The explanation is given in the [REC]: Historias Inéditas ("Untold Stories") comic book, where is told that Father Alberta took some neighboring children up there to test the demonic virus on them and study its effects (however, is not made clear how exactly he gets to take them or the infected Medeiros girl without attracting attention). After the chained Medeiros creature broke free, the fearful Alberta locked himself upon the ceiling and starved to death, leaving her and his infected children loose in the attic.
