Archeologist Steve Banning (Dick Foran) and his bumbling cohort Babe Jenson (Wallace Ford) stumble upon a vase with clues to the direction to the tomb of Egyptian princess Ananka. Laughed at by Professor Andoheb (George Zucco) of the Cairo Museum, they secure funding from magician Tim 'The Great Solvani' Sullivan (Cecil Kellaway) and his daughter Marta (Peggy Moran) and set out on an expedition, unknowingly awakening an ancient curse that revives the 3,000 year old living mummy Kharis (Tom Tyler), guardian of Ananka's tomb.

The Mummy's Hand follows La Momie (1932) (1932), which was based on a screenplay by American playwright/screenwriter John L Balderston. It is not an actual sequel to The Mummy and is mostly considered a retelling of Balderston's screenplay. The screenplay for The Mummy's Hand was written by American screenwriters Griffin Jay and Maxwell Shane. It was followed by La Tombe de la momie (1942) (1942), Le fantôme de la momie (1944) (1944), and La Malédiction de la momie (1944) (1944).

Steve discovers the secret passage into Ananka's tomb and attempts to rescue Marta from Andoheb. Before he can get the straps off her, however, the Mummy lumbers in looking for more tana fluid. Steve and Babe smash a vial and a pot containing more tana fluid, and Steve smashes a firey brazier on top of the mummy, setting him on fire. Some time later, Steve and Marta find another broken vase in the Cairo market, but Marta reminds him that they are going back to America. In the final scene, Babe shows Steve a cablegram informing him that he's got a position at the Scripps Museum.
