Nick Mancuso was considered for the role of Indiana Jones in Raiders of the Lost Ark along with Tom Selleck and Harrison Ford. Coincidentally, after being considered by Steven Spielberg to play Indiana Jones, Mancuso won the role of Nick in Death Ship opposite George Kennedy, which was about Nazi ghosts from WWII. Mancuso would also later star opposite Indiana Jones alum John Rhys-Davies in Marquis de Sade, produced by Roger Corman.

Steve Sohmer, the creator of "Mancuso, FBI" starring Robert Loggia, joined NBC in 1982 working directly with Stephen J. Cannell who created the series "Stingray" starring Nick Mancuso. Partially inspired by the work of Stephen J. Cannell and Nick Mancuso's work on "Stingray", Steve Sohmer created the character FBI agent Nick Mancuso for his novel "Favorite Son.".

In 1983 he was invited to become an associate of the Actors Studio by co-chairman, actor, Martin Landau.

While working with Tennessee Williams on his plays Tiger Tail and Night of the Iguana in the late 70s, Mancuso came to the attention of producers at Columbia Pictures who were eager to turn Mancuso into a new major star for their studio. Martin Ransohoff and Arthur Hiller were instrumental in getting Mancuso for the lead in the horror-thriller Nightwing, directed by Arthur Hiller and costarring David Warner. Ransohoff and Hiller hoped Nightwing would be a hit and be a breakthrough role for Mancuso, but it failed at the box office. However, Mancuso had his breakthrough performance just two years later in Ticket to Heaven.

In 1979 he made his American stage debut working directly with one of America's greatest playwrights, Tennessee Williams, for Tiger Tail and Night of the Iguana in Atlanta, Georgia.

He started his acting career in the mid-1960s as a founding member of several independent theaters in Toronto, such as Toronto Free Theatre, Canadian Stage, Factory Lab Theatre, and Theatre Pass Muraille.

He performed in plays at Canada's prestigious Stratford Festival in 1976, in the leading role of Bassanio in "The Merchant of Venice", as well as in "Antony and Cleopatra" and "A Midsummer Night's Dream", working with Maggie Smith, Jessica Tandy and Hume Cronyn.
