She studied acting at the Central School of Speech and Drama in the United Kingdom, where she was discovered to film a movie with Sean Connery.

Brantley began her career in the early 1980s, with her breakout role in the 1982 film Five Days One Summer opposite Sean Connery. She gained widespread recognition for her memorable performance in The Princess Bride (1987), establishing herself as a versatile character actress.
