Abstract

Se proporciona un metodo de transferencia nuclear que involucra el transplante o diferenciacion de nucleos de celula donadora en oocitos enucleados de una especie diferente a la celula donadora, en conjunto con un citoplasma y/o mitocondria compatible (de la misma especie del donador). Las unidades de transferencia nuclear resultante son utiles para la produccion de celulas madre embrionicas isogenicas, en particular celulas madre o embrionicas isogenicas humanas. Estas celulas tipo madre o embrionicas son utiles para producir las celulas diferenciadas deseadas y para la introduccion, remocion o modificacion, de los genes deseados, por ejemplo, en sitios especificos del genoma de tales celulas por recombinacion homologa. Estas celulas, que pueden contener un gen heterologo, son especialmente utiles en terapias de transplante celular y para estudio in vitro de diferenciacion celular. La eficiencia de transferencia nuclear se aumenta por la introduccion de citoplasma o ADN mitocondrial compatible (misma especie o similar a la celula o nucleo donador). Tambien, la eficacia de transferencia nuclear puede opcionalmente aumentarse ademas por celulas donadoras geneticamente alteradas para inhibir la apoptosis, seleccionadas para un ciclo celular especifico y/o cultivando bajo condiciones que aumentan el crecimiento y desarrollo embrionico.
