Abstract
1370548 Photo-metric chemical analysis MEDICOR MUVEX 11 July 1972 [13 Jan 1972] 32338/72 Heading G1A The concentrations of two components of a specimen having different optical properties are individually determined and visually displayed by passing light through the specimen, detecting the intensity at two different wavelengths differing from the wavelength of an isosbestic point of the two components, or over different wavebands, logarithmizing the outputs of the detectors and processing to produce signals proportional to the individual concentrations of the components. The system described is used to determine the concentrations of oxidized and reduced haemoglobin in a blood sample and also to determine the ratio of these concentrations i.e. the oxygen saturation and the total haemoglobin concentration. Light is passed through the sample and the intensities at two wavelengths (in the red and near IR portions of the spectrum respectively) are determined by means of a pair of detectors provided with appropriate filters. The light source may be an incandescent lamp or one or more LEDs may be used which because of their narrow spectral emission may allow the omission of the filters. The outputs of the detectors are applied to logarithmizers 8, 14 and signals corresponding to the signal obtained with zero haemoglobin concentration are subtracted by means of potentiometers 9, 15 and op amps 11, 17. The resulting signals are directly related to the concentrations of both the oxidized and reduced haemoglobin but differ due to the difference in absorption at different wavelengths resulting in a pair of linear equations with two unknowns. The signals are then combined by op amps 13 and 19 in such proportions that the output of op amp 13 is proportional to the concentration of oxidized haemoglobin only and that of op amp 19 to the concentration of reduced haemoglobin only. A further op amp 20 sums these signals to produce the total haemoglobin concentration and circuit 21 obtains their ratio to produce the oxygen saturation value. A switch K selects any one of the signals for display on a meter 23. Alternative circuit arrangements for solving the simultaneous linear equations to produce the separate concentration signals are described, Figs.3 to 5 (not shown). The detector may be arranged to provide a logarithmic output by terminating it with a large resistance and effectively operating it under noload conditions.
