que comprende dicha nucleasa o ácido nucleico que codifica dicha nucleasa, un método para producir una nucleasa, métodos para la administración, y una célula hospedadora que comprende un ácido nucleico que codifica dicha nucleasa. Reivindicación 1: Nucleasa PaCas9 que tiene una secuencia de aminoácidos de la SEQ ID Nº 2. Reivindicación 2: Una molécula de ácido nucleico aislada que codifica una nucleasa PaCas9 según la reivindicación 1, y que tiene una secuencia de nucleótidos de la SEQ ID Nº 1. Reivindicación 3: Un vector de expresión que comprende un ácido nucleico según la reivindicación 2. Reivindicación 12: Un método para producir una nucleasa PaCas9 según la reivindicación 1, que comprende cultivar una célula hospedadora según la reivindicación 10 en un medio de crecimiento en condiciones suficientes para producir dicha nucleasa PaCas9, si es necesario, seguido por el aislamiento y la purificación de la nucleasa PaCas9 obtenida.
