A high school virgin, who is eager to have sex, gets a guilt free pass from his girlfriend to lose his virginity while attending college parties for a whole weekend.

The movie will shift its focus on Erik Stifler, the cousin of Matt and Steve, a youngster who is nothing like his wild relations. Peer pressure starts to turn him to live up to the legacy of the other Stiflers when he attends the Naked Mile, a naked run across the college campus. As a Stifler you should have a lot going for you in high school, but this is not the case with Erik. Unlike his illustrious cousins he isn't a oversexed partying jock, but rather a good-natured shy guy. He hasn't even had sex, although he has a girlfriend, but Tracy Sterling is not quite ready for it yet. Erik is getting pretty desperate though. He can't leave high school as a virgin, not as a Stifler, can he? Then Tracy, after being ridiculed by her girlfriends, decides it's time. Erik is overjoyed, but when things go awry, Tracy is not in the mood anymore. She doesn't want to restrain him any further and gives him a guilt-free pass for the weekend. Erik Stifler (John White) Steve and Matt Stifler's cousin, has a difficult time living up to his family name. He is perhaps the only Stifler about to graduate from high school as a virgin. The Stifler name comes with responsibilities ad people expect Erik to live up to the legacy.

Erik feigns illness with his mother, so that he can stay home and masturbate. As soon as everyone is out of the house, Erik puts on a pornographic video cassette for his viewing pleasure. Since the neighbor was mowing his lawn, Erik turns the volume really high, as he was not able to hear anything. He puts on the headphones to drown out the noise, but due to the same he is not able to hear his parents coming back, to pick up a coupon that Erik's grandmother wanted to use.

Unfortunately, just as he is about to ejaculate, his parents Harry (Christopher McDonald), mother (Maria Ricossa) and grandmother (Jessica Booker) unexpectedly walk into the door and are hit with Erik's semen, after which his grandmother dies of a heart attack.

Erik's dad later surmises that, as a Stifler, his son should be out having sex instead of masturbating. Erik's girlfriend of two years, Tracy (Jessy Schram), loves him, but is not ready for intercourse. Now Erik and Tracy are very different from each other. She is into horse riding, while he is into video games. They are both very awkward around each other and yet Tracy keeps going out with him.

Erik tells Tracy that longer she stays a virgin, she seems purer and innocent, but the longer he stays a virgin he becomes a laughing-stock of the entire school. Tracy's girlfriends also counsel her that if she does not give Erik a reason to stick around, he will break up with her.

Tracy suddenly decides she is now ready, inviting Erik to her house later to have sex for the first time. They are interrupted when her father comes downstairs to the basement for a nightcap. Erik sneaks into Tracy's home at midnight, where Tracy is waiting for him in the basement. Before Tracy's father can catch him, a naked Erik flees from the dryer, in which he defecated while hiding as he has a weak stomach. Erik takes refuge with Ryan, as Tracy's father calls the police. Ryan lends Erik some clothes and drives him back home. The next morning, when Erik has talk with Tracy about another attempt, she backs out of trying again as she takes the interruption from her father as a sign from God.

Erik's friends Mike "Cooze" Coozeman (Jake Siegel) and Ryan (Ross Thomas) plan a road trip to visit Erik's cousin Dwight Stifler (Steve Talley) in Michigan during an event known as the Naked Mile. The event is where thousands of students get naked and run for a mile as a way of blowing off steam after the exams. Ryan is a true ladies' man, but Cooze keeps striking out with them due to his coarse approach. Ryan is the quarterback of the football team and the coolest guy in school.

Tracy sees this as an opportunity to give Erik a "guilt free weekend pass," hoping that he can quench his lust and get sex out of his system since she is not ready. But after attending her weekly yoga class, Tracy's girlfriends make her realize that she made a mistake as now Erik would go from having sex, back to not having sex with Tracy. And this would most certainly lead to Erik breaking up with her. Tracy tries calling Erik, but Ryan notices her call and throws Erik's phone out of the car.

Tracy has a run-in with Jim's Dad, Noah Levenstein (Eugene Levy) whom she confides in about her situation with Erik. Mr. Levenstein persuades her not to give up on what she and Erik have. Noah is alarmed when he finds that Tracy gave Erik a free pass to the Naked Mile weekend but assures her that it does not mean that Erik would necessarily do anything.

Erik and his friends arrive on campus and witness an over-the-top drinking contest where Dwight is crowned a campus champion. Dwight enlists the guys into a rough game of football against a bitter rival fraternity composed almost entirely of midgets. The college team were 4 guys short due to their arrest and if the team did not play every match, the team would be kicked out of the campus. Rock is the midget captain and plays dirty. Last year he bit a chunk out of Dwight's thigh. The team ends up losing to the midgets and end up in a brawl with those same midgets on several occasions.

Later at a bar, Erik meets college girl Brandy (Candace Kroslak), who has a fetish for virgins, prompting Ryan and Cooze to make a bet regarding whether or not Erik will sleep with her. Erik ends up telling Brandy about the time he almost had sex with Tracy and ran naked through the streets to escape her dad.

At the same time, Ryan and Cooze end up getting turned down by a couple of girls they have met at the bar, Jill (Jaclyn A. Smith) and Alexis (Angel Lewis), who are taken by a couple of the midgets. This rivalry comes to a head when the midget fraternity attacks Dwight, landing him in the hospital.

Dwight is back in full form later that evening when it is time for the Naked Mile run. Noah Levenstein (Eugene Levy) is a college legend who presides over the Naked Mile run. At first, they are reluctant to run, but when Brandy, Jill, and Alexis strip down stark naked, they are finally prompted to strip down as well. Finally, as Erik and Brandy reach the finish line, they share a kiss which is caught on camera for a TV news report. Watching the report about the Naked Mile at home, Tracy is upset and feels guilty that she allowed Erik the pass. Her friends persuade her also to lose her virginity before he gets back.

Later that evening, Erik realizes that he loves Tracy, so confesses to Brandy that he cannot sleep with her, and rushes back to see Tracy. However, Erik's SUV runs out of gas, and he is forced to steal a horse and ride barebacked into town.

When he gets to her house, Tracy's dad says she is at a party. Erik's car runs out of gas 10 miles out of town, so he rides a horse to the party, pounds on the closed bedroom door and proclaims his love for her. However, Tracy is not in the room because she had decided that she could not go through with her plans. They reconcile and have sex.

When Erik returns to the Beta house to pick up his friends the next morning, each guy shares stories of his sexual experiences from the night before. The guys then ask him if he "sealed the deal" with Brandy that night, and when Erik tells them no, Ryan pays up on the bet to Cooze. Erik tells them about his adventure back home to make up with Tracy and finally lose his virginity. The guys are proud of him for officially living up to the Stifler family name, and the three friends go home.

During the post-Naked Mile party, Dwight spots Vicky (Mika Winkler), the midget girlfriend of Rock, and the two of them head up to Dwight's room to have sex. Also at the Beta house, Cooze makes with Jill who ties him up and subjects him to S&M and bondage. Ryan has more pleasant luck when he has sex with Alexis.

As it turns out, Dwight and Vicky start dating, and they end up traveling the world together, as various postcard pictures show their many journeys together as the end credits roll.
