Abstract
Se proporcionan metodos para detectar un polinucleotido mutante en mezcla con polinucleotidos mutantes, polinucleotidos de tipo silvestre y polinucleotidos no relacionados; el metodo utiliza un iniciador de extension complementario a una primera secuencia objetivo tanto en los polinucleotidos de tipo silvestre como en los polinucleotidos mutantes; el metodo tambien utiliza una sonda complementaria a una segunda secuencia objetivo en los polinucleotidos de tipo silvestre, pero no en los polinucleotidos mutantes; la extension de los iniciadores unidos a la primera secuencia objetivo en los polinucleatidos mutantes produce productos de la larga extension; la extension de los iniciadores unidos a la primera secuencia objetivo en los polinucleotidos de tipo silvestre es bloqueada por la sonda que esta unida a la segunda secuencia objetivo; se producen productos de corta extension o productos sin extension; los productos de extension se aislan y se utilizan en una reaccion en cadena de polimerasa (PCR); la PCR amplifica preferencialmente los productos de larga extension.
