Shilpa Shukla comes from a family of scholars, political leaders, and bureaucrats. Her parents are Late N.K. Shukla, who was an income tax officer, and Namita Shukla.

Shukla was a student of Delhi Public School in R. K. Puram in New Delhi. She attended Miranda House at the University of Delhi for her college education.

She started modelling at the age of 16. She ventured into drama and started her stage career with eminent theatre personality Maharaj Krishna Raina. After working in several plays and a few TV serials on Doordarshan, she started receiving offers for minor movie roles.

Shukla made her debut on the big screen with the Indo-Pakistani film Khamosh Pani Eau dormante (2003). Her next movie was Hazaaron Khwaishein Aisi (2003), an Indian political drama film directed by Sudhir Mishra. It received positive reviews from critics.

She gained popular acclaim with Chak De! India (2007) starring Shah Rukh Khan. It was a sports drama film produced by Aditya Chopra under the Yash Raj Films banner and directed by Shimit Amin. She played the role of an arrogant hockey player in it. Shukla won the 2008 Screen Awards for Best Supporting Actress for her role.

She appeared in a lead role in the neo-noir thriller B.A. Pass (2012). She won the 2014 Screen Awards for Best Actor in a Negative Role (Female) for this film. Shukla also bagged the 2014 Filmfare Awards for Best Actress (Critics). The movie won the Best Film Award in the Indian competition section at the 2012 Osian's Cinefan Festival of Asian and Arab Cinema. It won The Prix Du Public at the first South Asian Film Festival in 2013, where it was voted best film by the French audience.

Shukla's repertoire of films includes Frozen (2007), Bhindi Baazaar (2011), Rajdhani Express (2013), Crazy Cukkad Family (2015), Bombairiya (2019), and HIT: The First Case (2022).

She debuted on the digital platform with the web series Mentalhood (2020). Since then, she has been a part of several other OTT shows, including Hostages (2020), Criminal Justice: Behind Closed Doors (2020), Four More Shots Please (2019), and Taaza Khabar (2023).
