Abstract

La presente invención se refiere a un método para generar un conjugado de anticuerpo-carga útil por medio de una transglutaminasa microbiana (MTG). El método comprende un paso para conjugar un ligador que tiene un residuo de amina primaria, el ligador tiene la estructura de péptido (mostrada en la dirección N->C) (Aax)m-(Aax)(NH2)-(Aax)n-B-(Aax)o, o (Aax)m-B-(Aax)n-(Aax)(NH2)-(Aax)o, a un residuo Gln comprendido en la cadena pesada o ligera de un anticuerpo. Aax(NH2) es un aminoácido, derivado de aminoácido o mimético de aminoácido que comprende una cadena lateral que tiene un grupo amina primaria (Figura 1).
