Abstract
Un método para producir una proteína de interés, que comprende la creacion de un mamífero transgénico no humano capaz de producir dicha proteína en su leche, la obtencion de dicha leche del mamífero transgénico no humano y la purificacion de dicha proteína de interés a partir de la leche. Se generaron animales bovinos transgénicos, que son capaces de producir hormona de crecimiento humana en sus glándulas mamarias. El método involucra el clonado de una construccion genética que codifica para el gen de hGh y el promotor de la beta caseína convenientemente ensamblados en un vector de expresion. También incluye los procedimientos de transfeccion de células somáticas fetales bovinas, generalmente fibroblastos, y la transferencia nuclear a ovocitos bovinos enucleados, generando así embriones transgénicos. El método también incluye otros procedimientos para generar embriones transgénicos para ulterior expansion del rebano transgénico, tales como el subclonado de hembras bovinas transgénicas, la superovulacion de vacas transgénicas y su inseminacion con semen de un macho bovino transgénico o no transgénico, y al superovulacion de vacas no transgénicas y su inseminacion con semen obtenido de un macho bovino transgénico. Posteriormente, los embriones transgénicos darán origen a ganado transgénico productor de grandes cantidades de hormona de crecimiento humana en su leche, material a partir del cual ésta se purifica en forma completa y se somete a los análisis que permitan garantizar el cumplimiento de todos los requisitos necesarios para la fabricacion de una producto biofarmacéutico seguro y eficaz.
