Abstract

Fremgangsmåte for sekvensering av et målpolymermolekyl karakterisert av trinnene: (i) behandling av målpolymeren med et virkemiddel som degraderer sekvensielt minst en ende av målpolymeren; (ii) omdannelse av minst en del av den degraderte enden av ulike degraderte polymerer over i en lesbar signalsekvens, og merking av hver av de degraderte polymerene med en merkelapp som representer den relative rekkefølgen av degraderingen; (iii) bestemmelse av sekvensen av den lesbare signalsekvensen; og (iv) bestemmelse av sekvensen av målpolymeren ved å benytte sekvensdata oppnådd i trinn (iii) og identifikasjon av hver assosierte merkelapp.
