Abstract

The present invention relates to the development of a quick, easy and reproducible method for evaluating the antioxidant ability in food samples, where a system of the model ABTS/lacasse was used for that purpose, the system including the chromogenic compound 2,2'-azino-bis(3-ethylbenz- thiazoline-6-sulfonic acid), (ABTS) and lacasse , which is a polyphenol oxidase for generating ABTS+ free radicals in the presence of molecular oxygen. The method involves preparing a pattern curve where a standard antioxidant, preferably glycolic acid, is used. The reaction mixture of 1 ml includes: Denilite IISTM or Rhus vernicifera lacasse, ABTS 0.05 mM in an acetate buffer of 0.1 M with pH 4 and glycolic acid at different concentrations and at a temperature of 25°C. The lag time for inhibiting the formation of the ABTS radical is determined by following the reaction in a spectrophotometer at 728 nm. An antioxidant concentration vs ABTSò+ inhibition lag time graphic is subsequently built. The quan tification of the sample is performed by substituting the standard antioxidant by a suitable aliquot from the aqueous extract of the food to be assessed. The result is expressed in equivalent pg of food gallic acid/g or any other antioxidant equivalent.
